Project description:Mitophagy, a central guarding process to maintain mitochondrial quality, is commonly impaired in human diseases, but its impacts in adaptive immunity remains unclear. Here, we found that Parkinson’s patients have reduced CD8+ memory T cell abundance and failed to form memory T cells upon COVID-19 vaccination. We further unveiled that regulators of mitophagy, including Parkin and NIX, were upregulated in response to IL-15 for supporting memory T cell formation. Our results indicate that the mitophagy machinery orchestrate survival and metabolic dynamics required for memory T cell formation.

Nitazoxanide impairs mitophagy flux through ROS-mediated mitophagy initiation and lysosomal dysfunction in bladder cancer

Project description:Bladder cancer is one of the most common malignancy in the urinary tract with high recurrence and drug resistance in clinics. Alternative treatments from existing drugs might be a promising strategy. Nitazoxanide (NTZ), an FDA-approved antiprotozoal drug, has got increasingly noticed because of its favorable safety profile and antitumor potential, yet the effects in bladder cancer and underlying mechanisms remain poorly understood. Herein, we find that NTZ induces mitochondrial damage and mitophagy initiation through PINK1-generated phospho-ubiquitin(pS65-Ub) and autophagy receptor-mediated pathway even in the absence of Atg5/Beclin1. Meanwhile, NTZ inhibits lysosomal degradation activity, leading to mitophagy flux impairment at late stage. Mitochondrial reactive oxygen species (ROS) production is critical in this process, as eliminating ROS with N-acetylcysteine (NAC) efficiently inhibits PINK1 signaling-mediated mitophagy initiation and alleviates lysosomal dysfunction. Co-treatment with NTZ and autophagy inhibitor Chloroquine (CQ) to aggravate mitophagy flux impairment promotes NTZ-induced apoptosis, while alleviation of mitophagy flux impairment with ROS scavenger reduced cell death. Moreover, we also discover a similar signaling response in the 3D bladder tumor spheroid after NTZ exposure. In vivo study reveals a significant inhibition of orthotopic bladder tumors with no obvious systemic toxicity. Together, our results uncover the anti-tumor activities of NTZ with the involvement of ROS-mediated mitophagy modulation at different stages and demonstrate it as a potential drug candidate for fighting against bladder tumors.

Mitophagy promotes resistance to BH3 mimetics in acute myeloid leukemia

Project description:BH3 mimetics are used as an efficient strategy to promote mitochondrial-dependent cell death in blood malignancies, including acute myeloid leukemia (AML). Venetoclax, a potent BCL2 antagonist, is used clinically in combination with hypomethylating agents for the treatment of AML, while various compounds targeting MCL1 are in clinical trials. Yet, drug resistance eventually ensues, highlighting the urgency to understand the underlying mechanisms. Our genome-wide CRISPR/Cas9 screens revealed that loss of mitophagy regulators sensitizes AML to BH3 mimetics. One such regulator is Mitofusin-2 (MFN2), a GTPase that controls mitochondrial dynamics and the degradation of damaged mitochondria through mitophagy. Resistance to BH3 mimetics is accompanied by alterations in mitochondrial morphology, enhanced mitochondria-endoplasmic reticulum interactions, and augmented mitophagy flux. MFN2 inactivation, using a novel small molecule inhibitor, and pharmacologic inhibition of mitophagy synergizes with BH3 mimetics. Overall, targeting of mitophagy along with BCL2-family members is a promising strategy to overcome drug resistance in AML.

Extracellular peptide Kratos restricts cell death during vascular development and stress in Arabidopsis

Project description:During plant vascular development, xylem tracheary elements (TEs) form water conducting, empty pipes through genetically regulated cell death. Cell death is prevented from spreading to non-TEs by unidentified intercellular mechanisms, downstream of METACASPASE9 (MC9)-mediated regulation of autophagy in TEs. Here, we identified differentially abundant extracellular peptides in vascular11 differentiating wild type and MC9-downregulated Arabidopsis cell suspensions. The peptide Kratos rescued the abnormally high ectopic non-TE death resulting from either MC9 knockout or TE-specific overexpression of the ATG5 autophagy protein during experimentally induced vascular differentiation in Arabidopsis cotyledons. Kratos also reduced cell death following mechanical damage and extracellular ROS production in Arabidopsis leaves. Stress-induced but not vascular non-TE cell death was enhanced by another identified peptide, Bia. Bia is therefore reminiscent of several known plant cell death-inducing peptides acting as damage-associated molecular patterns. In contrast, Kratos plays a novel extracellular cell survival role in the context of development and during stress response.

Project description:In this work, we set out experiments to determine the mechanisms that regulate mitochondrial trafficking of p17/PERMIT-CerS1 complex in response to Drp1 activation to induce mitophagy and cellular consequences of this process altering mitochondrial metabolism in cultured cells in situ and genetic mice models in vivo. The data revealed that mislocalized mitochondrial ribosomes on the outer membrane due to Drp1-mediated fission are recognized by p17/PERMIT for CerS1 localization leading to ceramide-dependent mitophagy in response to oxidative stress by the generation of nitric oxide species (NOS). This process then leads to mitochondrial dysfunction resulting in decreased malate/fumarate/aspartate exhaustion, which is restored and prevented by molecular and genetic ablation of p17/PERMIT, LC3, Drp1, and Parkin in cancer cells or patient-derived 2D-organoids or mice brains in response to SoSe or ceramide analog drug, LCL768.
