Project description:Articular cartilage has a limited capacity to self-heal once damaged. Tissue-specific stem cells are a solution for cartilage regeneration; however, ex vivo expansion resulting in cell senescence remains a challenge as a large quantity of high-quality tissue-specific stem cells are needed for cartilage regeneration. Our previous report demonstrated that decellularized extracellular matrix (dECM) deposited by human synovium-derived stem cells (SDSCs), adipose-derived stem cells (ADSCs), urine-derived stem cells (UDSCs), or dermal fibroblasts (DFs) provided a solution to rejuvenate human SDSCs in proliferation and chondrogenic potential. This study focused on the evaluation of ex vivo rejuvenation of rabbit infrapatellar fat pad-derived stem cells (IPFSCs) by the abovementioned dECMs to be used in functional cartilage repair in a rabbit osteochondral defect model and the potential cellular and molecular mechanisms underlying this rejuvenation. We found that dECM rejuvenation promoted rabbit IPFSCs’ cartilage engineering and functional regeneration in both in vitro and in vivo models, particularly for the dECM deposited by UDSCs, which was further confirmed by proteomics data. RNASeq analysis indicated that both mesenchymal-epithelial transition (MET) and inflammation-mediated macrophage activation and polarization are potentially involved in the dECM-mediated promotion of IPFSCs’ chondrogenic capacity, which needs further investigation.

Urine stem cells as a bona fide model to study cartilage associated disease

Project description:Non-invasive cellular models for genetic growth disorders affecting cartilage and bone remain rare, hampering mechanistic studies and the development of personalized therapies. Here, we present urine-derived stem cells (USCs), as a robust in vitro system for human chondrogenesis. Single-cell RNA sequencing of undifferentiated USCs revealed distinct clusters, of which a subset had high stem cell character. After differentiation, USCs underwent osteogenic and chondrogenic lineage with positive marker expression, confirmed by bulk-RNA-Seq. Single cell transcriptomic comparisons with healthy cartilage revealed a healthy cartilage mimicking, chondrocyte-like subcluster. Pseudotime analysis revealed multiple chondrogenic trajectories.

Scaffold-free cryopreservable cartilage grafts obtained from hiPSC-derived chondroprogenitor cells for airway reconstruction with growth adaptability

Project description:Pediatric tracheal reconstruction remains a major clinical challenge because of limited graft availability and the need for growth-adaptive materials. Current approaches, such as costal cartilage grafting and use of scaffold-based constructs, often suffer from complications including graft resorption, donor site morbidity, and poor integration. Here, we present scaffold-free cartilage grafts (chondro-plates) derived from expandable limb-bud mesenchymal cells generated from human leukocyte antigen-homozygous human induced pluripotent stem cells. These grafts are cryopreservable and maintain their hyaline cartilage phenotype after a brief pre-culture. In both rat and rabbit tracheal defect models, chondro-plates supported robust cartilage regeneration, epithelial reconstitution, and neovascularization. Importantly, in a pediatric-like growing rat model, chondro-plates preserved luminal patency and structural integrity, outperforming autologous costal cartilage. This study demonstrates a clinically viable, off-the-shelf strategy for tracheal reconstruction using scalable, immunocompatible, and growth-adaptive cartilage grafts.

Tissue Engineered Elastic Cartilage-Mimetic Auricular Grafts for Microtia Reconstruction

Project description:Microtia, the congenital malformation of the external ear, imposes a significant psychosocial burden on affected children1-4. The current gold standard treatment, autologous costal cartilage reconstruction, requires multiple complex surgeries, including the harvest of rib cartilage5-7. Tissue engineering offers a promising alternative, enabling the generation of patient-specific auricular grafts from a small biopsy (~5 mg). However, despite multiple decades of research, clinically viable, tissue engineered auricular grafts remain unavailable8. Key challenges include fibrocartilage formation in vitro, heterogeneous extracellular matrix development, and mechanical inferiority, all resulting in graft deformation and degradation in vivo9-15. Here, we report the successful fabrication of bioprinted auricular grafts that closely resemble native human auricular cartilage. Our grafts demonstrated a uniform distribution of elastin, glycosaminoglycans, and collagen II, lacked collagen I, and achieved a compressive modulus of 803 ± 134 kPa after 9 weeks of maturation in vitro (human auricular cartilage: 1023 ± 101 kPa). In vivo, grafts maintained their structural integrity for 6 weeks in a subcutaneous rat model and showed a transition towards mature elastic fiber. These grafts represent the closest approximation to native elastic cartilage reported to date and lay the foundation for a clinically viable, long-term treatment for children affected by microtia.
