Project description:We report the first application of human umbilical cord mesenchymal stem cell (HUCMSC)-derived therapy for pulmonary arterial hypertension (PAH). A 3-year-old female presented with heritable PAH (ACVLR1 missense mutation) associated with hereditary hemorrhagic telangiectasia (HHT). Serial intra-pulmonary arterial and centrally intravenous infusions of conditioned media (CM) from allogenic HUCMSCs resulted in marked clinical and hemodynamic improvement after six months. A consecutive multiple cord single-cell RNA-seq integrated analysis (HUCMSC and HUVEC control) revealed eight common cell clusters, of which two (overrepresented by HUCMSCs) had regenerative potential.

Project description:Whole transcriptome gene expression profiling of Normal -(HUVEC) Human Umbilical Vein Endothelial Cells The HUVEC gene expression results analyzed in this study are further described in Kustermann S. et al. (2014) A real-time impedance based screening assay for drug induced vascular leakage. A NimbleGen Homo sapiens Expression Array [100718_HG18_opt_expr] study using total RNA recovered from HUVECs grown until 80% confluency. Each microarray measures the expression level of 23,611 genes using 45,033 probe sets with three 60-mer probes (PM) per probe set. Each probe set is represented once on the array.

Sex-specific transcriptional differences between non-treated male and female human umbilical vein endothelial cells (HUVEC)

Project description:Significant sex differences exist in cardiovascular diseases. It is largely unknown whether sexually dimorphic gene expression plays a role, and whether cells themselves show intrinsic sex differences. We therefore performed whole genome expression analyses in human umbilical vein endothelial cells (HUVEC) from five sample pools with equal amounts of four individual RNAs from either male or female donors (altogether 20 male and 20 female donors) and compared levels of gene transcription between the sexes. Genes indicative for greater immune responsiveness were stronger expressed in female compared to male HUVEC. There was a significant enrichment of 77 immune-related genes in female HUVEC.

Serial infusions of HUCMSC-CM resulted in marked clinical and hemodynamic improvement after 6 months, and showed no adverse events. Differential expression analysis between conditioned media and cells was used to identify molecular processes with a putative role in treatment benefit.
