Project description:Gut microbiota in Machado Joseph Disease (MJD)
Project description:Spinocerebellar ataxia type 3 (SCA3), also called Machado-Joseph disease (MJD), is a polyQ neurodegenerative disease that is still seeking cure. We used SCA-3 84Q transgenic mice as a disease model to study the therapeutic effect of insulin-like growth factor 1 (IGF-1). Transcriptomic and metabolomic profiles were established and compared between the control (15Q), disease(84Q) and treatment(84Q+IGF-1) groups using RNA-seq and metabolic network analysis. SCA3-84Q mice showed a decrease in glycolysis, lipolysis and mitochondrial oxidative phosphorylation, and an increase in apoptosis, and IGF-1 treatment reversed the alterations slightly.
Project description:Proteomic analysis of mouse livers from animals maintained on 6-week diet of alcohol with a 10mg/kg ip LPS or saline (control) injection 24hr prior to sacrifice. Livers were extracted using a sequential extraction approach similiar to the method of de Castro Bras LE, Ramirez TA, DeLeon-Pennell KY, Chiao YA, Ma Y, Dai Q, et al. Texas 3-step decellularization protocol: looking at the cardiac extracellular matrix. J Proteomics 2013;86:43-52. Data collection and analysis are described in Massey VL, Dolin CE, Poole LG, Hudson SV, Siow DL, Brock GN, Merchant ML, Wilkey DW, Arteel GE.
