Project description:Background: Women represent the majority of Alzheimer’s disease patients and show typical symptoms. Genetic, hormonal and behavioral mechanisms have been proposed to explain gender differences in dementia prevalence. Whether sex differences exist in the epigenetic landscape of neuronal tissue during the progression of the disease is unknown. Methods: To investigate the differences of histone H3 modifications important for transcription we determined the ge-nome-wide profiles in K4me3, K27ac and K27me3 from brain cortexes of an Alzheimer mouse model (PSAPP). Gastrocnemius muscles were also tested since they are known to be different in the two sexes and are affected during the disease progression. Results: Correlation analysis dis-tinguished the samples based on sex for H3K4me3 and H3K27met3 but not for H3K27ac. The analysis of TSS signal distribution and of bounding sites revealed that the epigenetic landscape of gastrocnemius is more influenced than brain by sex, with the exception of H3K27me3 distribution on the X chromosome which showed sex-related differences in promoters belonging to behavior and cellular/ neuronal spheres in mice cortexes. Conclusions: The epigenetic landscape is slightly affected by sex in brain, with the exception of H3K27me3. On the other hand, a higher number of differences can be found in gastrocnemius.

Project description:Mechanisms of plasticity to acquire different cell fates are critical for adult stem cell (SC) potential, yet are poorly understood. Reduced global histone methylation is an epigenetic state known to mediate plasticity in cultured embryonic SCs and T-cell progenitors. Here we find histone H3 K4/K9/K27me3 levels actively reduced in adult mouse skin and hair follicle stem cells (HFSCs) during G0 quiescence. The level of marks over specific gene promoters did not correlate to mRNA level changes in quiescent HFSCs. Skin hypomethylation during quiescence was necessary for subsequent progression of hair homeostasis (cycle). Inhibiting BMP signal, a known HFSC anti-proliferative factor, elevated HFSC methylation in vivo during quiescence prior to proliferation onset. Furthermore, removal of proliferation factors and addition of BMP4 reduced histone methylases and increased demethylases mRNAs in cultured skin epithelial cells. We conclude that signalling couples hair follicle stem cell quiescence with reduced H3 K4/K9/K27me3 levels for proper tissue homeostasis.

Gene expression profiling of gastrocnemius of mini muscle mice

Project description:Few studies have investigated heterogeneity of selection response in replicate lines subjected to equivalent selection. We developed 4 replicate lines of mice based on high levels of voluntary wheel running (high runner or HR lines) while also maintaining 4 non-selected control lines. This led to the unexpected discovery of the HR mini-muscle (HRmini) phenotype, recognized by a 50% reduction in hindlimb muscle mass, which became fixed in 1 of the 4 HR selected lines. Here, we report genome-wide expression profiling describing transcriptome differences between HRnormal and HRmini medial gastrocnemius. This work provides a resource for understanding differences in muscle phenotypes in populations exhibiting high running capacity. Male mice were used from an animal model of 4 closed lines selectively bred for high voluntary wheel-running behaviour (HR) and 4 control lines bred without intentional selection, described in Swallow et al. Animals used in the current work were sampled from generation 37 at a mean age of 84 days (range = 79-86). For microarray analysis, we used gastrocnemius muscles (including both lateral and medial portions) from HRmini line #3 and HRnormal line #8 (n = 6, in each group). To obtain sufficient RNA, gastrocnemius samples of HRmini were pooled from left and right limbs from each animal, whereas HRnormal gastrocnemius was sampled from either left or right limb only.
