Project description:This study was designed to investigate to test the effect of exosomes from urine-derived mesenchymal stem cells (USCs) on the survival and viability of aging retinal ganglion cells (RGCs), and explored the preliminary related mechanisms. The sequencing outcomes demonstrated 117 upregulated genes and 186 downregulated genes in normal RGCs group vs aging RGCs group, 137 upregulated ones and 517 downregulated ones in aging RGCs group vs aging RGCs+USCs medium group. These DEGs involves in numerous positive molecular activities to promote the recovery of RGCs function.

Conditioning of porcine kidneys during dynamic preservation with exosomes derived from Urine Progenitor Cells

Project description:Backgroud: among strategies to limit ischemia/reperfusion (IR) injuries in transplantation, cell therapy using stem cells to condition/repair transplanted organ appears promising. We hypothesized that using a cell therapy based on exosomes derived from Urine Progenitor Cells (UPCs) during hypothermic and normothermic machine perfusion can prevent IR-related kidney damages. Methods: we isolated and characterized porcine UPCs and their exosomes (Exo). Then these were used in an ex vivo preclinical porcine kidney preservation model. Kidneys were subjected to warm ischemia (32min) then preserved by Hypothermic Machine Perfusion (HMP) for 24h before 5h of Normothermic Machine Perfusion (NMP). Three groups were performed (n=5-6): Group 1 (G1): HMP/vehicle + NMP/vehicle, Group 2 (G2): HMP/Exo + NMP/vehicle, Group 3 (G3): HMP/Exo + NMP/Exo. Results: porcine UPCs were successfully isolated from urine and fully characterized as well as their exosomes which were found of expected size/phenotype. RNA-seq performed on kidney biopsies showed different profiles between G1 and G3 with regulation of potential IR targets of Exo therapy. Conclusions: we showed the feasibility/efficacy of UPC-exosomes for hypothermic/normothermic kidney conditioning prior to transplantation, paving the way of combining machine perfusion with exosome-based cell therapy for organ conditioning.

MicroRNA sequencing of umbilical cord mesenchymal stem cells (uMSCs) and HEK293T cells derived exosomes

Project description:Exosomes are cell-derived vesicles that were found in many biological fluids such as blood, urine, and cultured medium. Exosomes are small vesicles (approximately 100nm in diameter) that contain many functional molecules like cytokines, receptors and regulating RNAs. In this study, we found that uMSC-derived exosomes accelerates wound healing and reduce myoblast formation in vivo. In vitro study showed uMSC-exosomes specific microRNAs take major roles in inhibiting myoblast differentiation of fibroblast. MicroRNA sequencing of both uMSC- and HEK293T-derived exosomes revealed significant differences between these exosomes. Further infomatic and functional analysis showed that uMSC-exosomes specific microRNA-21, -23,-145, and -125b can target different components of TGF-β signaling which attenuates the expression of α-SMA in fibroblasts, thus inhibiting myoblast differentiation

Single cell RNA sequencing of stem cell-derived retinal ganglion cells

Project description:We used human embryonic stem cell-derived retinal ganglion cells (RGCs) to characterize the transcriptome of 1,174 cells at the single cell level. The human embryonic stem cell line BRN3B-mCherry A81-H7 was differentiated to RGCs using a guided differentiation approach. Cells were harvested at day 36 and incubated with THY1 antibody (Miltenyi) before undergoing FACS. THY1 positive and THY1 negative cells were subsequently prepared for single cell RNA sequencing. Single cell suspensions were loaded onto 10X Genomics Single Cell 3' Chips along with the reverse transcription master mix as per the manufacturer's protocol for the Chromium Single Cell 3' v2 Library (10X Genomics; PN-120233), to generate single cell gel beads in emulsion. Libraries were then sequenced on an Illumina HiSeq 2500.
