Project description:The repertoire of tumor-infiltrating T cells is a novel perspective to characterize effective anti-tumor T cell responses. Previous studies reported that the oligoclonal expansion in tumor T-cell repertoire is associated with anti-tumor effects. However, the contribution of the expansion of diverse T-cell clones remained unclear. We demonstrated that the polyclonal fraction of tumor-reactive T-cell repertoire consisting of relatively minor clones, increased in tumor-bearing mice treated with anti-PD-L1 or anti-CD4 monoclonal antibodies (mAbs), while the oligoclonal fraction consisting of major clones was unchanged. Moreover, the polyclonal fraction was enriched with progenitor exhausted T cells, essential for a durable anti-tumor response, and more dependent on CCR7+ migratory dendritic cells, responsible for priming tumor-reactive T cells in the tumor-draining lymph node (dLN). These results proposed that the expansion of diverse tumor-reactive clones, “clonal spreading,” is an important mechanism by which anti-PD-L1 and anti-CD4 treatments exert robust and durable anti-tumor T-cell responses.

Project description:Depletion of CD4+ cells by anti-CD4 monoclonal antibody (anti-CD4 mAb) induces expansion of tumor-reactive CD8+ T cells and exhibits strong antitumor effects in several murine tumor models. However, whether the anti-CD4 mAb treatment activates particular or a broad variety of tumor-reactive CD8+ T cell clones is not answered. To investigate the changes of TCR repertoire induced by the anti-CD4 mAb treatment, we performed unbiased high throughput TCR sequencing in a B16F10 mouse subcutaneous melanoma model.

Expression data from OL and OC treated B16F10 cells

Project description:Oleocanthal (OL) and Oleacein (OC) are enriched components in Olive (oil and leaves). However, their effects on skin have not been widely studied compared to Oleuropein (OP). Here, we perfomed global gene expression profiling to screen the potential effects of OL and OC on the skin. Results showed that OL and OC have effects on skin development and keratinocyte differentiation by upregulation of key markers. Furthermore, OL and OC were effectively downregulated several melanogenic genes and it suggests that OL and OC can have a potential effect for pigmentation. B16F10 were murine melanoma cells line, treated with 5 µM of OL and OC for 24 hours. Microarray gene expression profiling was conducted technical replicates.
