Project description:We utilized Comparative Genomic Hybridization (CGH), using probes designed from de novo assembly of 10 tissues into a grand transcriptome to identify genes located on the X and Y chromosomes of a stalk-eyed fly, Teleopsis dalmanni. We use next generation sequencing of RNA to identify over 500 genes that are differentially expressed in the testes due to the effects of a driving X chromosome (XSR) in T. dalmanni. Most of these genes were X-linked, evolving more rapidly than control genes, and exhibit elevated expression in the gonads. Finally, XSR has become genetically differentiated from standard X chromosomes – using the RNA sequence data, we found nearly 1000 sites in X-linked genes and only a handful in autosomal genes where there was a fixed nucleotide difference. We conclude that XSR has led to widespread sequence and expression divergence on the X chromosome in T. dalmanni.

Project description:To define what genes are predominantly or specifically expressed in either soma or germline in C. elegans adults, total RNA was extracted from germline-less glp-4 mutant animals or from dissected gonads, respectively. Total RNA sequencing was peformed in duplicates. Four samples in total.
Project description:We performed miRNA transcriptome analysis of mature medaka tissues using high-throughput next generation sequencing technology. The small RNA libraries were contructed from brain, liver and gonads in mature male and female medaka and a total of more than 128 million sequencing reads were generated from the six small RNA libraries. By comparison to known miRNA database, a total of 223 known miRNA types were identified with more than 50% expressed in brain. The expression profiling showed that almost half of the miRNAs are tissue-specific, with only 55 miRNA types from 34 families common to all tissues. A small number of miRNAs are also gender-specific. The experimental validation was performed for five represented miRNA candidates and four of them are confirmed.
