Project description:B-cell acute lymphocytic leukemia (B-ALL), a common blood cancer in children, leads to high mortality. Cyclin-dependent kinase 9 inhibitor (CDK9i) effectively attenuates acute myeloid leukemia and chronic lymphoblastic leukemia by inducing apoptosis and inhibiting cell proliferation. However, the effect of CDK9i on B-ALL cells and the underlying mechanisms remain unclear. In this study, we showed that CDK9i induced the apoptosis of B-ALL cells in vitro by activating the apoptotic pathways. In addition, CDK9i restrained the glycolytic metabolism of B-ALL cells, and CDK9i-induced apoptosis was enhanced by co-treatment with glycolysis inhibitors. Furthermore, CDK9i restained the glycolysis of B-ALL cell lines by markedly downregulating the expression of glucose transporter type 1 (GLUT1) and the key rate-limiting enzymes of glycolysis, such as hexokinase 2 (HK2) and lactate dehydrogenase A (LDHA). Moreover, cell apoptosis was rescued in B-ALL cells with over-expressed c-Myc after treatment with CDK9i, which is involved in the enhancement of glycolytic metabolism. In summary, our findings suggest that CDK9 inhibitors induce the apoptosis of B-ALL cells by inhibiting c-Myc-mediated glycolytic metabolism, thus providing a new strategy for the treatment of B-ALL.

Project description:Chronic lymphocytic leukemia (CLL) is effectively treated with targeted therapies including Bruton tyrosine kinase inhibitors (ibrutinib or acalabrutinib) and BCL2 antagonists (venetoclax). When these become ineffective, treatment options are limited. Positive transcription elongation factor complex (P-TEFb), a heterodimeric protein complex composed of cyclin dependent kinase 9 (CDK9) and cyclin T1, functions to regulate short half-life transcripts by releasing RNA Polymerase II (POLII) from proximal-promoter pause by way of phosphorylation. The transcripts regulated by P-TEFb are frequently dysregulated in solid tumors and hematologic malignancies; however, therapies targeting inhibition of P-TEFb have not yet achieved approval for cancer treatment. This is due to lack of selective inhibitors and undesirable side-effect profiles of less selective inhibitors. VIP152 kinome profiling revealed CDK9 as the main enzyme inhibited at 100nM, with over a 10-fold increase in potency compared with other inhibitors currently in development for this target. VIP152 induced cell death as measured by apoptosis in CLL cell lines and primary patient samples. Transcriptome analysis revealed compensatory upregulation in translation machinery and inhibition of RNA degradation. Mechanistically, VIP152 inhibits the assembly of P-TEFb onto the transcription machinery and disturbs CDK9 binding partners such as HEXIM1, MEPCE, and LARP7. Finally, C57BL/6J immune competent mice were engrafted with CLL-like cells of Eµ-MTCP1 over-expressing mice and treated with either VIP152 or vehicle. VIP152-treated mice had a reduction of blood disease over time and an improved overall survival. These data suggest that VIP152 is a highly selective inhibitor of CDK9 that represents an attractive new therapy for CLL.

Project description:Phosphorylation of RNA polymerase II by cyclin-dependent kinase 9 (CDK9) is crucial for transcriptional regulation, and acute inhibition of CDK9 results in the rapid downregulation of genes with short-lived transcripts and labile proteins. Multiple non-selective CDK9 inhibitors have progressed clinically but were limited by a narrow therapeutic window. This work describes a novel, potent, and highly selective CDK9 inhibitor, AZD4573. Using integrated transcriptomic and proteomic analyses, loss-of-function pathway interrogation, and pharmacological comparisons, Mcl-1 depletion was identified as a major mechanism through which AZD4573 induces cell death in tumor cells. AZD4573 suppresses Mcl-1 in a dose- and time-dependent manner that causes caspase-3 cleavage in sensitive cancer cells, most notably in hematological malignancies. This pharmacodynamic response is observed in vivo as well, which leads to regressions in both subcutaneous tumor xenografts and disseminated models at tolerated doses as monotherapy or in combination with venetoclax. Fully understanding the mechanism, exposure, and anti-tumor activity of AZD4573 also informed the development of a robust PK/PD/efficacy model that helped select and predict the starting and efficacious dose range in man. Accordingly, AZD4573 is currently being evaluated in a phase I clinical trial for patients with hematological malignancies (clinicaltrials.gov identifier: NCT03263637).

Project description:CDK9, in complex with cyclin T1 or T2, is essential for mRNA transcription by enabling paused RNA polymerase II to proceed into elongation. Increasing evidence highlights CDK9’s involvement in transcriptional addiction in cancer. In this study, we explored the efficacy of novel CDK inhibitors and demonstrated that targeting CDK9 significantly impairs proliferation and induces apoptosis in various triple-negative breast cancer (TNBC) cell lines. Transcriptomic analysis revealed that CDK9 inhibition caused downregulation of genes governing transcription, cell cycle progression, and oncogenic signalling pathways, such as TGF-β and Wnt/β-catenin signalling. This disruption of transcriptional programs was further enhanced by combining CDK9 and EGFR inhibitors.
