Expression profiles and bioinformatic analysis of circRNAs in cardiac fibrosis of db/db mice

Project description:To identify the differential expression of circRNAs in cardiac fibrosis of db/db mice, db/db mice cardiac apex tissues were detected through microarray analysis at the age of 21 weeks. Compared with normal control mice, 77 circRNAs were up-regulated and 135 circRNAs were down-regulated in the chromosomes of db/db mice (fold changes ≥1.5, p-values ≤ 0.05). Moreover, Real-time qPCR was performed to validate the expression of circRNAs. In db/db mice, the top four up-regulated circRNAs, mmu_circRNA_28667, mmu_circRNA_32303, mmu_circRNA_20674, mmu_circRNA_25425 were verified. In db/db mice, the top four down-regulated circRNAs which were indexed by circBase database were verified. They were mmu_circRNA_014302, mmu_circRNA_001535, mmu_circRNA_005522 and mmu_circRNA_013422.

CircRNAs in Arabidopsis thaliana across the lifespan

Project description:To explore the overall circRNAs involved in growth and development of Arabidopsis thaliana across the lifespan, we deeply sequenced samples of whole plants from different developmental stages (cotyledons emergence, rosette leavesï¹¥1 mm, rosette growth complete, first flower open, flourishing florescence, first silique shattered, senescence). The total RNA was purified by rRNA-depletion and linear RNA removal with RNAseR, and sequenced by the Illumina HiSeq2500 platform. We obtained 31 Gb raw data and identified 1217 circRNAs with expression quantity. We annotated these circRNAs and predicted their targeted microRNA. The circRNAs involved in growth and development of Arabidopsis thaliana across lifespan were identified and analyzed using the Illumina HiSeq2500 platform.

Project description:To understand the molecular change occurring in the atria during obstructive sleep apnea, we have implemented a rat model of OSA involving the surgical implantation of a tracheal obstructive device, allowing the rats to remain conscious and free-roaming throughout 2 weeks of apnea administration. Rats were divided into severe and moderate apnea groups, receiving a 23 second or 13 second apneas per minute, respectively. The two-dimensional polyacrylamide gel electrophoresis (2D PAGE) of atrial homogenates to compare the dysregulations in the protein pattern in severe and moderate apnea when compared to control.
