Human Placental Exosomes in Gestational Diabetes Mellitus Carry a Specific Set of miRNAs Associated with Skeletal Muscle Insulin Sensitivity

Project description:The objective of this study was to investigate whether placental exosomes in gestational diabetes mellitus (GDM) carries a specific set of miRNAs associated with skeletal muscle insulin sensitivity. Exosomes were isolated from chorionic villi-conditioned media and plasma from normal and GDM pregnancies. A specific set of miRNAs was identified to be selectively enriched within exosomes when compared to their cells of origin indicating specific packaging of miRNAs into exosomes. In addition, miRNA expression varies in a consistent pattern in placenta, placental-derived exosomes, circulating exosomes and skeletal muscle in GDM.

Systematic characterization of human placenta-derived miRNAs and identification of autophagy-related miRNAs in gestational diabetes mellitus [miRNA-seq]

Project description:The objective of this study was to identify and investigate the roles of miRNAs in GDM. In the current study, placental miRNA expression profiles of normal controls and GDM patients were analyzed using high-throughput sequencing. Bioinformatics analysis identified that 4 were upregulated and 6 were downregulated in 10 most differential miRNA in GDM placentas compared with NC placentas. GO and KEGG enrichment analyses demonstrated that metabolic process-associated terms and metabolic pathways that may be related to GDM were significantly enriched. Our study indicates that aberrant expression of miRNAs may play roles in autophagy in GDM placentas, providing new insights into GDM.

Reduced representation bisulfite sequencing (RRBS) methylation profiles of placenta and umbilical cord blood with or without gestational diabetes mellitus (GDM)

Project description:Objective: To explore the characteristics and underlying molecular mechanisms of genome-scale expression profiles of women with- or without- gestational diabetes mellitus and their offspring. Materials and Methods: We recruited a group of 21 pregnant women with gestational diabetes mellitus (GDM) and 20 healthy pregnant women as controls. For each pregnant women, RRBS were performed using the placenta and paired neonatal umbilical cord blood specimens. Differentially methylated regions (DMRs) were identified. Then, functional enrichment analysis was performed to differential methylated genes (DMGs) separately or interactively in placenta and umbilical cord blood. Results: Through the comparison of GDM and healthy samples, 2779 and 141 DMRs were identified from placenta and umbilical cord blood, respectively. Functional enrichment analysis showed that the placenta methylation and expression profiles of GDM women mirrored the molecular characteristics of “type II diabetes” and “insulin resistance”. Methylation-altered genes in umbilical cord blood were associated with pathways “type II diabetes” and “cholesterol metabolism”. DMGs illustrated significant overlaps among placenta and umbilical cord blood samples, and the overlapping DMGs were associated with cholesterol metabolism. Conclusions: Our research demonstrated the epigenomic alternations of GDM mothers and offspring. Our findings emphasized the importance of epigenetic modifications in the communication between pregnant women with GDM and offspring, and provided reference for the prevention, control, treatment, and intervention of perinatal deleterious events of GDM and neonatal complications.

Project description:Affymetrix miRNA arrays were used to generate miRNA profiles in placenta of Meishan and Yorkshire pigs on stages of initiation (gestational day 26) and establishment (gestational day 50) of placental folds development. The study allowed for the determination of the miRNAs that were differentially expressed in porcine placenta on different gestational day. This study will provide the information to better understanding of the role miRNAs in porcine placental development.

Integrated Proteomic Characterization of Maternal Blood, Umbilical Cord Blood and Placenta Tissue of Gestational Diabetes Mellitus Patients

Project description:In this retrospective study, a diverse set of biological specimens was assembled, consisting of maternal blood, umbilical cord blood, and placenta tissue, from a cohort of 22 mothers with gestational diabetes mellitus and a matched group of 19 healthy mothers. Integrated proteomic characterization of these samples were performed, and functional enrichments based on GO and KEGG database, as well as Gene Set Enrichment Analysis were used to elucidate the pathways involved in the pathophysiology of gestational diabetes mellitus. Moreover, a weighted protein co-expression network was constructed to analysis the correlation of expression modules with clinical traits.

Our research helps to understand placental overgrowth in GDMM and its impact on fetal development, providing new insights into the pathogenesis of GDMM and predicting potential therapeutic interventions.
