Project description:Purpose: Macrophages are often classified into M1 ‘classical’ and M2 ‘alternatively-activated’ macrophages. Extracellular vesicles (EVs) are biomolecule carriers involved in cell-cell communication. Here, we provide a first insight into the complete small RNA cargo of human macrophage M1/M2 EVs. Methods: Monocyte-derived macrophages were polarised into M1 (GM-CSF+LPS+IFNγ) or M2 (M-CSF+IL-4+IL-13) and EVs isolated by size exclusion chromatography. EVs were characterised by nanoparticle tracking analysis, electron microscopy and ELISA. EV RNA samples were prepared for small RNA sequencing using Qiagen’s GIAseq small RNA Library Prep kit and sequenced on an Illumina NextSeq500, single end 75 bp. Functional enrichment analysis was performed using MIENTURNET, based on validated miR-target interactions from miRTarBase. Results: Many types of small non-coding RNAs were found in EVs from M1/M2 macrophages including miRNAs, isomiRs, tRNA fragments, piRNA, snRNA, snoRNA and yRNA fragments. Distinct differences were observed between M1 and M2 EVs, with higher relative abundance of miRNAs, and lower abundance of tRNA fragments in M1 EVs compared to M2 EVs. MicroRNA-target enrichment analysis identified several gene targets involved in gene expression and metabolic processes. Conclusions: M1 and M2 cells release EVs with distinct tRNA and miRNA cargo, which have the potential to contribute to the unique effect of these cell subsets on their microenvironment.

Project description:Melanoma progression and metastasis depend on intercellular communication within the tumor microenvironment (TME), where extracellular vesicles (EVs) have emerged as essential mediators through the transfer of molecular cargo. In melanoma, macrophages are enriched in the TME, and their abundance and infiltration into the tumor area are associated with poor prognosis. In this study, we examined the effects of pro-inflammatory M1 and immunosuppressive M2 macrophage-derived extracellular vesicles (EVs) on melanoma cells to gain insights into EV-mediated interactions between macrophages and tumor cells. Our results demonstrate that M1 EVs reprogram MV3 melanoma cells into a pro-inflammatory state by upregulating the expression and secretion of pro-inflammatory cytokines CXCL8, IL-6 and IL-1B. This M1 EV -induced inflammatory phenotype enhanced melanoma cell invasion, which was suppressed by CXCL8 silencing. Furthermore, M1 EV treatment activated the NF-κB signaling pathway, and its inhibition with the IKK16 inhibitor led to downregulation of inflammation-associated genes. These results suggest that M1 EVs promote tumor-associated inflammation in melanoma through NF-κB signaling, underscoring the pivotal role of EV-mediated macrophage–tumor cell communication in cancer progression.

Project description:Matrix-bound extracellular vesicles were isolated from the left ventricle tissue of de-identified patients with non-failing (healthy) hearts or with non-ischemi heart failure. mIRNA sequencing was performed on the miRNA cargo sequestered within the extracellular vesicles. Differences in the miRNA signature of healthy versus failing-tissue derived extracellular vesicles suggests that disease progression to heart failure is associated with dynamic changes in vesicular cargo.

Project description:Mesenchymal stem cells (MSCs) can respond to micrenvironmental cues to exert immune suppression or stimulation. Both MSCs and macrophages (MΦs) are part of the bone marrow microenvironment. Separately, both cell types can influence the behavior of breast cancer cells towards dormancy and reverse dormancy. This study assessed the MΦ−ΜSC axis, specifically determined how activated MSCs could influence the type of MΦ. The toll like receptor 4 (TLR4) on MSCs was activated with LPS and then used to assess how this influence the type of MΦ. We demonstrated that activated MSCs can convert M2 MΦs to M1 type and vice versa. Interestingly, the transition is not stable showing re-transition to the original type. This suggested the ability of activated and inactivated MSCs to maintain homeostasis by dictating the MΦ type. We quantitated cytokine levels in the media from activated and inactivated MSCs without evidence of significant changes. Using siRNA and pharmacological inhibitors, the results supported a role for MSC derived extracellular vesicles (EVs) as mediators of MΦ transition. We assessed our published miRNA arrays from cancer activated MSCs and identified one of four potential miRNAs that has been reported to regulate myeloid cells. We further investigated molecular changes with RNA-Seq using extracts from activated and inactivated MSCs, and EVs from M1 and M2 MΦs

