Project description:Downy mildew, caused by the biotrophic oomycete Plasmopara viticola, is one of the most economically significant grapevine diseases worldwide. Current strategies to cope with this threat rely on the massive use of phytochemical compounds during each cultivation season. The economic costs and negative environmental impact associated with these applications increased the urge to search for alternative strategies for sustainable disease control. Improved knowledge on plant mechanisms able to counteract pathogen infection may allow the development of alternative strategies of plant protection. Epigenetic regulation, in particular DNA methylation, is emerging as a key factor in the context of plant-pathogen interactions, associated with the expression modulation of defense genes. To improve our understanding of the genetic and epigenetic mechanisms underpinning grapevine response to P. viticola, we studied the modulation of both 5-mC methylation and gene expression at 6 and 24 hours post-infection (hpi). Leaves of two table grape genotypes (Vitis vinifera), selected by breeding activities for their contrasting level of susceptibility to pathogen, were analysed. Following pathogen infection, we found variations in the 5-mC methylation level and in the gene expression profile. The results indicate a genotypic-specific response to pathogen infection. The tolerant genotype (N23/018) at 6 hpi exhibits a lower methylation level compared to the susceptible one (N20/20), and its shows an early modulation (at 6 hpi) of defense and epigenetic-related genes during P. viticola infection. These data suggest that the timing of response is an important mechanism to efficiently counteract the pathogen attack.

Project description:Hymenoscyphus fraxineus, the causal agent of Ash Dieback (ADB), has been introduced to eastern Europe in the 1990s from where it spread causing decline in European ash populations. However, the genetic basis of the molecular response in tolerant and susceptible ash trees to this disease is still largely unknown. We performed RNA-sequencing to study the transcriptomic response to the disease in four ash genotypes (ADB-tolerant FAR3 and FS36, and ADB-susceptible UW1 and UW2), during a time-course of 7, 14, 21, and 28 days post-inoculation, including mock-infected trees as control samples for each sampling time point. The analysis yielded 395 and 500 Differentially Expressed Genes (DEGs) along the response for ADB-tolerant FAR3 and FS36, respectively, while ADB-susceptible UW1 and UW2 revealed 194 and 571 DEGs, respectively, with most DEGs found exclusively in just one of the genotypes. DEGs shared between tolerant genotypes FAR3 and FS36, included genes involved in the production of phytoalexins and other secondary metabolites with roles in plant defense. Moreover, we identified an earlier expression of genes involved in both pattern- and effector-triggered immunity (PTI and ETI) in ADB-tolerant genotypes, while in ADB-susceptible genotypes both responses were delayed (late response). Overall, these results revealed different transcriptomic expression patterns not only between ADB-tolerant and ADB-susceptible genotypes, but also within these two groups. This hints to individual response in the natural tolerance to ADB, possibly revealing diversified strategies across ash genotypes.

Project description:European common ash, Fraxinus excelsior L., is currently threatened by Ash dieback (ADB) caused by the fungus, Hymenoscyphus fraxineus. To detect and identify metabolites that may be products of pathways important in contributing to resistance against H. fraxineus we performed untargeted metabolomic profiling on leaves from selected F. excelsior individuals showing strong tolerance or susceptibility to H. fraxineus. We identified sets of "features" [small molecules] that enabled strong discrimination between tolerant or susceptible genotypes of F. excelsior. Strikingly, tolerant F. excelsior lines exhibited low levels of iridoid glycosides, known anti-feeding deterrents. As Europe is threatened by Emerald Ash Borer (Agrilus planipennis), an invasive wood boring beetle native to East Asia that has devastated North American ash, our study raises the question whether selection for resistance to H. fraxineus leads to ecological trade-offs that result in susceptibility to emerging pests such as emerald ash borer.

