Project description:Here we used RNAseq to compare the transcriptomes of the medio-basal hypothalamus of goats killed at two different times of the year (end of January and mid-May) and under different hormonal conditions (follicular and luteal phase, in January only). The current experiment was designed to identify the impact of photoperiod upon the transcriptome and to compare it with that of the ewe. The current design also tested the potential impact of progesterone (P4) upon this transcriptome; ewes were treated with a vaginal fluogestone acetate sponge during the breeding season (January) and killed either 2 days (follicular phase) or 8 days (luteal phase) after estrumate treatment and sponge removal.

Project description:Here we used RNAseq to assess the impact of progesterone (P4) on the transcriptome of the medio-basal hypothalamus of ewes. Ewes were treated with a vaginal fluogestone acetate sponge in mid-February. The sponge was removed in early March and ewes received a PMSG injection. Animals were then killed either 2 days (follicular phase) or 12 days (luteal phase) later.

Project description:Background: Small ungulates (sheep and goat) display a seasonal breeding, characterised by successive periods of sexual activity (SA) and sexual rest (SR). In sexual activity period the ovarian cycle of females is active and ready for reproduction (oestrus) whereas in sexual rest no ovulation and reproduction are possible (deep anoestrus). Odours emitted by a sexually active male can reactivate the ovarian cycle of anoestrus females. The plasticity of the olfactory system under these hormonal changes has never been explored at the peripheral level of odours reception. As it was shown in pig that the olfactory secretome (proteins secreted in the nasal mucus) could be modified under hormonal control, we monitored its composition in females of both species along several seasons, thanks to a non-invasive sampling of olfactory mucus. Results: In both species the olfactory secretome is composed of isoforms of OBP-like proteins, generated by post-translational modifications, phosphorylation, N-glycosylation and O-GlcNAcylation. Important changes were observed in the olfactory secretome between the sexual rest and the sexual activity periods, characterised in ewe by the specific expression of SAL-like proteins and the emergence of OBPs O-GlcNAcylation. In Goat, the differences between SA and SR did not come from new proteins expression, but from different post-translational modifications, the main difference between the SA and SR secretome being the number of isoforms of each protein. Conclusion: Despite common behaviour, seasonal breeding, and genetic resources, the two species seem to adapt their sensory equipment in SA by different modalities: the variation of olfactory secretome in ewe could correspond to a specialization to detect male odours only in SA, whereas in goat the stability of the olfactory secretome could indicate a constant capacity of odours detection suggesting that the hallmark of SA in goat could be the emission of specific odours by the sexually active male.

Project description:Background: The goat is an important farm animal. Reproduction is an important process of goat farming. The ovary is the most important reproductive organ for goats. In recent years, an increasing number of long non-coding RNAs (lncRNAs) have been implicated in the regulation of mammal reproduction. However, there are few studies on the function of lncRNAs in reproduction, particularly lncRNAs in the ovary. Results: The sequencing of goat ovaries generated 1,122,014,112 clean reads, and 4,926 lncRNAs and 1,454 TUCPs(transcripts of uncertain coding potential) were identified for further analysis by using the coding potential analysis software, CNCI, CPC and Pfam-sca. There were 115 /22 differential lncRNAs /TUCPs transcripts between the ovaries of the luteal phase and the follicular phase. We predicted the target genes of lncRNA /TUCP based on co-expression and co-localization methods. In total, 2584 /904 target genes were predicted by co-expression, and 326/73 target genes were predicted by co-localization. The functions of these target genes were further analyzed with GO and KEGG analysis. The results showed that lncRNAs /TUCPs, which are highly expressed in goat ovaries in the luteal phase, are mainly associated with the synthesis of progesterone, and we filtered the lncRNAs /TUCPs, such as XR_001918177.1 and TUCP_001362, which may regulate the synthesis of progesterone; lncRNAs /TUCPs, which are highly expressed in goat ovaries in the follicular phase, are mainly associated with oogenesis and the maturation of oocytes, and we filtered the lncRNAs /TUCPs that may regulate the oogenesis and maturation of oocyte, such as XR_001917388.1 and TUCP_000849. Conclusion: The present study provided the genome expression profile of lncRNAs /TUCPs in goat ovaries at different estrus periods and filtered the lncRNAs /TUCPs associated with goat reproduction. These results are helpful to further study the molecular mechanisms of goat reproduction.

