New genomic resources and comparative analyses reveal differences in floral gene expression in selfing and outcrossing Collinsia sister species

Project description:We used flower bud transcriptomes from Collinsia rattanii and its predominantly outcrossing sister species, C. linearis, to explore the genomic basis of mating system and phenotypic evolution in Collinsia, a self-compatible genus. Transcriptional regulation of enzymes involved in pollen formation may influence floral traits that distinguish selfing and outcrossing Collinsia species through pleiotropic functions. These patterns provide clues about parallel evolution in selfing plants.

Transcriptome profiling and weighted gene co-expression network analysis of early floral development in Aquilegia coerulea

Project description:The current experiment was designed to obtain a broad characterization of the genetic pathways acting in early Aquilegia coerulea floral meristem development. We conducted an in-depth transcriptome profiling of early floral development in A. coerulea at four finely dissected developmental stages, with eight biological replicates per stage. . The developmental window we sequenced sampled stages that started with a late phase of stamen initiation, covered the period of FM termination, and ended with the initial stage of morphogenesis of the floral organs.

RNA-seq of bird-pollianted, red-flowering and insect-pollinated, yellow-flowering Diplacus aurantiacus varieties sourced from various locations in San Deigo State, USA

Project description:Flower phenotypes in the species Diplacus aurantiacus in Southern California along an east west transect range from large, yellow, insect-pollinated flowers through orange flowers to small, red, bird-pollinated flowers. Until now, intermediate forms were attributed to recurrent hybridization at the (sub)-species level. However, by monitoring the flower phenotypes of these populations in field studies over the past 20 years, Rolf Baumberger observed that the transition in flower phenotype occurs during the lifespan of individual long-lived plants, thus ruling out a hybrid origin of intermediate forms. Further research has revealed that this transition bears the hallmark of an epigenetic transition. The small, red, bird-pollinated state is stable and heritable but reverts at frequencies of 1-2 %, much higher than that of genetic alterations. In our first approach a comparative transcriptome analysis of individuals of both stable morphotypes, we like to unravel candidate genes involved in floral colour and morphology determination.

Transcriptome Sequencing Analysis Reveals the Regulation of the Hypopharyngeal Glands in the Honey Bee, Apis mellifera carnica Pollmann

Project description:Transcriptome sequencing has become the main methodology for analyzing the relationship between genes and characteristics of interests, particularly those associated with diseases and economic traits. Because of its functional superiority, commercial royal jelly (RJ) and its production are major areas of focus in the field of apiculture. Multiple lines of evidence have demonstrated that many factors affect RJ output by activating or inhibiting various target genes and signaling pathways to augment their efficient replication. The coding sequences made available by the Honey Bee Genome Sequencing Consortium have permitted a pathway-based approach for investigating the development of the hypopharyngeal glands (HGs). In the present study, 3573941, 3562730, 3551541, 3524453, and 3615558 clean reads were obtained from the HGs of five full-sister honey bee samples using Solexa RNA sequencing technology. These reads were then assembled into 18378, 17785, 17065, 17105, and 17995 unigenes, respectively, and aligned to the DFCI Honey Bee Gene Index database. The differentially expressed genes (DEGs) data were also correlated with detailed morphological data for HGs acini. The results identify areas that warrant further study, including those that can be used to improve honey bee breeding techniques and help ensure stable yields of RJ with high quality traits.

Cis-regulatory divergence of eloF drives reproductive isolation in Drosophila

Project description:The differential production of pheromones is a major barrier to mating between species in Drosophila. Individuals from morphologically similar sister species can produce different sets of cuticular hydrocarbons that allow potential mates to identify them as a suitable partner. In order to elucidate cis-regulatory mechanisms behind speciation, we looked for allele-specific expression in hydrocarbon-producing oenocytes from F1 hybrids of the sister species D. simulans and D. sechellia. By focusing on cis-regulatory changes specific to female oenocytes, we rapidly identified a small number of candidate genes. Oour RNA-seq approach proved to be far more efficient than QTL mapping in identifying candidate genes, and it can be used to pinpoint the genetic basis for a wide range of traits differing due to cis-regulatory divergence between any interfertile species.