Project description:Augmented extracellular matrix (ECM) stiffness is a mechanical hallmark of cancer. Mechanotransduction studies have extensively probed the mechanisms by which ECM stiffness regulates intracellular communication. However, the influence of stiffness on intercellular communication aiding tumor progression in three-dimensional microenvironments remains unknown. Small extracellular vesicles (EVs) are communicators of altered biophysical cues to distant sites through EV-ECM interactions and EV-mediated recipient cell-ECM interactions. Here we demonstrate stiffness-mediated modulation of small EVs secretion and cargo from three-dimensional oral squamous cell carcinoma spheroids. Using a spheroid culture platform with varying matrix stiffness properties, we show that small EVs carry parental biomolecular cargo, including mechanosensitive Piezo1 ion channel and adhesion molecule CD44. We comprehensively validate the presence of both markers in our EV populations using proteomic and genetic analysis. Transcriptomic analysis of microRNA and long non-coding RNA cargo of small EVs released from soft and stiff ECM spheroids revealed enrichment of tumorigenic and metastatic profiles in EVs from stiff ECM cultures compared to that of soft ones. Gene set enrichment analysis of a comparative dataset obtained by overlaying spheroid mRNA and EV miRNA profiles identified key oncogenic pathways involved in cell-EV crosstalk in the spheroid model.

Project description:Macrophages are important antigen presenting cells which can release extracellular vesicles (EVs) carrying functional cargo including non-coding RNAs. Macrophages can be broadly classified into M1 'classical' and M2 'alternatively-activated' macrophages. M1 macrophages have been linked with inflammation-associated pathologies, whereas a switch towards an M2 phenotype indicates resolution of inflammation and tissue regeneration. Here, we provide the first comprehensive analysis of the small RNA cargo of EVs from human M1 and M2 primary macrophages. Using small RNA sequencing, we identified several types of small non-coding RNAs in M1 and M2 macrophage EVs including miRNAs, isomiRs, tRNA fragments, piRNA, snRNA, snoRNA and Y-RNA fragments. Distinct differences were observed between M1 and M2 EVs, with higher relative abundance of miRNAs, and lower abundance of tRNA fragments in M1 compared to M2 EVs. MicroRNA-target enrichment analysis identified several gene targets involved in gene expression and inflammatory signalling pathways. EVs were also enriched in tRNA fragments, primarily originating from the 5' end or the internal region of the full length tRNAs, many of which were differentially abundant in M1 and M2 EVs. Similarly, several other small non-coding RNAs, namely snRNAs, snoRNAs and Y-RNA fragments, were differentially enriched in M1 and M2 EVs; we discuss their putative roles in macrophage EVs. In conclusion, we show that M1 and M2 macrophages release EVs with distinct RNA cargo, which has the potential to contribute to the unique effect of these cell subsets on their microenvironment.

Project description:We tested the hypothesis that the biosensor capability of the endometrium is mediated in part, by the effect of different cargo contained in the extracellular vesicles secreted by the conceptus during the peri-implantation period of pregnancy. We transferredBos taurus taurusembryos of different origin:In vivo(high developmental potential (IV)),in vitro(intermediate developmental potential (IVF)), or cloned (low developmental potential (NT)), intoBos taurus indicusrecipients. Extracellular vesicles (EVs) recovered from Day 16 conceptus conditioned medium were characterized and their microRNA (miRNA) cargo sequenced alongside RNA sequencing of their respective endometria. There were substantial differences in the endometrial response toin vivoVsin vitroandin vivoVs cloned conceptuses (1153 and 334DEGs respectively) with limited differences betweenin vitroVs cloned conceptuses (36 DEGs). miRNA cargo was similar between all three groups (426 common cargo) differences betweenin vivoand cloned (8 miRNAs), andin vivoandin vitro(6 miRNAs) observed. Treatment of endometrial epithelial cells with mimic or inhibitors for miR-128 and miR-1298 changes to the proteomic content of target cells (96, and 85 respectively) of which mRNAs are altered in the endometriumin vivo(PLXDC2, COPG1, HSPA12A, MCM5, TBL1XR1, and TTF). In conclusion, we have determined that the biosensor capability of the endometrium is mediated in part, by its response to different EVs miRNA cargo produced by the conceptus during the peri-implantation period of pregnancy.
