Project description:A total of 4276 lncRNAs (1426 upregulated and 2850 down-regulated) and 9230 mRNAs (5187 upregulated and 4043 down-regulated) were differently expressed in SHR-C compared with WKY, whereas 265 lncRNAs (65 up-regulated and 200 down-regulated) and 267 mRNAs (132 upregulated and 135 down-regulated) were differently expressed after BSJY treatment (fold change >2. 0 and P value< 0. 05). Taking the intersection of the two sets, the analysis found that 8 lncRNAs and 35 mRNAs were upregulated, whereas 11 lncRNAs and 15 mRNAs were downregulated in the WKY vs. SHR-C group and SHR-Bh group (fold change > 2. 0 and P value < 0. 05). It showed the heatmap that were constructed using fold change, It showed the Volcano plots that were constructed using fold change values and P values and revealed the relationship between fold change and statistical significance.

Project description:Using high-throughput antibody microarray, through cross-sectional sample detection and verification of samples that had undergone physical changes over time, it was found that people with balanced constitution and dampness constitution in Chinese medicine showed differences in serum protein expression. The differences were expressed as the level changes of 19 proteins such as Dectin-2, Siglec-7, AIF and TACI. The research results provided the basis for the scientific expression of traditional Chinese medicine (TCM) constitution.

Project description:To identify biomarkers regulated by traditional Chinese medicine Astragalus membranaceus Fischer Bge. var. mongolicus Bge. Hsiao in colorectal cancer. We have identified several differentially expressed genes including microRNAs using Affymetrix HTA-2.0 array. In this dataset, we include the expression data obtained from colon cancer cell line HCT116 grafted into nude mice. The mice was treated either water or traditional Chinese medicine Astragalus membranaceus for 28 days. These data are used to obtain 1425 genes that are differentially expressed in response to Astragalus membranaceus treatment.

Traditional Chinese Medicine JianPiHuaTan Formula Improving Quality of Life and Survival in Patients with Colorectal Cancer through RAS/RAF Downstream Signaling Pathways

Project description:Abstract Objective: JianPiHuaTan Formula (JPHTF), a traditional Chinese medicine (TCM), has been utilized as an adjunctive therapy for colorectal cancer (CRC). The study aims to evaluate the potential clinical benefits of JPHTF and its effectiveness in inhibiting tumor growth. Methods: 300 stage II/III CRC patients and 412 advanced CRC patients were enrolled to verify the clinical value of JPHTF in CRC treatment. Furthermore, CRC patient-derived xenograft (PDX) mice were utilized to investigate the regulatory mechanisms of JPHTF. Results: JPHTF significantly improved abdominal distension, shortness of breath, drowsiness, loss of appetite, sleep, and tiredness in stage II/III CRC patients, thereby improving their quality of life. Simultaneously, JPHTF served as a supportive therapy in extending the overall survival (OS) of stage IV CRC patients with RAS/RAF mutations undergoing chemotherapy. Additionally, JPHTF effectively impeded tumor progression in CRC PDX models with RAS mutation, accompanied by a reduction in tumor cell content in the JPHTF group. Transcriptomic analysis revealed the involvement of the Hippo and Hedgehog signaling pathways in JPHTF-mediated CRC inhibition. Furthermore, mice in the JPHTF group exhibited increased immune cell infiltration. Conclusions: These findings suggested that JPHTF may inhibits tumor growth in CRC with RAS mutation by modulating RAS/RAF downstream signaling pathways, specifically the Hippo and Hedgehog signaling, leading to increased immune cell infiltration.
