These data were used to determine which genes are regulated by native HDL compared to components of HDL and categorize data based on shared sets of genes and distinct sets of genes regulated by each component. Each group had a n=5 and gene expression changes for each condition were compared to control PBS-treated HCAECs

Identification of potential abnormal methylation modified genes in coronary artery ectasia

Project description:Coronary artery ectasia (CAE) is a rare but easily recognized abnormality of coronary artery anatomy and morphology. Up to now, the pathogenesis of CAE remains unclear. Firstly, the peripheral venous blood samples of patients with CAE were obtained for the RNA sequencing to identify differentially expressed genes (DEGs), followed by functional analysis. Secondly, DNA methylation profile in CAE was downloaded from the dataset of GSE87016 to identify differentially methylated genes (DMGs). Lastly, after taking interaction genes between DEGs and DMGs, abnormal methylation modified genes were identified.

Transcriptomic analysis of human coronary artery endothelial cells stimulated with TNF-alpha in the presence and absence of ACSL3 siRNA

Project description:TNF-alpha has a number of pro-atherogenic effects in macrovascular endothelial cells, including induction of leukocyte adhesion molecules and chemokines. We investigated the role of acyl-CoA synthetase 3 (ACSL3) in the response of cultured human macrovascular endothelial cells to TNF-alpha. TNF-alpha induced ACSL3 both in human umbilical vein endothelial cells (HUVECs) and in human coronary artery endothelial cells (HCAECs). RNA sequencing demonstrated that knockdown of ACSL3 had no marked effects on the TNF-alpha transcriptome in HCAECs. Instead, ACSL3 was required for TNF-alpha-induced lipid droplet formation from fatty acids.
