RNA-Seq of Aged Polg Mutator Mouse Brains

Project description:We have performed both proteomic and transcriptomic analyses of the brains of one-year old Polg mutator mice. As part of this analysis, we generated RNA-Seq data from RNA isolated from one cerebral hemisphere of these mice. mRNA levels and protein abundance levels were compared in this study. 11 samples of RNA isolated from mouse cerebral hemisphere. 7 Polg D257A/D257A, 1 Polg D257A/WT, and 3 Polg WT/WT. The samples were collected from one-year old male mice.

Transcriptomic analysis of WT and Polg D257A/D257A bone marrow derived macrophages (BMDMs) challenged with Pseudomonas aeruginosa

Project description:Mitochondrial diseases (MtD) represent a significant public health challenge due to their heterogenous clinical presentation, often severe and progressive symptoms, and lack of effective therapies. Environmental exposures, such bacterial and viral infection, can further compromise mitochondrial function and exacerbate the progression of MtD. Infections in MtD patients more frequently progress to sepsis, pneumonia, and other detrimental inflammatory endpoints. However, the underlying immune alterations that enhance immunopathology in MtD remain unclear, constituting a key gap in knowledge that complicates treatment and increases mortality in this vulnerable population. This dataset reports transcriptomic changes in bone marrow derived macrophages isolated from wild type (WT) C57BL/6J mice and a mouse model of polymerase gamma (Polg)-related MtD (Polg D257A/D257A), both at rest and six hours after infection with Pseudomonas aeruginosa strain PAO1. These data reveal a hyperinflammatory innate immune status in Polg D257A macrophages characterized by elevated expression of genes involved in interferon and inflammatory cell death pathways. This work sheds new light on innate immune dysregulation in a model of mitochondrial dysfunction and reveals potential targets for limiting infection- and inflammation-related complications in Polg-related MtD.

RNAseq data from mtDNA mutator induced pluripotent stem cells

Project description:RNAsequencing data from POLG D257A mutant mouse iPS (induced pluripotent stem) cells. These mice accumulate random point mutations in their mitochondrial genome and manifest progeric features. The reprogramming and phenotype of the cells is described in Hämäläinen et al. 2015, Cell reports, mtDNA Mutagenesis Disrupts Pluripotent Stem Cell Function by Altering Redox Signaling. The goal of the study was to analyze altered gene expression profiles between mutant and wt iPS cells to identify altered biological pathways responsible for the stemness and proliferation defects seem in mtDNA mutator stem cells.

This data set utilizes an in vitro assay using Pseudomonas aeruginosa (PA) infection of bone marrow derived macrophages (BMDMs) to clarify the molecular and cellular basis for innate immune hyperactivity in a mouse model of polymerase gamma (Polg)-related MtD. This dataset reports the proteome signatures of WT and Polg D257A mutant BMDMs at rest and 2-8 hours post infection with PA.
