Project description:Neuroimmune interactions are critical regulators of tissue homeostasis and allergic inflammation, and group 2 innate lymphoid cells (ILC2s) have emerged as an important cell type for mediating these interactions. Here, we identified that the abundance of dopamine (DA) was negatively correlated with the number of circulating ILC2s and lung function in human. Dopamine potently suppressed lung ILC2 responses in mice, which was dependent on the expression of dopamine receptor DRD1. Correspondingly, the local ablation of dopaminergic neurons further augmented ILC2 responses and aggravated allergic lung inflammation. Transcriptome and metabolic analyses revealed that dopamine impaired oxidative phosphorylation (OXPHOS) pathway in ILC2s. Augmentation of OXPHOS activity with oltipraz antagonized the inhibitory effects of dopamine. Finally, the local administration of dopamine dramatically alleviated allergen-induced ILC2 responses and airway inflammation. Our work highlights a model where dopamine is critical for suppressing ILC2 responses and maintaining homeostasis of type 2 immune machinery in lung.

IL-22 inhibits allergic airway inflammation in house dust mite-induced asthma models

Project description:While the pathogenesis of asthma is mainly orchestrated by antigen-specific Th2 cells and their cytokines, recent findings indicate the involvement of other subsets of helper T cells including Th17 cells. Previous studies have shown that IL-22, one of Th17 cell-related cytokines, plays multiple roles in regulating allergic airway inflammation; however, the mechanism underlying the Il-22-mediated regulation remains unclear. Here, we show that allergic airway inflammation upon intratracheal administration of house dust mite extract (HDM), a representative allergen, were exacerbated in IL-22-deficient mice. To address the molecular mechanisms by which IL-22 inhibits the development of HDM-induced allergic airway inflammation, we next performed an unbiased comprehensive screening of genes induced by IL-22 administration in the lung by RNA-seq analysis.

MicroRNA regulation of type 2 innate lymphoid cell homeostasis and function in allergic inflammation

Project description:MicroRNAs (miRNAs) exert powerful effects on immunity through coordinate regulation of multiple target genes in a wide variety of cells. Group 2 innate lymphoid cells (ILC2s) are tissue sentinel mediators of allergic inflammation. We established the physiological requirements for miRNAs in ILC2 homeostasis and immune function, and compared the global miRNA repertoire of resting and activated ILC2s and T helper type 2 (TH2) cells. Mice lacking the miR-17~92 cluster in ILC2s displayed reduced lung inflammation following exposure to the natural allergen papain. Moreover, miR-17~92-deficient ILC2s exhibited defective growth and cytokine expression in response to IL-33 and TSLP in vitro. The miR-17~92 cluster member miR-19a promoted IL-13 production and inhibited expression of several target genes, including SOCS1 and A20, signaling inhibitors that limit IL-13 production. These findings establish miRNAs as important regulators of ILC2 biology, reveal overlapping but non-identical miRNA-regulated gene expression networks in ILC2s and TH2 cells, and reinforce the therapeutic potential of targeting miR-19 to alleviate pathogenic allergic responses .

Dopamine D2 receptor activation inhibits choroidal neovascularization in Parkinson patients with age related macular degeneration.

Project description:Neovascular age-related macular degeneration (nAMD) remains a major cause of visual impairment and puts considerable burden on patients and health care systems. L-DOPA-treated Parkinson Disease (PD) patients have been shown to be partially protected from nAMD, but the mechanism remains unknown. Using murine models combining 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD and laser-induced nAMD, standard PD treatment of L-DOPA/DOPA-decarboxylase inhibitor, or specific dopamine receptor inhibitors, we here demonstrate that L-DOPA treatment-induced increase of dopamine receptor D2 (DRD2) signaling inhibits choroidal neovascularization independently of MPTP-associated nigrostriatal pathway lesion. Analyzing a retrospective cohort of more than two hundred thousand nAMD patients receiving anti-VEGF treatment from the French nationwide insurance database, we show that DRD2-agonist treated (PD) patients have a significantly delayed age of onset for nAMD (81.4 (±7.0) vs 79.4 (±8.1) years old, respectively, p<0.0001) and reduced need for anti-VEGF therapies (-0.6 injections per 100 mg/day daily dose of DRD2 agonists the second year of treatment), similar to the L-DOPA treatment. While providing a mechanistic explanation for an intriguing epidemiological observation, our findings suggest that systemic DRD2 agonists might constitute an adjuvant therapy to delay and reduce the need for anti-VEGF therapy in nAMD patients.

The dopamine transporter antiports potassium to increase the uptake of dopamine

Project description:The dopamine transporter facilitates dopamine reuptake from the extracellular space to terminate neurotransmission. The transporter belongs to the neurotransmitter:sodium symporter family, which includes transporters for serotonin, norepinephrine, and GABA that utilize the Na+ gradient to drive the uptake of substrate. Decades ago, it was shown that the serotonin transporter also antiports K+, but investigations of K+-coupled transport in other neurotransmitter:sodium symporters have been inconclusive. Here, we show that ligand binding to the drosophila- and human dopamine transporters are inhibited by K+, and the conformational dynamics of the drosophila dopamine transporter in K+ are divergent from the apo- and Na+-states. Furthermore, we found that K+ increased dopamine uptake by the drosophila dopamine transporter in liposomes, and visualized Na+ and K+ fluxes in single proteoliposomes using fluorescent ion indicators. Our results expand on the fundamentals of dopamine transport and prompt a reevaluation of the impact of K+ on other transporters in this pharmacologically important family.

Project description:Allergic rhinitis is a prevalent condition affecting up to 25% of the global population. The role of innate lymphoid cells in the pathogenesis of allergic diseases have been previously described, however, a single cell transcriptomic analysis to investigate differences between allergic and non-atopic healthy individuals have yet to be fully investigated. Using a combination of single cell RNA/protein and FACS validation, we provide evidence that there is a predominance and dysregulation of a type 2 response in the ILCs of allergic patients.

Dopamine biosynthesis in plants

Project description:Dopamine, an essential human neurotransmitter, also plays important roles in plant-microbe interactions and abiotic stress tolerance in plants. Although dopamine biosynthesis is well-characterized in humans, it remains largely unclear in plants. Through metabolite-based genome-wide association studies, we identified a dopamine biosynthetic gene cluster in Brachypodium distachyon comprising two tyrosine decarboxylase genes and three putative polyphenol oxidase genes. Biochemical and knockout mutant analyses revealed that dopamine biosynthesis proceeds primarily via tyramine as an intermediate, and dopamine oxidation leads to tissue browning in B. distachyon. Homolog searches and enzymatic assays suggest that the dopamine biosynthetic pathway is likely conserved across numerous plant species. This discovery provides valuable insights into plant dopamine biosynthesis with potential applications in reducing food browning, improving plant stress resilience, and bioengineering valuable compounds.
