Project description:Background:
- Some bacteria found in the large and small intestines help keep people healthy and aid digestion. They may also affect a person s risk of developing cancer. Researchers want to study the relationship between intestinal bacteria and breast cancer risk factors. They can do this by looking at stool and urine samples from postmenopausal women.
Objectives:
- To study intestinal bacteria and its relationship to urine-based markers of breast cancer risk in women.
Eligibility:
- Women between 55 and 69 years of age with a recent mammogram that showed no signs of cancer.
Design:
* Participants will be screened with a medical history and basic health questionnaire.
* At home, participants will complete questionnaires about cancer risk factors and food consumption.
* Participants will also collect urine and stool samples. They will send the samples to the designated labs for study.
* No treatment will be provided as part of this protocol.

Project description:Probiotic bacteria, specific representatives of bacterial species that are a common part of the human microbiota, are proposed to deliver health benefits to the consumer by modulation of intestinal function via largely unknown molecular mechanisms. To explore in vivo mucosal responses of healthy adults to probiotics, we obtained transcriptomes in an intervention study following a double-blind placebo-controlled cross-over design. In the mucosa of the proximal small intestine of healthy volunteers, probiotic strains from the species Lactobacillus acidophilus, L. casei and L. rhamnosus each induced differential gene regulatory networks and pathways in the human mucosa. Comprehensive analyses revealed that these transcriptional networks regulate major basal mucosal processes, and uncovered remarkable similarity to response profiles obtained for specific bioactive molecules and drugs. This study elucidates how intestinal mucosa of healthy humans perceive different probiotics and provides avenues for rationally designed tests of clinical applications. Project description:Antibiotic-resistant “super-bug” bacteria represent a global health problem with no imminent solutions. Here, we demonstrate that AB569 (acidified nitrite (A-NO2-) and Na2-EDTA) killed all Gram-negative and Gram-positive bacteria tested. AB569 was also efficacious at killing the model organism P. aeruginosa in biofilms and in a murine chronic lung infection model. AB569 was not toxic to human cell lines at bactericidal concentrations. RNA-Seq analyses upon treatment of P. aeruginosa with AB569 revealed a catastrophic loss of the ability to support core pathways encompassing DNA, RNA, protein, ATP biosynthesis, and iron metabolism. Electrochemical analyses elucidated that AB569 produced more stable SNO-proteins, potentially explaining one mechanism of bacterial killing. Our data implicate that AB569 is a safe and effective means to kill pathogenic bacteria, suggesting that simple strategies could be applied with highly advantageous therapeutic/toxicity index ratios to pathogens associated with a myriad of peri-epithelial infections and related disease scenarios.

Project description:Gut-resident microbes contribute to host health via multiple mechanisms. Some of the most striking gut microbiota induced effects occur in the extraintestinal tissues and are restricted to early life. The mechanisms by which gut residing bacteria induce effects on distant host tissues and why this is restricted to a period in early life are largely unknown. We found that a subset of live gut-resident bacteria spontaneously translocate from gut to extraintestinal tissues in preweaning, but not adult mice. Translocation in preweaning mice appeared physiologic as it did not induce an inflammatory response and was in part controlled by sphingosine-1-phsophate receptor (S1PR) expressing host cells and host goblet cells. One translocating strain, Lactobacillus animalisWU, contained unique coding sequences for genes in the tyrocidine-gramicidin antibiotic-synthesizing gene cluster as well as five other regions putatively producing secondary metabolites with anti-microbial activity. Lactobacillus animalisWU exhibited antimicrobial activity against the late-onset sepsis pathogen E. coli ST69 in vitro, and translocation of L. animalisWU protected preweaning mice from systemic E. coli ST69 sepsis in vivo. These observations demonstrate a previously unappreciated higher-level symbiosis with our gut microbes.
