Conservation and divergence of vulnerability and responses to stressors between human and mouse astrocytes

Project description:Astrocytes were purified from mouse cortices and human cortex tissue by immunopanning with HepaCAM. All treatments were proceeded on 3DIV in serum-free medium. Astrocytes were treated by TNFα for 48hr, by Poly I:C for 72hr, or by hypoxia for 72hr. Acutely purified atrocytes RNA was harvest immediately after HepaCAM immunopanning. Serum selected astrocytes RNA was collected after 4-6 days culturing in serum medium. Serum-free cultured astrocytes RNA was collected on 5-6 DIV. Serum-selection purified human astrocytes were transplanted into P2-11 Rag2 immunodeficient mouse brains, and after about 8 months, transplanted human and host mouse astocytes RNA was collected acutely on HepaCAM immunopanning dish.

Expression data from SHP specific siRNA or nonspecefic siRNA transfected rat astrocytes

Project description:To clarify the effect of SHP in LXRs-mediated signaling pathway, we performed global gene expression analysis of SHP siRNA transfected- or control siRNA transfected- astrocytes after IFN-Î³ and LXRs agonist. Microarray analysis revealed that expression of several genes encoding inflammatory mediators were reversed in SHP siRNA transfected-astrocytes, when compared with control siRNA transfected-astrocytes. Primary astrocytes were cultured from the cerebral cortices of 1 day-old Sprague-Dawley rats. The astrocytes were transfected with control siRNA or SHP siRNA and treated with IFN-Î³ in the presence or absence of GW for 3 h.

Expression data from reactive astrocytes acutely purified from young adult mouse brains

Project description:Reactive astrogliosis is characterized by a profound change in astrocyte phenotype in response to all CNS injuries and diseases. To better understand the reactive astrocyte state, we used Affymetrix GeneChip arrays to profile gene expression in populations of reactive astrocytes isolated at various time points after induction using two different mouse injury models, ischemic stroke and neuroinflammation. Young adult male mice underwent middle cerebral artery occlusion (MCAO) to produce ischemic stroke or control sham surgery. Young adult mice were injected intraperitoneally with 5 mg/kg lipopolysaccharide (LPS) to produce neuroinflammation or saline for control. Astrocytes were acutely purified from control and injured brains at 1 day after injury for LPS/saline injection and 1, 3 days and 7 days after MCAO/sham surgeries.

Isolation of ferret astrocytes reveals their morphological, transcriptional, and functional differences from mouse astrocytes

Project description:To identify differently expressed genes between ferret and mouse astrocytes, we performed RNA-seq studies on primary ferret and mouse astrocytes cultured in serum free medium. We isolated primary astrocytes from mixed glial cultures which are prepared from the cortices of ferrets and mice at postnatal day 1. Each biological replicate was generated from 2 individuals of the same species. We detected substantial changes in the expression of genes associated with cell proliferation and migration, and identified ferret astrocyte-specific genes.

Project description:We report RNA sequencing data from the infraspinatus muscles of 3-month old male C57BL/6J (Jackson Labs stock 000664) or MRL/MpJ mice (Jackson Labs stock 000486). Mice underwent a full-thickness infraspinatus tenectomy and denervation, and four weeks later muscles were collected for analysis. Comparisons are made to control male mice of the same strains who did not undergo infraspinatus tenectomy and denervation.
