Project description:In recent years, extensive mortality caused by global-scale bleaching events has led to the rapid degradation of reef structures, resulting in severe environmental consequences. The hydrocoral Millepora complanata (fire coral) plays a critical role in reef structure and relies on a symbiotic relationship with Symbiodiniaceae. Environmental stressors derived from climate change, such as UV radiation and elevated temperatures disrupt this symbiosis, leading to bleaching and threatening reef survival. Despite its importance in reef building, the molecular response of this cnidarian to thermal stress have been poorly explored. For this reason, the objective of this study is to investigate the proteomic response of M. complanata to bleaching during the 2015–2016 El Niño event. Fragments from non-bleached and bleached colonies of the hydrocoral M. complanata were collected from a coral reef in the Mexican Caribbean, and proteomic extracts were analyzed using nano-liquid chromatography–tandem mass spectrometry (nano-LC-MS/MS). Uni- and multivariate analyses were applied to identify significant differences in protein abundance. A total of 52 proteins showed differential abundance, including 24 that showed increased expression and 28 whose expression decreased in bleached fragments. Differentially abundant proteins were associated with amino acid biosynthesis, carbohydrate metabolism, cytoskeleton organization, DNA repair, extracellular matrix composition, redox homeostasis, and protein modification. These findings indicate that heat stress triggers molecular responses involving protein refolding, enhanced vesicle transport, cytoskeletal reorganization, and adjustments in redox activity, thereby contributing to a better understanding of the molecular mechanisms underlying bleaching in reef-building hydrocorals.

Project description:<p>Benthic organisms sustain coral reefs through their growth and metabolism, but less is known about how their released metabolites influence reef seawater microorganisms. To investigate metabolite composition of benthic exudates and their ecological significance for reef microbial communities, we harvested exudates from six species of Caribbean benthic organisms including stony corals, octocorals, and an invasive encrusting algae, and subjected these exudates to untargeted and targeted metabolomics approaches using liquid chromatography-mass spectrometry. Incubations with reef seawater microorganisms were conducted to monitor changes in microbial community composition using 16S rRNA gene sequencing and abundance in relation to exudate source and three specific metabolites. Exudates tended to be enriched in amino acids, nucleosides, and vitamins, indicating that benthic organisms contribute labile organic matter to reefs. The phytohormone indole-3-acetic acid was detected in octocoral exudates, suggesting that this metabolite facilitates microbial interactions within and outside of benthic organisms. Exudate compositions were species-specific and significantly enriched in the indole class of metabolites. Microbial abundances and specific microbial taxa responded differently in relation to exudates from stony corals and octocorals, demonstrating the link between benthic organismal composition, metabolite exudates, and microbial growth. Conversely, microbial communities did not respond to additions of the individual metabolites, suggesting that reef microorganisms likely provide diverse metabolite pools that support microbial growth. Project description:<p>Sponges are sessile filter-feeders that can process vast amounts of water and are known to influence the chemistry of the surrounding seawater. There has been limited work however to understand the extent to which sponges alter dissolved organic matter (DOM), yet in areas where sponges are abundant, sponges may contribute significantly to the reef seawater profile of DOM. This work provides an in-depth examination of six prevalent sponges on Caribbean reefs and how they alter DOM and other seawater nutrients. Incurrent and excurrent seawater samples were collected for each of the six sponge species and processed for: inorganic nutrients, fluorescent dissolved organic matter (fDOM), untargeted and targeted metabolomics, and particulate matter by flow cytometry. Sponges were sampled from two coral reef sites in the Florida Keys (Florida, USA): Looe Key reef and Wonderland reef in the southern Florida Keys. We found higher that sponges altered a relatively small subset of the DOM profile and were a net sink of most mass features from untargeted metabolomics. However, sponges also released some putatively labile metabolites and processed DOM in a species-specific manner. These results provide additional support for the large impact that sponges have in the dissolved nutrient profile on coral reefs and provide support for a species-specific impact, with some species altering the DOM profile, fDOM profile, and/or inorganic nutrients to a greater extent than other species. These results have implications for better understanding the influence of the sponge community on coral reef nutrient dynamics.</p>
