Project description:Different intensities of high temperatures affect the growth of photosynthetic cells in nature. To elucidate the underlying mechanisms, we cultivated the unicellular green alga Chlamydomonas reinhardtii under highly controlled photobioreactor conditions and revealed systems-wide shared and unique responses to 24-hour moderate (35oC) and acute (40oC) high temperatures and subsequent recovery at 25oC. We identified transcripts/proteins with unique differential regulation at 35oC, uncovering previously overlooked novel elements in response to moderate high temperature. Heat at 35oC increased transcripts/proteins involved in gluconeogensis/glyoxylate-cycle for carbon uptake, promoted growth, and increased starch accumulation. Heat at 40oC inhibited growth, resulting in carbon uptake over usage and increased starch accumulation. Heat at 35oC transiently arrested the cell cycle followed by partial synchronization while 40oC inhibited DNA replication and arrested the cell cycle. Both high temperatures induced photoprotection, while 40oC decreased photosynthetic efficiencies, distorted thylakoid/pyrenoid ultrastructure, and affected the carbon concentrating mechanism. We demonstrated increased transcript/protein correlation during heat, which decreased during recovery, suggesting reduced post-transcriptional regulation during heat may help coordinate heat tolerance activities efficiently. During recovery after both treatments, transcripts/proteins related to DNA synthesis increased while those involved in photosynthetic light reactions decreased. We propose down-regulating photosynthetic light reactions during DNA replication benefits cell cycle resumption by reducing ROS production. Our results provide potential targets to increase thermotolerance in algae and crops.

Project description:Different high temperatures adversely affect crop and algal yields with various responses in photosynthetic cells. The list of genes required for thermotolerance remains elusive. Additionally, it is unclear how carbon source availability affects heat responses in plants and algae. We utilized the insertional, indexed, genome-saturating mutant library of the unicellular, eukaryotic green alga Chlamydomonas reinhardtii to perform genome-wide, quantitative, pooled screens under moderate (35oC) or acute (40oC) high temperatures with or without organic carbon sources. We identified heat-sensitive mutants based on quantitative growth rates and identified putative heat tolerance genes (HTGs). By triangulating HTGs with heat-induced transcripts or proteins in wildtype cultures and MapMan functional annotations, we present a high/medium-confidence list of 933 Chlamydomonas genes with putative roles in heat tolerance. Triangulated HTGs include those with known thermotolerance roles and novel genes with little or no functional annotation. About 50% of these high-confidence HTGs in Chlamydomonas have orthologs in green lineage organisms, including crop species. Arabidopsis thaliana mutants deficient in the ortholog of a high-confidence Chlamydomonas HTG were also heat sensitive. This work expands our knowledge of heat responses in photosynthetic cells and provides engineering targets to improve thermotolerance in algae and crops.

Project description:Algal photo-bio hydrogen production, a promising method for producing clean and renewable fuel in the form of hydrogen gas, has been studied extensively over the last few decades. In this study, microarray analyses were used to obtain a global expression profile of mRNA abundance in the green alga Chlamydomonas reinhardtii at five different time points before the onset and during the course of sulphur depleted hydrogen production. The present work confirms previous findings on the impacts of sulphur deprivation but also provides new insights into photosynthesis, sulphur assimilation and carbon metabolism under sulphur starvation towards hydrogen production. For instance, while a general trend towards repression of transcripts encoding photosynthetic genes was observed, the abundance of Lhcbm9 (encoding a major light harvesting polypeptide) and LhcSR1 (encoding a chlorophyll binding protein) was strongly elevated throughout the experiment, suggesting remodeling of the photosystem II light harvesting complex as well as an important function of Lhcbm9 under sulphur starvation. This study presents the first global transcriptional analysis of C. reinhardtii during hydrogen production using five major time points at Peak Oxygen, Mid Oxygen, Zero Oxygen, Mid Hydrogen and Peak Hydrogen. Project description:Genus Halimeda is a worldwide distributed reef-building organism facilitated by fast growth and a rapid turnover rate in tropical and sub-tropical oceans. It is an excellent candidate for calcareous algae research because its calcification occurs extracellular, which deposits aragonite in the semi-enclosed inter-utricle space. In this study, we sampled the thalli of Halimeda macroloba in Sanya Bay of the South China Sea. The mean winter and summer water temperatures in Sanya Bay were 25.05℃ and 28.72℃, respectively. Additionally, the mean surface and bottom layer water temperatures in summer were 29.61℃ and 27.23℃. In order to simulate the natural temperature range, a factorial experimental design with three treatments (25°C, 28°C, and 31°C) was set. The goal of this study was to decipher the underlying traits that may facilitate H. opuntia to maintain high productivity and active physiology under different temperatures. Here, we employed iTRAQ-based quantitative proteomics and untargeted metabolomics approaches to investigate the protein and metabolite profiles of H. macroloba.
