Project description:Epithelial ovarian cancer (EOC) is the most common type of ovarian cancer. Although studies have reported that downregulation of HOXD10 expression may contribute to the migration and invasion abilities in EOC, much remains to be fully elucidated. The present study aimed to identify different gene expression profiles of HOXD10 overexpression from the Human OneArray microarray and bioinformatics for the regulation of the progression of EOC cells. Bioinformatics analysis revealed that the pathways of the genes with different expressions were related to oxidative phosphorylation and mitochondrial function. Although the detailed regulatory mechanism between HOXD10 and the targeted oxidative phosphorylation genes is not yet fully understood, the findings of the present provide novel insight on HOXD10-mediated pathways for cancer metabolism, carcinogenesis, and the progression of EOC.

Project description:Ovarian cancer is common among women. Cancer in ovaries can arise from three different tissues, epithelial, germ and stromal cells. Majority of ovarian cancer arise from epithelial tissues. However, cancer in the germ cells or stromal cells are histologically, genetically and clinically heterogeneous, which result in a challenge in terms of treatment. In this study, we generated sRNA libraries of germ cell tumors (7 benign and 2 malignant) as well as 3 stromal tumors. In particular, we analyzed the miRNA profile of these tumors and found that malignant ovarian germ cell tumors (OGCT) are genetically distinct from benign OGCT and sex cord-stromal tumors (SCST). We detected several miRNAs that were deregulated in malignant OGCT compared to benign and SCST and fewer deregulated miRNAs between benign OGCT and SCST. However, we were still able to detect several deregulated miRNAs between benign OGCT and SCST.

Project description:Protein biomarkers of early stages of disease are critical for managing the disease and improving outcomes. Unfortunately, discovery of protein biomarkers is hampered by the limited availability of assays for sensitive and reproducible quantification of proteins in complex biological matrices such as blood plasma. In the recent years, Selected Reaction Monitoring (SRM) has emerged as a robust mass spectrometric method, capable of overcoming this limitation. Here, we present a comprehensive SRM-based strategy for developing protein biomarkers for epithelial ovarian cancer, and illustrate the extent to which SRM platform, combined with sound experimental design and statistical analysis, results in robust detection of predictive analytes. First, the biomarker development was guided by a discovery-driven proteomic effort to detect potential N-glycoprotein biomarker candidates in tissue of a genetically stable ovarian cancer mouse model. Next, 65 candidate markers were reproducibly quantified with SRM assays across a large cohort of over 200 plasma samples from ovarian cancer patients and healthy controls. Finally, these measurements were used to derive 5-protein signatures for distinguishing individuals with epithelial ovarian cancer from healthy controls. The sensitivity of the candidate biomarker signature in combination with CA125 ELISA-based measurements currently used in clinic, exceeded that of CA125 ELISA-based measurements alone. The SRM-based strategy in this study is broadly applicable. It can be used in any study that requires accurate and reproducible quantification of selected proteins in a high-throughput and multiplexed fashion.

Then cells were harvested for RNA isolation for microRNA expression analysis.

Project description:Epithelial ovarian cancer (EOC) is the most common type of ovarian cancer. Although studies have reported that downregulation of HOXD10 expression may contribute to the migration and invasion abilities in EOC, much about its regulation remains to be fully elucidated. The present study aimed to identify different gene expression profiles associated with HOXD10 overexpression in EOC cells. The present study confirmed that HOXD10 overexpression effectively inhibited the proliferation and motility of the TOV21G and TOV112D cells. Further, we overexpress HOXD10 in TOV112D cells, the different gene expression (DEGs) profiles induce by HOXD10 was analyze by the Human OneArray microarray. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG), ingenuity pathway analysis (IPA) was used to perform the pathway enrichment analysis for the DEGs. Integrated bioinformatics analysis showed that the DEGs were enriched for terms related to oxidative phosphorylation and mitochondrial function pathways. Dysfunction oxidative phosphorylation metabolic pathway occurs frequently in many tumors. We validated the expression of NDUFA7, UQCRB and CCL2 using qPCR, involving in metabolism-related pathway, were significantly changed by HOXD10 overexpression in EOC. The detailed regulatory mechanism that links HOXD10 and the oxidative phosphorylation genes is not yet fully understood, our findings provide novel insight into HOXD10-mediated pathways and their effects on cancer metabolism, carcinogenesis, and the progression of EOC. Thus, the data suggest that strategies to interfere with metabolism-related pathways associated with cancer drug resistance could be considered for the treatment of ovarian tumors.

Project description:Background: Germline truncating mutations in DICER1, an RNase III-type endoribonuclease essential for processing of microRNAs, are seen in families with the pleuropulmonary blastoma-family tumor and dysplasia syndrome; these individuals have a propensity to develop non-epithelial ovarian tumors (i.e. sex-cord stromal and germ cell tumors). Methods: We sequenced the whole transcriptomes or exomes of 14 non-epithelial ovarian tumors and noted closely clustered mutations in the RNase IIIb domain of DICER1 in four cases. Using Sanger sequencing, we then analyzed additional ovarian tumors for mutations in this region. The impact of the mutations on the enzymatic activity of Dicer1 was determined using in vitro RNA cleavage assays. Results: RNase IIIb domain DICER1 mutations were found in 30/102 non-epithelial ovarian tumors (29%), predominantly Sertoli-Leydig cell tumors (26/43, 60%), including 4/4 from individuals with germline DICER1 mutations. The mutations were restricted to metal binding sites within the RNase IIIb catalytic centres critical for miRNA interaction and cleavage, and were somatic in all 14 cases where germline DNA was available for testing. Of 266 epithelial ovarian and endometrial cancers tested, one case, an ovarian carcinosarcoma, had a hotspot mutation. In vitro, the mutant DICER proteins showed reduced RNase IIIb activity but retained RNase IIIa activity. Conclusions: Somatic missense mutations of the RNase IIIb domain of DICER1 are common in non-epithelial ovarian tumors. Genetic, pathologic, and functional evidence suggest that these mutations do not obliterate DICER1 function, rather, they alter it in specific cell types, a novel mechanism through which perturbed microRNA expression is oncogenic.
