Project description:Circulating sex steroids are critical for the development of neuroplasticity in the respiratory motor system. A primary locus of inducible respiratory neuroplasticity is the phrenic motor nucleus, a column of motor neurons in the ventral cervical spinal cord (C3-C5). In specific, exposure to acute intermittent hypoxia (AIH) induces strengthened synaptic connections to phrenic motor neurons and increased motor output; a form of neuroplasticity called phrenic long-term facilitation (pLTF). Gonadectomy (surgical removal of the testes in males and ovaries in females) reduces circulating sex steroids and eliminates AIH-induced phrenic pLTF in both females and males, implicating sex steroids as critical mediators for the development of neuroplasticity. To begin interrogating mechanisms of sex steroid influence on phrenic motor neurons and to determine the effect of gonadectomy on local gene expression, we performed RNA sequencing on ventral cervical spinal cord samples containing the phrenic motor nucleus. Our findings identified sex-specific gene expression in gonadally-intact rats and suggested that gonadectomy caused greater changes to the transcriptomes of male rats compared to females.

Single cell transcriptome of regional human embryonic stem cell-derived ventral cultures from the spinal cord

Project description:We generated single RNA-seq data to measure transcriptional profiles of six hESC-derived ventral populations, representing distinct regions of the developing human spinal cord. These cells are differentiated using a protocol that induces collinear activation of region-specific HOX genes during exposure to FGF8 and Wnt signaling (Lippmann et al, 2015 PMID:25843047). By transitioning to media containing retinoic acid, smoothened agonist (SAG), Purmorphamine (Pur), and DAPT after varying durations of Wnt signaling, ventral neuronal cultures are generated with unique rostrocaudal identities. In total we recovered the transcriptomes of 12,540 cells. Analysis verified expression of increasingly caudal HOX paralogs that could be correlated to cervical (HOX1-8; H24-vN, H48-vN, and H72-vN), thoracic (HOX1-9; H120-vN), lumbar (HOX1-11; H168-vN), and lumbosacral (HOX1-13; H216-vN) spinal regions. However, inconsistent differentiation between samples resulted in a mix of neural progenitor, neuron, mesodermal, and neural crest derivatives.

Gene expression data from cervical spinal cord regions of MPTP-intoxicated mouse by acupuncture

Project description:It has been shown that acupuncture at acupoints GB34 and LR3 inhibits the degeneration of nigrostriatal neurons in an 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) mouse model of Parkinsonâ??s disease. This preventative effect of peripheral acupuncture stimulation is hypothesized to be transmitted through the spinal cord to the nigrostriatal neurons. The gene expression profile changes following acupuncture at the acupoints were investigated in the cervical spinal cord of an MPTP-induced parkinsonism model using an Affymetrix genechip mouse gene 1.0 ST array. C57BL/6 mice were divided into four experimental groups; â? C: Control, â?¡ M: MPTP-treatment only, â?¢ MA: MPTP- and acupuncture-treatment at acupoints GB34 and LR3, â?£ MNA: MPTP- and acupuncture-treatment at non-acupoints. Total RNA was isolated from the cervical spinal cord regions of each experimental group (4 experimental group Ã? 2 samples of each experimental group = total 8 samples).

Project description:To comprehensively elucidate metabolite changes in different anatomical structures (e.g., gray matter and white matter) after spinal cord injury(SCI), our study utilized air-flow-assisted desorption electrospray ionization mass spectrometry imaging platforms to perform untargeted metabolomic studies. These analyzes are designed to identify metabolites critical in spinal cord injury. confirmed the profile differences in white and gray matter as well as in ventral and dorsal horns after SCI. These results provide valuable information for understanding in situ metabolite alterations after SCI.

The conversion of PrPC and the replication of prions, in cases of peripheral infection, occur following the uptake and subsequent spread of prions to brain via spinal cord. We investigated the changes of gene expression profiles in the spleen, cervical spinal cord and thalamus following intraperitoneal inoculation of ME7 scrapie prion and the inter-tissue alterations of the differentially expressed genes through Venn diagram to find genes which are commonly influenced by ME7 scrapie prion spread in the three tissues.

Project description:We profiled populations of mouse spinocerebellar tract neurons (SCTNs) in cervical and thoracic sections of mouse spinal cord, as defined by retrograde projection labeling. For each sample, SCTNs were labeled by injecting CTB (Alexa555 conjugated form, 1µg/µl in PBS) into the cerebellum using NanojetII at P4 and examined at P6-P7. Labeled SCTNs were collected manually from 150-300µm transverse spinal cords slices were generated using a razor blade. Cells were collected from each injection experiment and then profiled using standard Illumina bulk RNA-seq protocols
