Project description:The decision of whether to grow or to divert energy to stress response is a key physiological trade-off for plants seeking to survive under fluctuating environments. However, the cell-to-cell communication underlying this decision is largely unknown. Here we show that three orphan leucine-rich repeat receptor kinases (LRR-RKs) act as direct ligand-perceiving receptors for PSY-family peptides and play a key role in local switching between two opposing pathways. In contrast to the known LRR-RKs which activate signaling upon ligand binding, PSY receptor (PSYR) signaling, which intrinsically induces a stress response, usually is repressed by ligands to permit growth. Conversely, PSYRs activate the expression of various genes encoding stress response transcription factors upon depletion of the ligands. Loss of PSYRs resulted in defects in the plant’s tolerance to both biotic and abiotic stresses. This ligand-deprivation-dependent activation system enables plants to exert exquisitely tuned regulation of stress responses in the tissues proximal to metabolically dysfunctional damaged sites where ligand production is impaired.

Project description:Seed defence priming is emerging as a novel, cost-efficient and environmentally safe tool for pest management. Previous reports suggest that this strategy could potentially uncouple the defence-growth trade-off in plants, but this role and the mechanisms underlying it remain elusive. Here, we investigated the ability of seed priming with four phytohormones and two plant vitamins to uncouple the growth-defence trade-off in Arabidopsis thaliana upon pest infestation, focusing on its molecular dynamics. To broaden our scope, two phytophagous species from different feeding guilds were used: the generalist cell-content sucker Tetranychus urticae, and the specialist chewer Pieris brassicae. A combined -omics approach revealed distinct and interactive effects of seed priming and herbivory. Specifically, seed priming with 0.1 mM methyl jasmonate induced a state of readiness characterized by reduced stomatal aperture, high transcriptomic variation, increased sugar content and enhanced translational rate. Upon pest infestation, primed plants exhibited herbivore-specific responses. Interaction of seed priming and T. urticae infestation increased amino acid levels, hydroxycinnamic acid amide production, and reallocated resources from arabidopsides and anthocyanin biosynthetic pathways. In contrast, P. brassicae infestation in primed plants led to enhanced anthocyanin biosynthesis, alterations in lignin metabolic intermediates, and sustained carbon and nitrogen sources. These findings demonstrate that seed priming can enhance pest-specific defences in plants without incurring a fitness penalty, effectively managing infestations while maintaining growth.

Project description:The evolutionary transition of multicellular life initially involves growth in groups of undifferentiated cells followed by differentiation into soma and germ-like cells. This is facilitated by trade-offs between traits determining survival and reproduction, favoring the coexistence of cells with extreme trait values and a convex trade-off curve as the multicellular state dominates. However, these transitions remain poorly characterized at the ecological and genetic level. Here, we studied the evolution of cell groups in ten isogenic lines of the unicellular green algae Chlamydomonas reinhardtii with prolonged exposure to a rotifer predator. We confirmed that this trait was heritable and characterized by a convex trade-off curve between reproduction and survival. Identical mutations evolved in all cell group isolates which were linked to survival and reducing associated cell costs. Overall, we show that just 500 generations of predator selection is sufficient to lead to a convex trade-off and incorporate evolved changes into the prey genome.

Project description:Activation of immunity via LPS injection causes an energetic trade-off with the somatic maintenance program of homeothermy, resulting in hypometabolism and hypothermia. To better understand the pathways involved in this trade-off, genome-wide expression profiling was performed on livers of mice housed at 2 different ambient temperatures (22°C and 30°C) with tissue collected at several timepoints post-LPS injection (0, 4, 12, 24hours). Our analysis revealed that regardless of ambient temperature, LPS reprograms host metabolism with distinct metabolic programs engaged to support entry and recovery from the hypometabolic state.

Project description:Plant Elicitor Peptides (Peps) are conserved regulators of defense responses across diverse plant species and are multigene families in most species. However, the functional relevance of Peps as multigene families remains largely undefined. While Arabidopsis Peps appear largely redundant in function, previous work examining Pep-induced responses in maize implied specificity of function. To better define function of individual ZmPeps, activities of each peptide were examined by assessing changes in defense-associated phytohormones, specialized metabolites and global gene expression patterns. In addition to delineating individual ZmPep and ZmPEPR activities, these experiments led to a number of new insights into Pep signaling. ZmPROPEP precursors were found to harbor multiple ZmPeps, a phenomenon not previously observed. In all, seven new ZmPeps were identified and together the family members were found to have specific activities defined by relative magnitude of response output rather than uniqueness. A striking correlation between ZmPep-elicited changes in levels of jasmonic acid and ethylene and the magnitude of induced defense responses was observed, indicating that ZmPeps regulate immune output through rheostat-like regulation of phytohormone levels. Peptide structure-function studies and ligand-receptor modeling revealed structural features critical to ZmPep signaling function. Structural analysis also led to identification of ZmPep5a as a potential antagonist peptide able to competitively inhibit activity of other ZmPeps, a regulatory mechanism not previously observed for this family. Using heterologous expression assays, endogenous gene expression patterns and analysis of CRISPR/Cas9 generated knockout plants, ZmPEPR1 was found to be the dominant receptor regulating ZmPep-mediated anti-herbivore defenses in planta.

Project description:The decision of whether to grow and proliferate or to restrict growth and develop resilience to stress is a key biological trade-off. Multiple tumour suppressors have roles in suppressing growth and proliferation when conditions are unfavourable, and aggressive tumours show re-wiring of the metabolic pathways and removal of these restraints on growth1,2. In plants, constitutive growth results in increased sensitivity to environmental stress3,4. However, the underlying mechanisms controlling this decision are not well understood. We used temperature as a cue to discover regulators of this process in plants, as it both enhances growth and development rates within a specific range, and is also a stress at extremes. We find the conserved chromatin protein DEK plays a central role in balancing the response between growth and arrest in Arabidopsis, and it does this via H2A.Z-nucleosomes. DEK target genes show two distinct categories of chromatin architecture, and these predict induction or repression by DEK. We show that these chromatin signatures of DEK target genes might be conserved in human cells, suggesting that DEK may act through a fundamental evolutionarily conserved mechanism to control the balance between growth and arrest in plants and animals.

Project description:Microbes experience high cell density in many environments that come with diverse resource limitations and stresses. However, high density physiology remains poorly understood. We utilized well-controlled culturing systems to grow wild-type and metabolically engineered Escherichia coli strains into high cell densities (50–80 g Cdry cell weight L-1) and explore the associated transcriptional dynamics. Knowledge-enriched machine-learning-based analytics reveal distinct stress-related gene expression patterns that are consistent with a fundamental trade-off between resistance and persistence. We suggest that this trade-off explains observed growth arrests in high-density cultures and that it results from the disruption of cellular homeostasis, due to reallocation of limited cellular resources from resistance functions towards maintenance requirements of engineered production pathways. This study deepens our understanding of high-density physiology and demonstrates its importance to fundamental biomanufacturing challenges.
