microRNA and the Posttranscriptional Response to Oxidative Stress During Neuronal Differentiation

Project description:Oxidative stress is an important environmental exposure associated with psychiatric disorders, but the underlying molecular mechanisms remain elusive. We recently observed enrichment of neurodevelopmental processes and diseases among a large number of differentially expressed genes during or after exposure to oxidative stress in an in vitro model of neuronal differentiation. To further explore the regulatory mechanisms that might account for a coordinated response to this exposure, we investigated changes in the expression of small non-coding miRNA in this system and estimated their interaction with differentially expressed mRNA. These molecules are thought to play a crucial role in brain development and its response to stress. Here we observed more than a hundred differentially expressed miRNAs, including 72 previously reported to be dysregulated in psychiatric disorders. The 7 most influential miRNAs associated with pre-treatment exposure were miR-138-5p, miR-96-5p, miR-34c-5p, miR-1287-5p, miR-497-5p, miR-195-5p, and miR-16-5p. These were sup-ported by at least 10 negatively correlated mRNA connections, and formed hubs in interaction network with 134 genes enriched with neurobiological function. Whereas in the co-treatment con-dition, miRNA-mRNA interaction pairs were enriched not only in neuronal processes, but also in cardiovascular and immunity-related disease ontologies. Interestingly, 12 differentially expressed miRNAs originated from the same genomic location, DLK1-DIO3, which encodes a schizophre-nia-associated miRNA signature. Collectively, these findings suggest that early exposure to oxida-tive stress, before and during prenatal neuronal differentiation, might increase the risk of mental illnesses in adulthood by disturbing the expression of miRNAs that regulate neurodevelopmental-ly significant genes and networks.

Expression profiles in Caenorhabditis elegans mutants lacking an intestinally expressed microRNA mir-60 that promote adaptive response against chronic oxidative stress

Project description:Recent revelations into microRNA function suggest that microRNAs serve as a key player in a robust adaptive response against stress in animals through their fine-tuning capability in gene expression. However, it remains largely unclear how a microRNA-modulated downstream mechanism contributes to the process of homeostatic adaptation. Here we show that loss of an intestinally expressed microRNA gene mir-60 in the nematode C. elegans promotes adaptive response against oxidative stress; animals lacking mir-60 dramatically extend lifespan under a mild and long-term oxidative stress condition, while they do not increase resistance against a strong and transient oxidative stress exposure. We found that canonical stress responsive factors, such as DAF-16/FOXO, are dispensable for mir-60 loss to enhance oxidative stress resistance. Gene expression profiles revealed that genes encoding lysosomal proteases and those involved in the xenobiotic metabolism and pathogen defense response are up-regulated by the mir-60 loss. Detailed genetic studies and computational microRNA target prediction suggest that endocytosis components and a bZip transcription factor gene zip-10, which functions in innate immune response, are directly modulated by miR-60 in the intestine. Our findings suggest that the mir-60 loss facilitates adaptive response against chronic oxidative stress by ensuring the maintenance of cellular homeostasis.

microRNA-125b Promotes Neuronal Differentiation in Human Cells by Repressing Multiple Targets

Project description:We report here the expression profile of microRNAs in human neuronal differentiation in the neuroblastoma cell line SH-SY5Y. Six microRNAs were significantly upregulated during differentiation induced by all-trans¬-retinoic acid and brain-derived neurotrophic factor. We demonstrated that ectopic expression of either miR-124a or miR-125b increases the percentage of differentiated SH-SY5Y cells with neurite outgrowth. Subsequently, we focused our functional analysis on miR-125b and demonstrated the important role of this miRNA in both spontaneous and induced differentiation of SH-SH5Y cells, based on neurite outgrowth and neuronal marker expression. In human neural progenitor ReNcell VM cells, miR-125b is also upregulated during differentiation and miR-125b ectopic expression significantly promotes neurite outgrowth. To identify the targets of miR-125b regulation, we profiled the global changes in gene expression following miR-125b ectopic expression in SH-SY5Y cells. miR-125b represses 164 genes that contain the seed match sequence of the microRNA and/or predicted to be direct targets of miR-125b by conventional methods. Pathway analysis suggests that a subset of miR-125b-repressed targets antagonize neuronal genes in several neurogenic pathways, thereby mediating the positive effect of miR-125b on neuronal differentiation. We have further validated the binding of miR-125b to the microRNA response elements of ten selected targets. Together, we report here for the first time the important role of miR-125b in human neuronal differentiation.
