Differential expression of Transcriptomics and Metabolomics in different developmental periods of poria cocos

Project description:<p>Poria cocos, a traditional medicinal fungus, exhibits a wide array of biological activities. Through the integration of metabolomic and transcriptomic analyses, we have determined that the development of Poria cocos mycelia is predominantly propelled by biochemical reactions triggered by oxidative stress. Within this process, the mitogenactivated protein kinase (MAPK) signaling pathway assumes a pivotal regulatory function. As Poria cocos reaches its maturation stage, we have identified several functional substances, such as oleanolic acid, 18β-glycyrrhetinic acid, hawthorn acid, ergosterol and dehydroergosterol, among others. Our findings have unveiled the molecular mechanism underlying the formation of sclerotia in Poria cocos, providing crucial insights into its biological traits and its response to environmental stressors. These functional substances underscore the potential of Poria cocos in the development and application of nutraceuticals.</p>

RNA-seq of mouse small intestine samples from different Fecal Microbiota Transplantation treatment groups

Project description:The aim of this project was to explore the role of gut microbiota in the development of small intestine. The gut microbiota from different groups was used to treat the mice for 1 or 2 weeks. Then the small intestine samples were collected. The RNA was used for the RNA-seq analysis to search the role of gut microbiota in the development of small intestine. Groups: IMA100 mean gut microbiota from Alginate oligosaccharide 100mg/kg treated mice; IMA10 mean gut microbiota from Alginate oligosaccharide 10mg/kg treated mice; IMC mean gut microbiota from control group mice (dosed with water); Sa mean dosed with saline (no gut microbiota). "1" mean dosed for 1 week, "2" means dosed for 2 weeks.

Project description:Huatan Jiangzhuo Decoction, a traditional Chinese medicine composition, has been used to treat patients with hyperlipidemia (HLP). To investigate the therapeutic mechanism of Huatan Jiangzhuo decoction(HTJZD) and Poria cocos (PC) - Alismatis Rhizoma (AR) on gene expression in liver of rats with HLP, this study performed gene expression profiling analysis using data obtained from RNA-seq technology. It’s predicted that the potential role of regulating the expression of differentially expressed genes in the treatment of HLP by HTJZD and PC-AR separately.

IL-22 from enteroendocrine cells promotes early-life gut motility via microbiota in zebrafish

Project description:The gut microbiota, immune system, and enteric nervous system interact to regulate adult gut physiology. Yet the mechanisms establishing gut physiology during development remain unknown. We report that in developing zebrafish, enteroendocrine cells produced IL-22 in response to microbial signals before lymphocytes populate the gut. In larvae, IL-22 shaped the gut microbiota, increased Lactobacillaceae abundance and ghrelin expression to promote gut motility. Impaired motility and ghrelin expression were restored in il22-/- zebrafish by transfer of microbiota from wild-type zebrafish or by monoassociation with Lactobacillus plantarum. IL-22-deficient mice had impaired gut motility and reduced ghrelin expression in early life too, indicating a conserved function. Thus, before immune system maturation, enteroendocrine cells regulate early-life gut function by controlling the microbiota via IL-22.
