Project description:Lung adenocarcinoma (LUAD) is a leading cause of cancer-related mortality globally. TAR DNA-binding protein 43 (TDP-43) is involved in multiple tumor-associated biological processes, while its functional role and underlying mechanism in LUAD progression remain to be elucidated. Ferroptosis, a form of iron-dependent regulated cell death, plays a critical role in tumor development and progression. In this study, we investigated the regulatory effect of TDP-43 on ferroptosis and its subsequent impact on LUAD malignant progression. The expression pattern of TDP-43 in LUAD tissues and its correlation with clinical prognosis were analyzed. Functional experiments were performed to verify the effects of TDP-43 on LUAD cell proliferation, cell death and ferroptosis progression. Whole transcriptome sequencing combined with ferroptosis-related indicator detection was used to explore the regulatory mechanism of TDP-43 in LUAD ferroptosis. The results of this study reveal the regulatory role of TDP-43 in LUAD ferroptosis and malignant progression, providing experimental data and research basis for further exploration of LUAD pathogenesis and potential therapeutic targets.