Project description:Local breeds retained unique genetic variability important for adaptive potential especially in light of challenges related to climate change. Our objective was to perform, for the first time, a genome-wide diversity characterization using Illumina GoatSNP50 BeadChip of autochthonous Drežnica goat breed from Slovenia. Genetic diversity analyses revealed that the Slovenian Drežnica goat has a distinct genetic identity and is closely related to the neighboring Austrian and Italian alpine breeds. These results expand our knowledge on phylogeny of goat breeds from easternmost part of the European Alps.

Project description:Hibernation is a physiological and behavioural adaptation that permits survival during periods of reduced food availability and extreme environmental temperatures. This is achieved through cycles of metabolic depression which results in reduced body temperature (torpor) and rewarming (arousal). This cannot be achieved without extensive preparative physiological changes prior to the hibernation season. Dramatic changes in body temperature set-point observed over the hibernation season, pointing towards the hypothalamus, and several lines of evidence implicate a specialised glial cell type, the tanycyte, in hypothalamic control mechanisms linked to metabolic regulation. To investigate changes in tanycytic cell phenotype over the course of the hibernation season, we exposed golden hamsters (Mesocricetus auratus) to short photoperiod (SP) and reduced ambient temperature (8C) and then LASER capture microdissected the tanycytic region from animals at physiologically defined points across the season. RNAseq followed by combined hierarchical clustering and pathway analysis defined major changes in the tanycytic transcriptome linked to physiological status. Our analysis revealed a marked reduction in the expression of genes linked to ciliary assembly and GPCR-signalling during the hibernation season, as well as evidence for a shift towards increased glycolytic metabolism. These aspects all spontaneously reversed in refractory animals which have ceased to express torpor despite continued exposure to SP 8C. Tanycytes sampled mid-torpor show increased expression of immediate-early genes compared to the interbout euthermic state, while genes linked to RNA processing and translation show the reverse. The putative involvement of tanycytes in hibernation control are discussed.

Project description:Epigenetic regulation of gene transcription may play a critical role in the onset of puberty. Previous studies have confirmed that methylation regulates gene transcription in the hypothalamus-pituitary-gonadal axis during puberty initiation, but little is known about the regulation of DNA methylation on gene expression in the pineal gland. This study aims to screen pineal gland candidate genes related to the onset of goat puberty and regulated by genome methylation, to clarify the initiation mechanism of goat puberty and lay the foundation for improving goat reproductive performance. Pineal gland was collected from three female Anhui white goats during prepuberty (2.5 to 3.0 mo) and three during puberty (4.0 to 4.5 mo). Genome-wide hydrogen sulfite sequencing determined the DNA methylation profile, while RNA sequencing determined the transcriptome. The relationship between gene transcription and DNA methylation was determined by joint analysis. We found that there was no significant change in the whole genome methylation level of goat pineal gland before and after puberty, but the methylation pattern changed significantly, indicating that genomic DNA methylation of the pineal gland may be involved in regulating goat puberty initiation. Changes in DNA methylation patterns affected some pineal gland transcriptomes, while the transcriptional level of most genes was not affected by DNA methylation differences. Genes regulated by DNA methylation were mainly involved in metabolic processes, oxidative phosphorylation (OXPHOS), and signaling pathways related to thermogenesis. The differential genes ATP5F1D, CACNB2, and PTEN were significantly regulated by methylation, and the difference multiples of LIN28B, GIP, OPN1SW, and DCC were the most significant, possibly involved in puberty initiation.
