Phenotypic characterization of a Col6a3 knockout mouse model targeting exon 3

Project description:Mutations in COL6A3 lead to collagen VI-related myopathies due to COL6A3 loss. Therefore, studying the consequence of Col6a3 deregulated expression in mouse models is relevant, but the Col6a3 knockout mouse models currently available through a public repository does not entirely abolish COL6A3 protein expression. Here, we present the validation and phenotypic characterization of a novel CRISPR-based knockout mouse model targeting Col6a3 exon 3 (Col6a3 d3/d3). In this mouse model, Col6a3 mRNA is still expressed at a similar level to that of wild-type littermates, although the expected protein is undetectable. Histological analysis of Col6a3 d3/d3 quadriceps revealed an abnormally high frequency of muscle cells with a centralized nuclei, consistent with a muscular dystrophy phenotype. Interestingly, Col6a3 d3/d3 mice are smaller in size with their fat, muscle, and bone to body ratios kept proportional compared to wild type littermates. Thus, we developed a novel Col6a3 knockout mouse model that could be further used to study Col6a3 biology and collagen VI-associated diseases.

KTN-FLAG-IP for identification of KTN1-variant specific binding proteins

Project description:The endoplasmic reticulum (ER) forms sheet or tubule organization, and functions in regulating various cellular processes. Kinectin 1 (KTN1), an integral ER sheet protein involved in ER organization and translation elongation, has alternative exons on the cytoplasmic side, but their roles remain elusive. Here we found that the multi-tRNA synthetase complex (MSC), which is composed of eight aminoacyl-tRNA synthetases (ARSs) and three non-enzymatic proteins, specifically interacts with KTN1 depending on the alternative exon V2 in mouse and human cells. Through a combination of KTN1 knockout and rescue using variant-specific KTN1 expression, we show that the V2 exon is essential for MSC recruitment to the ER, where it contributes to the formation of rough ER stack organization. Our study thus identifies a novel role of KTN1 splicing variant in regulating the ER localization of the MSC, which is likely related to ex-translational activities of MSC to regulate ER sheet organization.

Comparative Transcriptomic Analysis of Embryonic Hearts in Atf4 Exon 2 and Exon 2–3 Conditional Knockout Mice

Project description:This dataset comprises RNA sequencing (RNA-seq) profiles of E11.5 embryonic hearts from two Atf4 conditional knockout (cKO) mouse models, targeting different exons of the gene. In one model, exons 2 and 3 were floxed and excised in cardiomyocytes using the Xmlc2-Cre driver, while the second model involved deletion of exon 2 only. The aim of this study is to determine whether Atf4 exon-targeting strategy influences transcriptomic outcomes and to evaluate the impact of ATF4 deficiency on early cardiac development. RNA-seq was performed on ventricular tissues from both models and their respective littermate controls, each with four biological replicates. The dataset serves to clarify the necessity and function of ATF4 in the embryonic heart using refined conditional knockout strategies.

Minimal expression of dysferlin prevents development of muscular dystrophy in dysferlin exon 40a knockout mice

Project description:Dysferlin is an essential muscle membrane repair protein and autosomal recessive mutations in its gene result in progressive muscular dystrophies collectively called dysferlinopathies. We previously showed that calpain-mediated cleavage within dysferlin exon 40a releases a 72kDa C-terminal minidysferlin recruited to injured sarcolemma. In the current study, we hypothesised that knocking out exon 40a will lead to defective membrane repair and development of muscular dystrophy over time. To address this hypothesis, we created three exon 40a knockout (40aKO) mouse lines with dysferlin protein levels ranging from ~80% of wildtype (WT) in 40aKO-3, ~50% in 40aKO-2 and ~10-20% in 40aKO-1 mice. Histopathological analysis of skeletal muscles harvested from all 12-month-old 40aKO mice showed little evidence of dystrophic features seen in dysferlin-null BLAJ mice. Lipidomics analysis on 18wk old quadriceps showed that all 40aKO lines had a similar lipidomic profile to WT and distinct from BLAJs. The proteomic profile of 40aKO lines was intermediate between that of WT and BLAJs. Laser membrane damage assays showed normal membrane repair capacity in all three 40aKO lines. These results collectively indicate that ~10-20% of dysferlin protein expression is sufficient for maintenance of the lipidome and membrane repair capacity, and crucially prevents development of muscular dystrophy.
