Project description:The brainstem dorsal vagal complex (DVC) is known to regulate energy balance and is the target of appetite suppressing hormones, such as glucagon-like peptide-1 (GLP-1). Here we provide a comprehensive genetic map of the DVC and identify neuronal populations that control feeding. Combining bulk and single-nucleus gene expression and chromatin profiling of DVC cells, we reveal 25 neuronal populations with unique transcriptional and chromatin accessibility landscapes and peptide receptor expression profiles. GLP-1 receptor (GLP-1R) agonist administration induces gene expression alterations specific to two distinct sets of Glp1r neurons – one population in the area postrema (AP) and one in the nucleus of the solitary tract (NTS) that also expresses calcitonin receptor (Calcr). Transcripts and regions of accessible chromatin linked to obesity-associated genetic variants are enriched in AP and NTS neurons that express Glp1r and/or Calcr, and activation of several of these neuronal populations decreases feeding in rodents. Thus, DVC neuronal populations associated with obesity predisposition suppress feeding and may represent targets for therapy of obesity.

Project description:We investigated neural populations mediating the effects of cagrilintide on energy balance in the dorsal vagal complex (DVC). Single-nucleus RNA sequencing and spatial transcriptomics were performed across mouse, rat, and macaque brainstem tissue. Treatment groups include diet-induced obese animals treated acutely or subchronically with cagrilintide or vehicle. Diurnal patterns of gene expression in the dorsal vagal complex and the central nucleus of the amygdala

Project description:Genes that establish the circadian clock have differential expression with respect to solar time in central and peripheral tissues. Here, we find circadian-time-induced differential expression in a large number of genes not associated with circadian rhythms in two brain regions lacking overt circadian function: the dorsal vagal complex (DVC) and the central nucleus of the amygdala (CeA). These regions primarily engage in autonomic, homeostatic, and emotional regulation. However, we find striking diurnal shifts in gene expression in these regions of male Sprague Dawley rats with no obvious patterns that could be attributed to function or region. These findings have implications for the design of gene expression studies as well as for the potential effects of xenobiotics on these regions that regulate autonomic and emotional states. Micropunches of dorsal vagal complex (DVC) and central nucleus of the amygdala (CeA) were collected from male, Sprague Dawley rats at three different times of the day. Animals were not experimentally different in any way except for the time of sacrifice and collction. Gene expression of 145 genes were measured with Fluidigm's Biomark 96.96 platform.

Neuromodulatory co-expression in cardiac vagal motor neurons of the Dorsal Motor Nucleus of the Vagus [RNAseq]

Project description:The activity of the cardiac vagal innervation is well known to be crucial to the maintenance of cardiac health, and to protect and recover the heart from injury. Only recently has this role been shown to depend on the activity of the underappreciated Dorsal Motor Nucleus of the Vagus (DMV). By combining neural tracing, transcriptomics, and 3D mapping in male and female Sprague Dawley rats we characterize cardiac-specific neuronal phenotypes in DMV. We find that the DMV cardiac-projecting neurons differentially express PACAP, CART, and synucleins, as well as evidence that they participate in neuromodulatory co-expression involving catecholamines. The significance of these findings is enhanced by previous knowledge of the role of PACAP at the heart and of the other neuromodulators in peripheral vagal targets.
