Project description:Endometriosis is characterized by growth of endometrial-like tissue outside of the uterus affecting many women during their reproductive age, causing years of pelvic pain and potential infertility. Its pathophysiology remains largely unknown, limiting early diagnosis and treatment. We characterized peritoneal and ovarian lesions at single-cell transcriptome resolution and compared to matched eutopic endometrium, control endometrium, and organoids derived from these tissues, generating data on over 122,000 cells across 14 individuals. We spatially localized many of the cell types using imaging mass cytometry. We identify a perivascular mural cell unique to the peritoneal lesions with dual roles in angiogenesis promotion and immune cell trafficking. We define an immunotolerant peritoneal niche, fundamental differences in eutopic endometrium and between lesions microenvironments, and an unreported progenitor-like epithelial cell subpopulation. Altogether, this study provides a holistic view of the endometriosis microenvironment representing a comprehensive cell atlas of the disease in hormonally treated patients, essential information for advancing therapeutics and diagnostics.

Accumulating evidence suggests that aberrant mRNA and lncRNA expressions may be involved in the pathogenesis and development of endometriosis. This study therefore aims to survey key TFs, genes and lncRNAs and further understand the mechanism of endometriosis. Paired expression profiling of mRNA and lncRNA in ectopic endometria compared with eutopic endometria were determined by high-throughput sequencing techniques in eight patients with ovarian endometriosis. Our study presents a unique insight into the regulatory network of this enigmatic disorder and possibly provides clues regarding replacement therapy of endometriosis.

Transcriptomic changes in eutopic endometrium and ectopic lesions in mouse model during progression of endometriosis-related infertility

Project description:In this study, we performed transcriptomic analysis in ectopic lesions and eutopic endometrial tissues from both fertile and subfertile mice with endometriosis. We identified the positive correlation of the gene signatures between the mouse and human in ectopic lesions. Conserved gene networks were activated in all the ectopic lesions including estradiol, immune, fibrosis, and angiogenesis pathways. The interactions mediated through hormone, cytokine, and growth factor as well as their corresponding receptors were predicted between the ectopic and eutopic endometrium. EGF and WNT signaling were more suppressed in the eutopic endometrium from subfertile mice. Our results revealed that our mouse endometriosis model recapitulates the important transcriptomic changes of endometriosis progression in human ectopic lesions including the essential regulator network and intensive inter-communications between ectopic and eutopic endometrium. Our preclinical animal model for endometriosis will be invaluable to understand etiology and pathophysiology on endometriosis.

The differential expression of tRF&tiRNA in eutopic and ectopic endometrial tissue of patients with ovarian endometriosis.

Project description:Endometriosis is a chronic gynecological disease that afflicts millions of women of reproductive age and has traditionally been diagnosed by laparoscopic surgery. This diagnostic barrier delays identification and treatment by years, resulting in prolonged pain and disease progression. Development of a noninvasive diagnostic test could significantly improve timely disease detection. We characterized eutopic and ectopic endometrium from five patients with ovarian endometriosis using tRF&tiRNA sequencing. We identified 294 tsRNAs by RNA microarray technique in eutopic and ectopic endometrium. Under the criteria of |log2FoldChange >2.0| and p-value < 0.05, 11 tRFs/tiRNAs were reported to be dysregulated in ectopic endometrial tissue compared with eutopic endometrial tissue, of which eight were up-regulated, and three were down-regulated.
