DNA damage invokes mitophagy through a pathway involving Spata18

Project description:Mitochondria are vital for cellular energy supply and intracellular signaling after stress. Here, we aimed to investigate how mitochondria respond to acute DNA damage with respect to mitophagy, which is an important mitochondrial quality control process. Our results show that mitophagy increases after DNA damage in primary fibroblasts, murine neurons, and C. elegans neurons. Our results indicate that modulation of mitophagy after DNA damage is independent of the type of DNA damage stimuli used and that the protein Spata18 is an important player in this process. Knockdown of Spata18 suppresses mitophagy, disturbs mitochondrial Ca2+ homeostasis, affects ATP production, and attenuates DNA repair. Importantly, mitophagy after DNA damage is a vital cellular response to maintain mitochondrial functions and DNA repair.

Protein import motor complex reacts to mitochondrial misfolding by reducing protein import and activating mitophagy

Project description:Mitophagy is essential to maintain mitochondrial function and prevent diseases. It activates upon mitochondria depolarization, which causes PINK1 stabilization on the mitochondrial outer membrane. Strikingly, a number of conditions, including mitochondrial protein misfolding, can induce mitophagy without a loss in membrane potential. The underlying molecular details remain unclear. Here, we report that a loss of mitochondrial protein import, mediated by the pre-sequence translocase-associated motor complex PAM, is sufficient to induce mitophagy in polarized mitochondria. A genome-wide CRISPR/Cas9 screen for mitophagy inducers identifies components of the PAM complex. Protein import defects are able to induce mitophagy without a need for depolarization. Upon mitochondrial protein misfolding, PAM dissociates from the import machinery resulting in decreased protein import and mitophagy induction. Our findings extend the current mitophagy model to explain mitophagy induction upon conditions that do not affect membrane polarization, such as mitochondrial protein misfolding.

Regulatory circuits of mitophagy restricts cell death for mounting memory CD8+ T cell formation

Project description:Mitophagy, a central guarding process to maintain mitochondrial quality, is commonly impaired in human diseases, but its impacts in adaptive immunity remains unclear. Here, we found that Parkinson’s patients have reduced CD8+ memory T cell abundance and failed to form memory T cells upon COVID-19 vaccination. We further unveiled that regulators of mitophagy, including Parkin and NIX, were upregulated in response to IL-15 for supporting memory T cell formation. Our results indicate that the mitophagy machinery orchestrate survival and metabolic dynamics required for memory T cell formation.

Project description:Gemcitabine resistance in gallbladder cancer poses a significant challenge to patient prognosis. This study identified RUVBL2 as a critical factor contributing to gemcitabine resistance in gallbladder cancer. Further analysis, combining ChIP-seq and gene enrichment studies, revealed that RUVBL2 regulates the mitophagy pathway. This regulation helps maintain intracellular redox homeostasis in gallbladder cancer cells following gemcitabine treatment.

Urolithin A rewires inter-organellar communication via calcium signaling to promote mitophagy and longevity

Project description:Mitochondrial dysfunction and mitophagy deregulation are hallmark features of aging and age-related pathologies. Urolithin A (UA), a potent mitophagy inducer, is known to confer neuroprotection, maintain muscle integrity, and extend healthspan and lifespan across diverse species. Nonetheless, the molecular mechanisms underlying UA-mediated mitophagy remain largely unknown. Here, we demonstrate that UA treatment modulates cytosolic calcium levels, which are essential for initiating robust mitophagy in both neurons and muscles. Transcriptomic and proteomic analyses reveal that UA facilitates the reorganization of inter-organellar communication between endoplasmic reticulum (ER), lysosomes and mitochondria; processes that are highly dependent on calcium signaling. Our findings suggest that UA induces calcium release from the ER and enhances lysosomal activity, while downstream uptake of released calcium from mitochondria, ultimately leads to mitochondrial fission and the successful execution of mitophagy. Consistently, calcium chelation abolishes UA-induced mitophagy, leading to impaired muscle function and diminishes lifespan extension, underscoring the critical role of calcium dynamics. We further found that UA-induced calcium elevation triggers mitochondrial biogenesis through the activation of UNC-43/CaMKII and SKN-1/Nrf2; these pathways are also critical for healthspan and lifespan extension. In human cells, UA supplementation not only induces mitophagy but also enhances mitochondrial metabolism and prevents stress-induced senescence in a calcium-dependent manner. Ultimately, our findings uncover the mechanistic insights of UA-mediated geroprotection and underscore the central role of calcium dynamics in orchestrating the crosstalk and functional wiring of different cellular compartments, thereby sustaining energy homeostasis and overall organismal physiology.

Nitazoxanide impairs mitophagy flux through ROS-mediated mitophagy initiation and lysosomal dysfunction in bladder cancer

Project description:Bladder cancer is one of the most common malignancy in the urinary tract with high recurrence and drug resistance in clinics. Alternative treatments from existing drugs might be a promising strategy. Nitazoxanide (NTZ), an FDA-approved antiprotozoal drug, has got increasingly noticed because of its favorable safety profile and antitumor potential, yet the effects in bladder cancer and underlying mechanisms remain poorly understood. Herein, we find that NTZ induces mitochondrial damage and mitophagy initiation through PINK1-generated phospho-ubiquitin(pS65-Ub) and autophagy receptor-mediated pathway even in the absence of Atg5/Beclin1. Meanwhile, NTZ inhibits lysosomal degradation activity, leading to mitophagy flux impairment at late stage. Mitochondrial reactive oxygen species (ROS) production is critical in this process, as eliminating ROS with N-acetylcysteine (NAC) efficiently inhibits PINK1 signaling-mediated mitophagy initiation and alleviates lysosomal dysfunction. Co-treatment with NTZ and autophagy inhibitor Chloroquine (CQ) to aggravate mitophagy flux impairment promotes NTZ-induced apoptosis, while alleviation of mitophagy flux impairment with ROS scavenger reduced cell death. Moreover, we also discover a similar signaling response in the 3D bladder tumor spheroid after NTZ exposure. In vivo study reveals a significant inhibition of orthotopic bladder tumors with no obvious systemic toxicity. Together, our results uncover the anti-tumor activities of NTZ with the involvement of ROS-mediated mitophagy modulation at different stages and demonstrate it as a potential drug candidate for fighting against bladder tumors.

Pairwise multiplex CRISPR/Cas9 screen of mitophagy receptors in acute myeloid leukemia

Project description:The selective autophagic degradation of mitochondria via mitophagy is essential for preserving mitochondrial homeostasis and thereby disease maintenance and progression in acute myeloid leukemia. The process of mitophagy is orchestrated by a variety of mitophagy receptors whose interplay in AML is not well understood. Here, we established a dual multiplexed CRISPR screen targeting mitophagy receptors to elucidate redundancies and individual contributions of mitophagy receptors and gain a deeper understanding of the functional interactome governing mitophagy in AML.