Project description:In present study we compared transcriptional response to salinity stress between susceptible CO 12 and tolerant genotype trichy 1 of finger millet. We found out that several functional group of genes like transporters, transcription factors, genes involved in cell signalling, osmotic homeostasis, compatible solutes biosynthesis were upregulated more in tolerant genotype as compared to susceptible genotype in response to salinity stress. Salnity inhibited photosynthetic capacity and photosynthesis related genes more in susceptible genotype as compared to tolerant genotype. Identified genes are excellent targets for further functional studies in order to understand more specific molecular mechanisms of salt tolerance in two genotypes of finger millet and can be further used for developing salinity tolerant crops through genetic engineering.

Project description:In many potato cultivation regions, production is constrained by abiotic stresses such as drought and high temperatures which are often present in combination. We aimed to identify key mechanisms and processes underlying single and combined abiotic stress tolerance by a comparative analysis of tolerant and susceptible cultivars. Physiological data supported cultivars Desiree and Unica as being abiotic stress tolerant, while Agria and Russett Burbank were stress susceptible. This was indicated by the stronger impact of abiotic stress on photosynthetic carbon assimilation in the susceptible cultivars. Similarly, susceptible cultivars exhibited a lower leaf transpiration rate following stress, particularly combined heat and drought stress. Transcript profiles using microarrays were highly divergent both between genotypes and following the application of stress treatments. However, relatively few transcripts or metabolites exhibited genotype specific responses to abiotic stress treatment. Furthermore, apart from a decrease in the abundance of transcripts associated with PSII, particularly the light harvesting complex in both Desiree and Unica, there were very few changes that were consistent across stress susceptible or stress tolerant genotypes following stress treatment.

Natural sources of resistance are available in several wild Vitis species, which are being used in traditional breeding approaches. However, molecular switches, signals and effectors involved in resistance are poorly understood. In this paper we report a microarray analysis of early transcriptional changes associated to P. viticola infection in both susceptible Vitis vinifera and resistant Vitis riparia plants (12 and 24 h post inoculation). To provide a biological basis to the choice of time points for transcriptome analyses, we performed microscopic examinations of infected tissues at 12, 24, 48 and 96 hpi. Data suggest that resistance in V. riparia is mainly a post-infectional event and involves a large reprogramming of host metabolism. Transcripts of signal transduction-related genes are specifically and often strongly accumulated in response to infection. Well known defence genes also show marked transcript increases, especially pathogenesis-related proteins PR-10 and stylbene synthases, and genes related to an hypersensitive reaction. On the other hand, V. vinifera mounts a much weaker transcriptional response, involving mainly defence genes, not effective enough in preventing pathogen infection. Leaves from one resistant (V. riparia cv. Gloire de Montpellier) and one susceptible (V.vinifera cv. Pinot Noir) grapevine cultivars grown in vitro were infected with the oomycete Plasmopara viticola, and transcriptome changes were investigated at 12h and 24h after infection. Project description:Grapevine downy mildew is an important disease affecting crop production and causing severe losses. To identify genotype-dependent responses towards this pathogen and to explore the molecular mechanisms involved in grapevine-P. viticola resistance, we have conducted a proteomic analysis of leaf samples from resistant and susceptible grapevine genotypes prior and post-inoculation with the pathogen. Proteins were analyzed by quantitative two-dimensional differential gel electrophoresis (2D-DIGE). The analysis able to identified 50 unique proteins. Functional analysis showed that photosynthesis and metabolism were the main categories differentiating genotypes at 0h and that P. viticola-responsive proteins were mainly involved in photosynthesis, carbohydrate metabolism, stress and defense responses and redox homeostasis. ROS production, total antioxidant capacity and lipid peroxidation on both genotypes were determined and together with the proteome data suggest that Regent presents a strict balance between ROS control and signaling leading to plant cell death activation. Our data reveals the genotype-dependent modulation of plant metabolism and defense responses providing new insights into underlying molecular processes of grapevine resistance against the downy mildew fungus.
