Project description:Aberrant mechanotransduction and compromised epithelial barrier function are associated with numerous human pathologies including inflammatory skin disorders. However, the cytoskeletal mechanisms regulating inflammatory responses in the epidermis are not well understood. Here we addressed this question by inducing a psoriatic phenotype in human keratinocytes and reconstructed human epidermis using a cytokine stimulation model. We show that the inflammation upregulates the Rho-myosin II pathway and destabilizes adherens junctions (AJs) promoting YAP nuclear entry. The integrity of cell-cell adhesion but not the myosin II contractility per se is the determinative factor for the YAP regulation in epidermal keratinocytes. The inflammation-induced disruption of AJs, increased paracellular permeability, and YAP nuclear translocation are regulated by ROCK2, independently from myosin II activation. Using a specific inhibitor KD025, we show that ROCK2 executes its effects via cytoskeletal and transcription-dependent mechanisms to shape the inflammatory response in the epidermis.

Inflammation drives TGFβ1 activation via the αvβ6 integrin-mechanotransduction pathway in human skin

Project description:Transforming growth factor β (TGFβ) is a critical regulator of skin homeostasis and is implicated in skin inflammation, including psoriasis. We investigated the molecular mechanisms of TGFβ1 regulation during cutaneous inflammation using reconstructed human epidermis. We report a mechanosensitive positive feedback loop, that occurs downstream of inflammatory stimuli and involves TGFβ-mediated activation of mechanotransduction pathways, further promoting αvβ6 integrin-dependent TGFβ1 activation in keratinocytes.

Hic-5 drives epithelial mechanotransduction promoting a feed-forward cycle of bronchoconstriction.

Project description:Mechanical forces are essential for organ function, but excessive or dysregulated forces can promote pathologic conditions. In asthma, bronchoconstriction narrows the airway, compressing the airway epithelium and activating mechanotransduction, yet key regulators of mechanotransduction remain unclear. Here we show that Hic-5, a focal adhesion adaptor, is a key regulator of epithelial mechanotransduction. In human airway epithelial cells at air–liquid interface exposed to mechanical compression that mimics bronchoconstriction, we find that compression induces Hic-5 expression in airway basal cells. We further validated these in vitro findings by reanalyzing single-cell RNA-seq data from patients with asthma undergoing bronchoconstriction after allergen challenge, which revealed increased Hic-5 expression in airway basal cells. Hic-5 knockdown in human airway epithelial cells markedly attenuates mechanoresponses to compression, including stress fiber formation, differential gene expression, and increased secretion of endothelin-1 (ET-1). Through secretion of ET-1, a potent bronchoconstrictor, Hic-5 drives epithelial mechanotransduction and promotes a feed-forward cycle of bronchoconstriction, thereby highlighting dysregulated mechanical forces as active drivers of human disease.

Project description:Splenic follicular B cells (FoB; B220+GL7-CD38hiCD23+) and Germinal Center B cells (GCB; B220+GL7+CD38lo) were FACS sorted from WT or CD23-Cre.Rock2 mice after 7d post-immunization with NP-CGG. RNA-seq was then employed to compare the transcriptome of GCBs from the indicated mice. Here we show that loss of ROCK2 leads to altered gene expression in GCBs. We also employed ATAC-seq analysis and found that differentially expressed genes from the ROCK2-deficient GC B cells contained loci that were enriched for IRF binding sites.

Talin force coupling underlies eukaryotic cell-substrate adhesion

Project description:Integrin-mediated cell adhesion and mechanotransduction are considered key innovations in animal evolution, enabling a specific and physically robust attachment of cells to the extracellular environment. Here, we show that this type of cell adhesion is a specification of an evolutionary conserved force coupling mechanism that originated in unicellular organisms and is mediated by the actin-binding protein talin. Unlike heterodimeric integrin receptors, talin is widely distributed in unicellular organisms, including amoebae. By comparing piconewton-scale mechanics of talin-A from amoeboid cells with that of mammalian talin-1, we uncover a conserved role of talin as a force transducer, transmitting mechanical forces of 6–8 pN into cells, even in unicellular organisms lacking integrin receptors. Our data show that the critical evolutionary steps towards integrin-mediated cell adhesion were the specification of talin as an adaptor protein allowing activation of integrin receptors, regulation of biochemical signalling via paxillin, FAK and YAP, and control of cell adhesion turnover by KANK recruitment. Collectively, our experiments identify a pivotal yet hitherto underappreciated role for talin in the evolution of eukaryotic cell-substrate adhesion and mechanotransduction.
