The four treatments were: sterile water irrigation alone, FS root irrigation, FS and AM201 root irrigation, and FS combined with methyltobuzin (TM) root irrigation. And the differential genes between AM201 and FO groups were identified and compared, which helps to reveal the resistance mechanism of AM201 to Atractylodes macrocephala root rot disease

Project description:Atractylodes macrocephala is a traditional Chinese medicine in China. It is widely used in clinic and the market demand is great. The quality difference between A. chinensis(Bunge) Koidz and A. japonica Koidz.ex Kitam is great, which seriously affects its clinical curative effect. Determination of Atractylosin by High-performance liquid chromatography. Transcriptomics coupled with Metabonomics for elucidating differences between the components in A. chinensis(Bunge) Koidz and A. japonica Koidz.ex Kitam. It was found that ACS, ACC, PAL and NOS were the key genes and metabolites related to the synthesis of atractylodes macrocephala. The decrease of salicylic acid content in A. chinensis(Bunge) Koidz may lead to the decrease of its ability to mediate the elicitor of endophytic fungi, resulting in the down-regulation of the expression of TGAL4 transcription factor and the up-regulation of DOGL3 transcription factor in A. chinensis(Bunge) Koidz cells, which may affect the accumulation of Atractylosin. The study above provides a theoretical basis for elucidating the biosynthesis pathway of atractylosin in A. chinensis(Bunge) Koidz.

Project description:Sorghum (Sorghum bicolor) is one of the world's most important cereal crops. S. propinquum is a perennial wild relative of S. bicolor with well-developed rhizomes. Functional genomics analysis of S. propinquum, especially with respect to molecular mechanisms related to rhizome growth and development, can contribute to the development of more sustainable grain, forage, and bioenergy cropping systems. In this study, we used a whole rice genome oligonucleotide microarray to obtain tissue-specific gene expression profiles of S. propinquum with special emphasis on rhizome development. A total of 548 tissue-enriched genes were detected, including 31 and 114 unique genes that were predominantly expressed in the rhizome tips (RT) and internodes (RI), respectively. Further GO analysis indicated that the functions of these tissue-enriched genes corresponded to their characteristic biological processes. A few distinct cis-elements, including ABA-responsive RY repeat CATGCA, sugar-repressive TTATCC, and GA-responsive TAACAA, were found to be prevalent in RT-enriched genes, implying an important role in rhizome growth and development. Comprehensive comparative analysis of these rhizome-enriched genes and rhizome-specific genes previously identified in S. propinquum indicated that phytohormones, including ABA, GA, and SA, are key regulators of gene expression during rhizome development. Co-localization of rhizome-enriched genes with rhizome-related QTLs in rice and sorghum generated functional candidates for future cloning of genes associated with rhizome growth and development. In conclusion, a whole rice genome oligonucleotide microarray was used to profile gene expression across five tissues of the perennial wild sorghum S. propinquum. Expression patterns of the five tissues were consistent with the different functions of each organ, and RT- and RI-enriched genes revealed clues regarding molecular mechanisms of rhizome development. Plant hormones, including ABA, GA, and SA, function as key regulators of rhizome gene expression and development. To shed further light on the identities of rhizome-specific genes, rhizome-enriched candidates were identified using QTL co-localization and comparative analysis.

Project description:Ulcerative colitis (UC), a subtype of inflammatory bowel disease (IBD), is characterized by chronic and relapsing mucosal inflammation initiating in the rectum and extending upward through part or the entire colon in a continuous fashion. Currently, 5-aminosalicylic acid agents, corticosteroids, immunomodulators, and surgery are the main treatments for UC, which are limited in clinical practice due to common nonadherence, serious adverse effects and heavy financial burden. Atractylodes macrocephala Koidz. (Baizhu) has been used for improving gastrointestinal function and treating digestive disorders for thousands of years, and is currently one of the most frequently used traditional Chinese medicines for the treatment of UC. The purpose of this study was to evaluate the effect and underlying mechanism of polysaccharide from Atractylodes macrocephala Koidz. (PAMK) on UC based on a mouse model of dextran sodium sulfate (DSS)-induced colitis. To investigate the mechanism of therapeutic effect of PAMK on colitis, we performed the microarray analysis of colonic tissues in Control, DSS and DSS+PAMK groups. The results indicated that the beneficial effect of PAMK on DSS-induced colitis may rely on the regulation of the immune response.

Project description:Introduction: Cisplatin (DDP) is a highly effective and broad-spectrum chemotherapeutic agent. However, its clinical application is limited by severe hepatotoxicity. Currently available hepatoprotective agents are often associated with insufficient efficacy or side effects. Therefore, developing new hepatoprotective agents that are highly efficient and low in toxicity is urgent. Objectives: Atractylodes macrocephala Koidz. is a well-known traditional Chinese medicine for strengthening the spleen function. According to the theory of traditional Chinese medicine, “When liver disease occurred, the spleen should be strengthened first” suggests that A. macrocephala Koidz. might possess hepatoprotective properties. Therefore, this study focuses on the molecular mechanism by which the active factors of A. macrocephala Koidz. exert their liver-protective effects. Methods: Animal experiments were adopted to screen the active factors of A. macrocephala Koidz. Multi-omics integrated analysis, including gut microbial diversity, transcriptomics, and metabolomics, combined with fecal microbiota transplantation (FMT) experiments were performed to clarify the mechanism of liver-protective effects of atractylenolide III (AT-III). Results: We found that AT-III-derived from A. macrocephala Koidz. significant decreased ALT, AST levels and hepatic pathological symptoms in DDP-induced acute liver injury (ALI). AT-III alleviated DDP-induced ALI via the “gut-liver axis” by inhibiting the ACT1/TRAF6/NF-κB-mediated IL-17 signaling pathway. Eight distinct microbial species, including Parabacteroides and Alloprevotella, were identified with significant differences. Besides, metabolite N6-(1,2-dicarboxyethyl)-AMP was identified as a novel biomarker for hepatotoxicity. Meanwhile, the FMT results indicated that the intestinal microbiota has a better alleviating effect on ALI than the metabolites of the intestinal microbiota.
