Project description:The objective of modern pig breeding is to exhaust the genetic potential in reproduction performance of sows regarding to litter size and litter weight of piglets. During gestation period, umbilical cord contributes to placenta-fetal communication and plays an indispensable role in the intrauterine embryonic development. In this study, we attempted to analyze the molecular mechanism of reproductive declined in high-parity sows from the perspective of umbilical cord blood. Firstly, we analyzed the reproductive character data of sows, and then the histological analysis of umbilical cord phenotype was performed. Next, we evaluated the effect of umbilical cord blood exosomes (UCB-EXO) on angiogenesis. Moreover, the expression characteristics of miRNA in UCB-EXO of high-parity sows with poor reproductive performance (OS) and multiparous sows with excellent reproductive performance (MS) were analyzed. Results showed that the reproductive performance performed best at 3rd-7th and gradually decreased after 8th parities. Angiogenesis was repressed in OS piglets. Moreover, the Exo-MS exhibited pro-angiogenesis properties, with those of Exo-OS were diminished. With the increase of parities, the angiogenesis and immune function of sows decreased significantly, greatly limited the reproductive potential of sows. The data demonstrated that miRNAs of UCB-EXO played a central role in intrauterine development and suggested a novel possible explanation for reproductive potential, provides reference for increasing female reproductive efficiency.

Herein, we analyzed the molecular mechanism in declining reproductive potential in high-parity sows by assessing the changes in the umbilical cord blood. Firstly, we analyzed the reproductive characteristics data of sows, followed by histological analysis of the umbilical cord phenotype. Overall, the best reproductive performance was at parity 3-7, gradually decreasing after parity 8 and angiogenesis was repressed in OS. However, exosomes derived from MS (Exo-MS) exhibited pro-angiogenesis properties but were diminished in exosomes derived from OS (Exo-OS). Additionally, the angiogenesis of sows was significantly decreased, increasing the risk of disease with the increase in parity, greatly limiting the reproductive potential of the sows. At the same time, miR-188-5p expression in Exo-OS was significantly higher than in Exo-MS (p < 0.01), implying that it may play an important role in regulating the lifespan and reproductive potential of sows. These findings demonstrated that miRNAs in UCB-EXO play a central role in intrauterine development. Further, the findings suggest novel insights on reproductive potential, which provide a reference for increasing the sow reproductive efficiency.

Project description:To identify the molecular markers of early pregnancy in pigs, we compared global gene expression profiles of the maternal peripheral blood in pregnant sows with the non-pregnant sows. Peripheral blood sample was collected at 14 days after insemination from the submandibular vein of pregnant and non-pregnant sows respectively, and total RNA was isolated, purified and sent for microarray analysis. This study identified 127 up-regulated and 56 down-regulated genes (FC >= 1.5 and P < 0.05) in peripheral blood from pregnant sows versus non-pregnant sows. Of the differently expressed genes, nine (including LPAR3, RXFP4, GALP, CBR1, CBR2, GPX6, USP18, LHB and NR5A1) were found to exert function related to early pregnancy processes. Seven differentially expressed genes (CHGB, USP18, VWF, LPAR3, NR5A1, PPARD, BIN1) were selected to perform qRT-PCR in the same RNA samples.The expression profiles of these genes detected by qRT-PCR were consistent with those by microarray, which confirmed the reliability of our microarray data.

Project description:To investigate the effects of nano selenium (nano-Se), curcumin (CUR), and glycyrrhiza extracts (GE) on reproductive performance, antioxidant and immune functions of primiparous sows and parity-2 sows, 54 primiparous sows (Landrace × Yorkshire) were randomly divided into three groups (18 sows per group): (1) CON group, basal diet (0.30 mg·kg-1 Se, sodium selenite); (2) CUR group, basal diet + 0.20 mg·kg-1 Se (nano-Se) + 300 mg·kg-1 CUR; (3) GE group, basal diet + 0.20 mg·kg-1 Se (nano-Se) + 500 mg·kg-1 GE. The trial lasted for approximately 180 days from day 90 of gestation of primiparous sows to parity-2 sows. There were no significant differences in reproductive performance among three groups (p > 0.05), but the litter weight gain of piglets from primiparous sows in the GE group was 16.49% higher than that in the CON group (p < 0.05). Compared with the CON group, the serum SOD and GSH-Px levels of primiparous sows in the GE group were significantly increased, and the MDA content was extremely decreased. The concentrations of serum IL-6 and IL-1β (p < 0.05) of primiparous sows in the GE group were significantly lower than those in the CON group, and the serum IL-10 and TNF-α concentrations (p < 0.05) was significantly higher. The combination of nano-Se and CUR decreased the serum IL-1β level and increased the TNF-α concentration (p < 0.05). In conclusion, the addition of nano-Se along with CUR or GE in the diet of primiparous sows significantly increased the antioxidant and immune levels in the serum of primiparous sows at parturition, enhanced their stress resistance, and thus improved growth performance of offspring piglets and reproductive performance of parity-2 sows.

Project description:In our previous sow study, two subpopulations of feed-restricted sows (60% of anticipated feed intake) were identified: ‘Restrict (Risk)’ that mobilized higher levels of body tissue stores (>40MJ ME day-1) compared to ‘Restrict (Non-Risk)’ sows (Patterson et al. Reprod. Fert. Deveopl., 2011, 23, 889-898) and although Risk sows maintain higher litter growth in the weaned litter, this was at the expense of lower embryonic weight in their subsequent litter compared with Non-Risk sows. To understand the underlying molecular mechanisms involved, we investigated the gene expression profiles of embryos from Risk and Non-Risk litters in this experiment.

Project description:Exosomes derived from mesenchymal stem cells (MSCs) have shown to have effective application prospects in the medical field, but exosome yield is very low. The production of exosome mimetic vesicles (EMVs) by continuous cell extrusion leads to more EMVs than exosomes, but whether the protein compositions of MSC-derived EMVs (MSC-EMVs) and exosomes (MSC-exosomes) are substantially different remains unknown. The purpose of this study was to conduct a comprehensive proteomic analysis of MSC-EMVs and MSC-exosomes and to simply explore the effects of exosomes and EMVs on wound healing ability. This study provides a theoretical basis for the application of EMVs and exosomes.In this study, EMVs from human umbilical cord MSCs (hUC MSCs) were isolated by continuous extrusion, and exosomes were identified after hUC MSC ultracentrifugation. A proteomic analysis was performed, and 2,315 proteins were identified. The effects of EMVs and exosomes on the proliferation, migration and angiogenesis of human umbilical vein endothelial cells (HUVECs) were evaluated by cell counting kit-8, scratch wound, Transwell and tubule formation assays. A mouse mode was used to evaluate the effects of EMVs and exosomes on wound healing . Bioinformatics analyses revealed that 1,669 proteins in both hUC MSC-EMVs and hUC MSC-exosomes play roles in retrograde vesicle-mediated transport and vesicle budding from the membrane. The 382 proteins unique to exosomes participate in extracellular matrix organization and extracellular structural organization, and the 264 proteins unique to EMVs target the cell membrane. EMVs and exosomes can promote wound healing and angiogenesis in mice and promote the proliferation, migration and angiogenesis of HUVECs.
