Project description:p38 mitogen-activated protein kinases play fundamental roles in the regulation of the cellular response to environmental stress. p38 is known to regulate translation, mRNA processing and stability, and transcription. The transcriptional response mediated by phosphorylated p38 (P-p38) to extracellular stimuli, such as growth factors, cytokines and different environmental agents, has been thoroughly analysed in a variety of tissues and organisms. However, the genomic localisation of P-p38 is poorly understood. Here, we analysed the chromatin binding of activated P-p38 and the transcriptional response triggered by reactive oxygen species (ROS) in Drosophila S2 cells. We found that P-p38 is already bound to chromatin in basal conditions and relocates in the genome after exposure to high levels of ROS. The distribution pattern of P-p38 suggests that it is involved in the activation and repression of transcription in S2 cells, mainly regulating house-keeping functions.

Project description:Cryptococcus neoformans is an opportunistic fungal pathogen that is responsible for an estimated 112,000 deaths per year. We studied the role of the Hog1/p38 and Gcn2 signal transduction pathways during adaptation of C. neoformans to oxidative stress, a physiologically relevant stressor during infection. Using a combination of molecular assays and RNA-sequencing analysis in Hog1-, Gcn2- and double-deleted mutants, we identify shared targets of these pathways, and investigate a negative feedback loop that regulates induction of the integrated stress response (ISR). We found that simultaneous loss of Hog1 and Gcn2 profoundly impacts the ability of C. neoformans to tolerate oxidative stress. We also show that Hog1 regulates induction of the ISR, and that the Hog1 and Gcn2 pathways converge on repression of abundant, pro-growth mRNA, a key step in translatome reprogramming. Our results establish Hog1 and Gcn2 as key regulators of the response to oxidative stress, and demonstrate that the interplay between these pathways is critical for ROS tolerance in C. neoformans. More broadly, our results demonstrate the extent to which signaling via Hog1/p38 and Gcn2 are linked, and point to extensive recalibration of the cellular signaling apparatus when one of these pathways is disrupted.

Single nuclei transcriptomic profiling of adult Drosophila upon oxidative stress

Project description:Environmental factors, infection, or injury, can cause oxidative stress in diverse tissues and loss of tissue homeostasis. Effective stress response cascades, conserved from invertebrates to mammals, ensure reestablishment of homeostasis and tissue repair. Hemocytes, the Drosophila blood-like cells, rapidly respond to oxidative stress by immune activation. However, the precise signals how they sense oxidative stress and integrate these signals to modulate their immune response during oxidative stress are ill-defined. Furthermore, it remains unclear if all hemocytes respond similarly to oxidative stress or if there is a specification in distinct subsets. here, we employes single nuclei RNA-sequencing (snRNA-seq) to elcuidate the induced transcriptomic chnages in hemocytes and other cell types by Paraquat feeding compared to control food. Paraquat induces a pronounced oxidative stress response in Drosophila and we used this mdoel to study the diversification of immune cells upon this immune challenge in the adult fly.
