Project description:Programmed cell death plays a vital role in shaping limb morphology in amniotes. BMP signaling has been shown to be essential for inducing interdigital cell death, but its relationship with reactive oxygen species (ROS) production, a known driver of this process, remains unclear. Here, we explore the molecular mechanisms by which BMP signaling regulates ROS production and cell death in the interdigital regions of chicken hindlimbs. To identify the ROS-producing cascade genes regulated by BMP signaling, we performed RNA-Seq of the interdigital region with PBS or Noggin bead implantation in chicken hindlimb buds. Subsequently, we performed RNA-seq of the interdigital region from stage 30 to stage 31 chicken hindlimb buds to analyze temporal expression changes of ROS-related genes during the normal progression of interdigital cell death in chicken hindlimbs. This study provides new insights into the ROS-mediated mechanisms of BMP-driven programmed cell death, highlighting the coordinated regulation of ROS generation and scavenging in limb morphogenesis.

Project description:Nonsteroidal anti-inflammatory drugs (NSAIDs), the quintessential medicines to treat pain and inflammatory conditions, induce cell death in human cancer cells, as repurposed anticancer agents, and in normal gastric mucosa, as a major side effect. The subcellular target/s of NSAIDs that leads to the cell death remained elusive so far. Here, by venturing transcriptomics followed by functional validation, we, for the first time, identified mitochondrial deacetylase Sirtuin 3 (Sirt3) as a non-canonical target of NSAIDs whose depletion induced the hyperacetylation of mitochondrial proteome, OGG1 depletion, mtDNA damage, electron transport chain defect associated mitochondrial dysfunction and finally cell death. Silencing of Sirt3 in AGS cells (a human gastric adenocarcinoma cell line) significantly aggravated NSAID-induced cytopathology. Whereas, honokiol mediated induction of Sirt3 corrected the NSAID-induced transcriptome alteration and gastropathy in rodent model. Together, the results identify Sirt3 as a common target used by NSAIDs to induce gastric carcinoma cell death and gastric mucosal injury.

A novel lncRNA FUAT1/TNS4 axis confers chemoresistance by suppressing ROS-mediated apoptosis in gastric cancer

Project description:Reactive oxygen species (ROS) play a vital role in conveying the cytotoxicity and resistance of most chemo-drugs. Thus, a better understanding of the detailed activities against oxidative stress in cancer cells may provide novel insights into the discovery of common mechanisms underlying chemoresistance. In this study, taking advantage of a customized screening strategy based on transcriptome sequencing (RNA-Seq) technology, we identified a novel lncRNA, designated FUAT1, as a key non-genetic player that participates in ROS-mediated intrinsic chemoresistance in gastric cancer (GC) cells. The role of the FUAT1 regulatory axis in chemoresistance was further explored by using a series of in vitro and in vivo assays, including gain/loss-of-function and rescue experiments. Mechanistically, FUAT1 could upregulate TNS4 by sponging miR-140-5p, allowing GC cells to survive upon chemotherapy via inhibition of ROS-mediated apoptosis. Clinically, the FUAT1/TNS4 regulatory axis is negatively associated with overall and progression-free survival in gastric and colon cancer patients treated with 5-FU adjuvant chemotherapy. Our findings touch on a fundamental adaptive mechanism for the survival of cancer cells upon chemotherapy, providing a novel insight into the development of strategies to overcome chemoresistance.
