Quantitative proteomic study reveals differential expression of matricellular proteins between fibrous dysplasia and cemento-ossifying fibroma pathogenesis

Project description:Fibrous dysplasia (FD) and cemento-ossifying fibroma (COF) are benign fibrous-osseous lesions of the maxillofacial bones that exhibit numerous similarities in their clinicopathological aspects. However, due to differences in their clinical evolution and treatment, it is essential to diagnose these diseases with accuracy. The differences in the pathogeneses of FD and COF are largely unknown and currently there is no biomarker available for their differential diagnosis. Nonetheless, there are few studies comparing these lesions and predominantly comprised of immunohistochemical investigations of traditional bone biomarkers. Therefore, the present study aimed to quantitatively compare the proteomic and phosphoproteomic profiles of cemento-ossifying fibroma and fibrous dysplasia to shed light onto the mechanisms involved in their molecular pathogenesis and identify potential differential diagnosis biomarkers.

Expression data of bone marrow-derived osteoclasts expressing wild-type or mutant PSS1

Project description:Phosphatidylserine (PS) is an acidic phospholipid that is involved in various cellular events. Heterologous dominant mutations have been identified in the gene encoding PS synthase 1 (PSS1) in patients of a congenital disease called Lenz-Majewski syndrome (LMS). LMS patients show various symptoms, including craniofacial/distal-limb bone dysplasia and progressive hyperostosis. The LMS-causing gain-of-function mutants of PSS1 (PSS1-LMS) have been shown to synthesize PS without control, but why the uncontrolled synthesis would lead to LMS is unknown. We investigated the effect of PSS1-LMS(Q353R) on osteoclasts to elucidate the causative mechanism of LMS and found that PSS1-LMS inhibited the formation, multinucleation, and activity of osteoclasts. We used microarrays to comprehensively examine the effects of PSS1-LMS on osteoclast gene expression.

De novo variant in RING finger protein 213 causes systemic vasculopathy.

Project description:Systemic arterial stenosis, including moyamoya disease (MMD) and middle aortic syndrome (MAS), is a rare condition of unclear etiology. MMD is a cerebral angiopathy, whereas MAS affects abdominal and thoracic aortas, leading to various complications. Although some genetic associations with MAS have been identified, the causes remain elusive. In this study, whole-exome sequencing was used to identify de novo heterozygous missense variants of RING finger protein 213 (RNF213) (p.His4058Pro and p.Thr4155Pro) in two unrelated families exhibiting both MAS and MMD. To understand the significance of these variants, we generated knockin mice carrying the Rnf213 p.His4058Pro variant. Notably, homozygous knockin mice exhibited perinatal lethality due to respiratory failure and lung dysplasia, suggesting that this variant is pathogenic. Lung dysplasia in homozygous knockin mice was associated with upregulated innate immunity and inflammatory responses and downregulated cell proliferation. The findings suggested that in mice, the RNF213 p.His4058Pro variant plays critical roles in lung development, innate immunity, and inflammation, revealing the complexity of RNF213 function in various tissues and species. In conclusion, this study provides insights into the genetic basis of MAS and MMD, highlights the potential involvement of RNF213 variants in systemic vasculopathy, and identifies unexpected associations with lung development and immune processes.

Gene expression analysis of corneal endothelial cells in iridocorneal endothelial syndrome

Project description:Iridocorneal endothelial (ICE) syndrome is a rare ocular disease affecting the anterior segment, characterized by progressive deterioration leading to significant complications, including corneal endothelial compensation and secondary glaucoma. The unclear etiology of this syndrome limits clinical management to symptomatic interventions. The abnormality of corneal endothelial cells (ICE cells) in patients with ICE syndrome is considered the initial step of this disease. This study endeavors to elucidate the transcriptomic profiles of ICE cells utilizing smart-seq2 single-cell RNA sequencing techniques.
