Single-nucleus transcriptomic profiles of glial cells in human dorsal root ganglion and spinal cord

Project description:Glial cells are present throughout the entire nervous system and paly a crucial role in regulating physiological and pathological functions, such as infections, acute injuries and chronic neurodegenerative disorders. The glial cells mainly include astrocytes, microglia, and oligodendrocytes in the central nervous system (CNS), and satellite glial cells (SGCs) in the peripheral nervous system (PNS). Although the glial subtypes and functional heterogeneity is relatively well understood in mice by recent studies using single-cell or single-nucleus RNA-sequencing, no evidence yet has elucidate the transcriptomic profiles of glia cells in PNS and CNS. Here, we used high-throughput single-nucleus RNA-sequencing to map the cellular and functional heterogeneity of SGCs in human dorsal root ganglion (DRG), and astrocytes, microglia, and oligodendrocytes in human spinal cord. In addition, we compared the human findings with previous single-nucleus transcriptomic profiles of glial cells from mouse DRG and spinal cord. This work will comprehensively profile glial cells heterogeneity and will provide a powerful resource for probing the cellular basis of human physiological and pathological conditions related to glial cells.

Mouse E14.5 Ret+ mechanoreceptors vs. laser captured spinal cord dorsal column midline cells

Project description:Our study examined a population of radial glial-like cells (RGLCs) in the dorsal spinal cord midline that we showed provide long-distance growth support for longitudinal rapidly adapting (RA) mechanoreceptor axons during development of spinal cord dorsal column. To evaluate potential molecular markers of these cells, we isolated the RGLCs using hematoxylin and eosin staining to visualize the cell bodies in the dorsal column midline from E14.5 mouse embryos, and used laser capture microdissection for each sample ("LCM"). To compare transcript expression to the adjacent RA mechanoreceptive axons, we performed FACS of dorsal root ganglion of E14.5 Ret-Tdtomato+ RA mechanoreceptors. Our analyses revealed a high enrichment of radial glial-specific markers in the LCM replicates compared to Ret-Tdt samples. In contrast, neuronal-specific markers were more highly enriched in the Ret-Tdt samples, as expected. These data suggest the midline RGLCs are of radial glial identity. Others may find these data helpful in determining potential RGLC-mechanoreceptor molecular interactions in subsequent studies.

Proteomic profile of (Laser-dissected) Motoneurons, Ependymal Cells and Dorsal Root Ganglia Cells, and after Spinal Cord Injury in Rat

Project description:Spinal cord injury triggers extensive molecular remodeling in neural and glial compartments. To resolve these processes at cell-type-specific resolution, we performed quantitative proteomic profiling of laser-microdissected motor neurons (MNs), dorsal root ganglia (DRG), and ependymal cells (EC) from control Wistar rats and 2 or 6 weeks after complete spinal cord transection at Th10. Retrogradely traced MNs innervating the soleus (SOL; 172 ± 39 cells) and tibialis anterior (TA; 262 ± 74 cells) muscles, ECs from the central canal (L3–L6) were microdissected from spinal cord slices with Leica LMD7000 Laser Microdissection System. Bilateral DRG (L3–L5) were also isolated. Samples from five animals per group were analyzed using TMT-based LC–MS/MS, yielding 1 221 proteins in SOL MNs, 1 186 in TA MNs, 1,520 in ECs, and 5 087, 3 740, and 3 086 in DRG L3, L4, and L5, respectively. The dataset supports cell-type-specific characterization of molecular responses after spinal injury.

Project description:This experiment aims at characterizing the transcriptome of embryonic mouse dorsal spinal cord. Dorsal spinal cords dissected from litters of E14.5 wild type embryos of unknown sex were processed for RNA extraction using Trizol and RNeasy Mini kit (Qiagen) extraction procedures. Five replicates of wild type embryos were analyzed, each sample with tissue pooled from three embryos.

Project description:An exquisite example of form serving function is the dorsal horn of the spinal cord, the gateway to the central nervous system for sensory information from the body. Each sensory input to the dorsal horn targets a specific address within its laminated arrangement of diverse neuronal populations. However, it is not known how this organization emerges during development from an apparently homogenous pool of neural progenitors. Here, we found that both the excitatory and inhibitory cell families of the mouse dorsal horn were born in successive waves as temporal cohorts. The excitatory families then settled into a chronotopic map that transformed their birth order into the dorsal laminae. Diversification of families into refined neuron types was mediated by a dorsal-ventral progenitor gradient of Zic transcription factors. This work uncovered fundamental temporal and spatial factors that establish the cell types and structure of the spinal cord dorsal horn.
