Project description:Maternal blood, as well as umbilical cord blood samples, were collected and DNA methylation levels were determined by Illumina MethylationEPIC microarray. Methods: Twenty-four subjects were chosen from a previous clinical study. Overweight/obese pregnant women (body mass index ≥24kg/m2) who had an uncomplicated pregnancy at <12+6 weeks of gestation were randomly allocated to either an exercise or a control group. Patients allocated to the exercise group performed 3 exercise bouts per week (at least 30 min/session with a rating of perceived exertion between 12-14) via a cycling program that was initiated within 3 days of randomization until 37 weeks of gestation. Patients allocated to the control group continued their usual daily activities. Maternal blood, as well as umbilical cord blood samples, were collected and DNA methylation levels were determined by Illumina MethylationEPIC microarray.

Shotgun metagenomic profiling of human fecal microbiota before and after aerobic exercise intervention

Project description:Lean nonalcoholic fatty liver disease (NAFLD) is increasingly recognized as a distinct clinical phenotype with limited evidence for effective non-pharmacological interventions and unclear mechanistic pathways. Aerobic exercise is recommended for NAFLD management; however, its effects and the gut microbiota–associated mechanisms in lean NAFLD remain incompletely understood. This dataset was generated from a randomized controlled trial (ClinicalTrials.gov identifier: NCT04882644). Participants assigned to the aerobic exercise intervention group provided fecal samples at baseline and after the 3-month intervention. A total of 33 paired fecal samples were included in this dataset. Gut microbiota profiles were generated using shotgun metagenomic sequencing. The dataset includes processed and de-identified species-level relative abundance tables derived from fecal samples collected before and after the intervention. These data were used to characterize exercise-induced alterations in gut microbial composition and interindividual variability in microbiota responses to aerobic exercise in lean NAFLD. The data support integrative analyses with clinical phenotypes and circulating metabolomic profiles to explore gut microbiota–associated mechanisms underlying the metabolic benefits of aerobic exercise.

16S rRNA gene sequencing of human fecal microbiota before and after exercise intervention

Project description:Lean nonalcoholic fatty liver disease (NAFLD) is increasingly recognized as a distinct clinical phenotype with limited evidence for effective non-pharmacological interventions and unclear mechanistic pathways. Aerobic exercise is recommended for NAFLD management, yet its effects and underlying gut microbiota–mediated mechanisms in lean NAFLD remain insufficiently characterized. This study is based on a randomized controlled trial (ClinicalTrials.gov identifier: NCT04882644) in which 100 adults with lean NAFLD were randomly assigned to a 3-month aerobic exercise intervention or usual care. 63 paired fecal samples were collected at baseline and after intervention. Gut microbiota profiles were generated using 16S rRNA gene sequencing. The dataset includes processed taxonomic abundance tables derived from fecal samples collected before and after the intervention. These data were used to characterize exercise-induced alterations in gut microbial diversity, composition, and functional potential, and to explore interindividual heterogeneity in microbiota responses to aerobic exercise in lean NAFLD. The microbiome data deposited in this series support integrative analyses with clinical phenotypes and circulating metabolomic profiles, aiming to elucidate gut microbiota–associated mechanisms underlying the metabolic benefits of aerobic exercise in lean NAFLD.

Effects of 20-week exercise on whole-blood genome-wide DNA methylation profile in children with overweight/obesity

Project description:In this project, the effects of a 20-week exercise intervention on whole blood genome-wide DNA methylation signatures (CpG sites level) in boys and girls with overweight/obesity (OW/OB) were investigated. Twenty-three children (10.05 ± 1.39 years, 56% girls) with OW/OB, were randomized to either a 20-week exercise intervention (exercise group [EG]; n=10; 4 boys/ 6 girls), or to usual lifestyle (control group [CG] (n=13; 6 boys/ 7 girls). Whole blood genome-wide DNA methylation and transcriptome profile (RNA-seq) analyses were performed before and after the intervention period. Changes in the DNA methylation sites of 485 and 386 CpGs were induced by the exercise intervention (compared to the control group) in boys and girls respectively (p < 0.001). These CpG sites mapped to loci enriched in distinct gene pathways related to metabolic diseases, fatty acid metabolism, and immune function in boys or girls (p < 0.05). In boys, changes on the DNA methylation status of 87 CpG sites (from the subset of 485 significant CpGs) associated with changes in the gene expression levels of 51 gene transcripts which were regulated by exercise (p < 0.05). Among girls, changes on DNA methylation at 46 CpG sites (from the initial 386 significant CpGs) were associated with changes in the gene expression levels of 30 gene transcripts affected by exercise. Gene transcripts affected by exercise-induced DNA methylation were related to obesity, metabolic syndrome, and inflammation. Nevertheless, none of the presented analyses survived multiple testing correction (FDR > 0.05). This multi-omics approach reveals that exercise may regulate gene pathways involved in metabolism and immune functions in blood cells of boys and girls with OW/OB. These preliminary results provide an important first step to characterize the molecular response to exercise interventions in pediatric populations.

Proteomic and Metabolomic Profile of Cognitive Stability and Decline Over an Aerobic Exercise Intervention and 5-years later in Older Adults

Project description:. In this study we identify a unique signature protein and metabolic biomarker panel for those who improve in cognition and those who decline, assessed by the Montreal Cognitive Assessment (MoCA). We investigate a discovery cohort of Brain in Motion participants involving a quasi-experimental aerobic exercise intervention study and analyze blood plasma before and after a six-month aerobic exercise intervention and 5 years later. We relate the differentially expressed proteins and metabolites to subjective sleep quality measures and fitness level. This study informs on the differences in the plasma of those who decline and those who improve in cognition, identifying pathways that are of importance for preventing cognitive decline.
