Immunological Sexual Dimorphism in Placenta and Its Response to Benzene Exposure

Project description:There is increasing concern regarding the adverse effects of air pollution on human health, and benzene is a major toxic compound in air pollution. Maternal benzene exposure has been associated with reproductive complications, such as preterm birth, low birth weight, and immunological and neurological complications in the offspring. However, it is poorly understood how benzene induces these complications. Our objective was to establish a full body inhalation mouse model for maternal benzene exposure that mimics clinical phenotypes observed in human populations, and characterize the maternal immune activation and placental response in our model. Here, we report that maternal immune activation triggered by benzene exposure during pregnancy leads to increased resorptions, abnormal placenta development and low birth weight of fetus. More importantly, there is a sexual dimorphic response to benzene exposure in female and male placentas. In the male placenta, the transcriptome changes reveal a more immunologically relevant profile, while females have a metabolically related profile. Furthermore, we discover the sexual dimorphic response could be a consequence of the sexual dimorphism of placenta at baseline, which indicates the significant difference between sexes in terms of the immunological processes in the placenta, both in human and mouse. Therefore, our findings established a benzene exposure mouse model and indicated the sexual dimorphism of placenta, which provides valuable reference for the future pregnancy studies.

Project description:The molecular and functional bases of sexual dimorphism in skeletal muscle remain poorly understood. The androgen receptor (AR) is a major regulator of sex-biased gene expression in muscle, but its genomic targets and associated coregulators in vivo are incompletely defined. Using ChIL-seq and an AirID-AR knock-in mouse, we mapped AR-bound genes and AR-associated proteins in skeletal muscle and identified histone deacetylase–linked corepressors. We further identified myosin binding protein H (Mybph) as a female-biased AR-repressed gene conserved in mouse and human muscle. Mybph loss disrupted sarcomeric organization and selectively delayed postinjury force recovery in female mice. These findings define an in vivo AR regulatory network and identify AR-dependent Mybph repression as a potential mechanism contributing to skeletal muscle sexual dimorphism.

Dissecting Sexual Dimorphism in Aortic Valve Stenosis by Proteomics

Project description:Managing aortic valve stenosis (AVS) is challenging because of the lack of pharmacotherapies to halt/revert stenosis, deeming aortic valve replacement (AVR) the only effective treatment. AVS management is further confronted with staggering sexual dimorphism; women display more extensive fibrosis, while men present remarkably higher valve calcification. To accelerate the development of effective and sex-personalised pharmacotherapies, deeper molecular insights are needed. Hence, we aimed to characterise AVS sexual dimorphism using proteomics. Aortic valves were obtained from surgical AVR. Disease severity was assessed by echocardiography. Fifty valves (50% women) were homogenised, and the proteins were quantified by LC-MS/MS. The relevance of differentially expressed proteins (DEPs) in sexual dimorphism was appraised using protein-protein interaction (PPI) and functional enrichment analyses (FEA). DEPs were validated using immunohistochemistry, qRT-PCR and ELISA, with the aid of 30 additional independent valves.
