Project description:The functional role of the maternal protein during early embryo development remains to be elucidated. Here we show that the maternal protein, Rbpms2, is highly expressed in mouse oocytes and that knockdown of Rbpms2 inhibits development from the morula to blastocyst stage. To study the underlying mechanism, we firstly analysed the differential expressed genes after Rbpms2 knockdown through single-cell sequencing. Combined the bioinformatics analysis of sequencing data, we then identify the impaired location of E-cadherin after knockdown of Rbpms2 may leading to the morula arrested development . Furthermore, Rbpms2 is important for development of mouse early embryos, most likely because it activates the BMP pathway. Taken together, our study uncovers that Rbpms2 activate the BMP pathway, and thus adjust the location of E-cadherin which is important for mouse early embryos development.

RBPMS and RBPMS2 Cooperate to Safeguard Cardiac Splicing P0

Project description:RBPMS and RBPMS2 share a high amino acid sequence similarity and are interchangeable in in vitro splicing assays. However, it remains unclear whether RBPMS and RBPMS2 play similar or distinct function during heart development in vivo. To address this question, we constructed Rbpms and Rbpms2 floxed mice with an established CRISPR-Cas9 method, and validated the efficacy of Rbpms or Rbpms2 floxed alleles in ablating RBPMS or RBPMS2 proteins by crossing them with germline-expressing Sox2-Cre. Transcriptome were abtained by RNA seq to investigate the cardiac function of RBPMS and RBPMS2. We performed RNA sequencing on RbpmscmKO, Rbpms2cmKO, and Rbpms/Rbpms2dcmKO hearts along with their littermate controls.

Rbpms2 promotes female fate upstream of the nutrient sensing Gator2 complex component, Mios

Project description:Reproductive success relies on proper establishment and maintenance of biological sex. In many animals, including mammals, the primary gonad is initially ovary in character. We previously showed the RNA binding protein (RNAbp), Rbpms2, is required for ovary fate in zebrafish. Here, we identified Rbpms2 targets in oocytes(Rbpms2-bound oocyte RNAs; rboRNAs). We identify Rbpms2 as a translational regulator ofrboRNAs, which include testis factors and ribosome biogenesis factors. Further, genetic analyses indicate that Rbpms2 promotes nucleolar amplification via the mTorc1 signaling pathway, specifically through the mTorc1-activating Gap activity towards Rags 2 (Gator2) component, Missing oocyte (Mios). Cumulatively, our findings indicate that early gonocytes are in a dual poised, bipotential state in which Rbpms2 acts as a binary fate-switch. Specifically, Rbpms2 represses testis factors and promotes oocyte factors to promote oocyte progression through an essential Gator2-mediated checkpoint, thereby integrating regulation of sexual differentiation factors and nutritional availability pathways in zebrafish oogenesis.

The impact of maternal age on trophectoderm gene expression profiles in human.

Project description:Ageing is a crucial factor that affects embryonic development and implantation. Maternal age has an impact on the blastocyst's gene expression levels from the fertilised oocyte and throughout the development of the preimplantation embryo. The current study reported differential gene expression analysis among trophectoderm samples that derived from young maternal age women (YMA - below 30 years), intermediate maternal age (IMA - 30-39 years old) and advanced maternal age women (AMA - at least 40 years old). The molecular approach was assayed by a low-input next generation RNA sequencing approach. Hundreds of significantly differentially expressed transcripts were reported. Extracellular exosome-related transcripts were significantly higher in the trophectoderm cells that derived from young women. According to our analysis, several important transcripts were reported. These molecular biomarkers may have a potential role in embryonic-endometrial communication and could be used in a diagnostic setting, that could help to improve the blastocyst implantation potential.
