Project description:We described, for the first time, a robust protocol for direct differentiation of hPSCs into SGEP by mimicking the activity of retinoic acid and WNT signaling. These hPSC-derived SGEP expressed SOX9, KRT5 and KRT19, which were the important progenitor markers of developing salivary glands. The CD24 and α-SMA positive cells that are capable to restore the function of injured salivary glands were also present in SGEP cultures. Importantly, RNA-seq was performed to examine the similarity of SGEP to human fetal and mature salivary glands. The result revealed the SGEP resemble the transcript profile of human fetal submandibular glands. Therefore, we successfully induced hPSCs into SGEP and demonstrated that these SGEP potentially serves as models for studying mechanism underling salivary development, and as cell sources for clinical use.

Project description:The aim of this study was to use pluripotent stem cells (PSCs) as a starting point to progressively differentiate into induced salivary gland progenitor cells (iSGPCs) by mimicking salivary gland development. In this study, we further explored and optimized the passage strategy and organoid formation protocol of iSGPCs based on our previous studies. After 17 days of differentiation, PSC-derived iSGPCs exhibited a marker expression profile similar to that of adult salivary gland progenitors, including KRT5, KRT14, KRT19, and CD49F. Functionally, iSGPCs exhibit self-renewal capacity, are able to form salivary gland-like branching organoids, and have therapeutic potential in models of radiation-induced salivary gland hypofunction.

Project description:The differentiation of pluripotent stem cells has been broadly used in studying the disease mechanism and development process. We previously described a method for differentiating human pluripotent stem cells (hPSCs) into salivary gland epithelial progenitors (SGEPs). Here, to investigate whether the hPSCs-derived SGEPs are capable of modeling the characteristics of cystic fibrosis (CF), a disease impact SG function caused by mutation of cystic fibrosis transmembrane conductance regulator (CFTR) gene, the CFTR knockout hPSCs were differentiated into CF-SGEPs using the same protocol. Firstly, we successfully generated CFTR knockout hPSCs with reduced CFTR protein expression using the CRISPR-Cas9 system. After 16-day of differentiation, the protein expression of CFTR was also decreased in SGEPs generated from CFTR knockout hPSCs. RNA-sequencing shows that multiple genes modulating SG development, function and were down-regulated and positive regulators of inflammation were up-regulated in CF-SGEPs, which was correlated with salivary phenotype in CF patients. These results demonstrated that suppression of CFTR disrupted the differentiation of hPSCs-derived SGEPs, which modeled the SG development of patients with CF. In summary, this study not only provided the proof that the hPSCs-derived SGEPs could serve as manipulatable and easily accessible models to study the SG developmental diseases, but also provided new avenues for the study of the CF mechanism.

Project description:The derivation of salivary gland (SG) progenitors from pluripotent stem cells (PSCs) presents significant potential for developmental biology and regenerative medicine. However, the existing protocols for inducing SG include limited factors, making it challenging to mimic the in vivo microenvironment of embryonic SGs. We reported a cocktail factor approach to enhance the differentiation of mouse embryonic stem cell (mESC)-derived oral epithelium (OE) into SG progenitors through a three-dimensional co-culture method. Upon confirming that the embryonic SG can promote the differentiation of mESC-derived OE, we performed RNA sequence analysis to identify factors involved in the differentiation of SG progenitors. Our findings highlight several efficient pathways related to SG development, with frequent appearances of four factors: IFN-γ, TGF-β2, EGF, and IGF-1. The combined treatment using these cocktail factors increased the expression of key SG progenitor markers, including Sox9, Sox10, Krt5, and Krt14. However, absence of any one of these cocktail factors did not facilitate differentiation. Notably, aggregates treated with the cocktail factor formed SG epithelial-like structures and pre-bud-like structures on the surface. In conclusion, this study offers a novel approach to developing a differentiation protocol that closely mimics the in vivo microenvironment of embryonic SGs. This provides a foundation for generating PSC-derived SG organoids with near-physiological cell behaviors and structures.

Project description:Derivation of transplantable T cell progenitors from human pluripotent stem cells is the key step towards regenerative therapy of hereditary and acquired immunodeficiencies. Here, we describe an original methodology for differentiation of genetically non-modified human pluripotent stem cells (hPSCs) that yields T cell progenitors capable for direct and unmediated reconstitution of mouse thymus. The hPSC-derived T cell progenitors exhibited a strong thymus and spleen homing potential and developed into single positive human T cells that could enter circulation. Detailed single cell transcriptome analysis confirmed the similarity of hPSC-T cell progenitors with their in vivo counterparts. The transcription profiling attested the emergence of cells that displayed the transcription signature of early T cell progenitors.

Project description:Severe salivary gland injury causes long-term structural and functional impairment, yet the epithelial populations associated with repair remain incompletely defined. Here, we identify NGFR as a surface marker of a basal duct-associated epithelial population with organoid-forming capacity in the human salivary gland and show that Ngfr marks a conserved injury-responsive epithelial population in the mouse salivary gland. Single-cell RNA sequencing of human salivary gland tissue revealed that NGFR was enriched in a restricted basal duct subpopulation distinct from differentiated epithelial and myoepithelial compartments. NGFR-positive cells exhibited a progenitor-like transcriptional profile and were enriched at early pseudotime positions in inferred ductal-acinar differentiation trajectories. Functionally, NGFR-positive cells isolated from human tissue and organoids showed enhanced organoid-forming capacity, and NGFR-enriched organoids engrafted into injured mouse salivary glands. In mouse salivary glands, Ngfr marked a rare epithelial population whose isolated Ngfr-positive fraction showed enriched organoid-forming activity and was detected in injury-associated ductal regions after duct ligation and local inflammatory injury. Lineage tracing using Ngfr-CreERT2; Rosa26-tdTomato mice further showed that Ngfr-lineage cells contribute to ductal and acinar epithelial compartments during post-injury regeneration. Together, these findings define NGFR in human and Ngfr in mouse as conserved markers of an injury-responsive basal duct epithelial population that can be prospectively isolated and contributes to ductal and acinar epithelial compartments during post-injury salivary gland regeneration.

Project description:Regeneration of human cartilage and bone remains a challenge due to the limitations from current stem cell sources. Here, we developed a four-day differentiation strategy that achieved near uniform derivation (>98.5%) of SOX9+ sclerotomal progenitors from multiple human pluripotent stem cells (hPSCs). Importantly, SOX9+ sclerotomal progenitors exhibited typical bipotential of mesenchymal progenitors during skeletal development. Upon lineage-specific induction, they were able to differentiate either into chondroprogenitors that could repair articular cartilage defects and early chondrocytes that could undergo endochondral ossification for human bone formation. Furthermore, we identified ITGA9 as the specific surface marker that facilitated reporter-independent isolation of SOX9+ sclerotomal progenitors and established an in vitro culture system that could expand these cells for at least 15,000-fold. Collectively, these findings highlight the SOX9+ sclerotomal progenitors as a promising stem cell source for next-generation human cartilage/bone bioengineering.
