Project description:Glacier ice algae of the streptophyte genus Ancylonema live on glaciers globally, including the Greenland Ice Sheet. Each summer these algae bloom despite freezing temperatures, low nutrient availability, and very high light intensities. These algae also survive continual darkness during polar night. However, little is known about the cellular mechanisms underpinning glacier ice algae resistance and adaptation to high light or how they survive during dark periods. To address this knowledge gap, we evaluated the response of Ancylonema-dominated samples from the Greenland Ice Sheet to light and dark conditions over a 12-day incubation experiment using combined multi-omics analyses: amplicon sequencing, metatranscriptomics, and metaproteomics.

Algae-dominated metaproteomes uncover cellular adaptations to life on the Greenland Ice Sheet

Project description:Eukaryotic algae-dominated microbiomes thrive on the Greenland Ice Sheet (GrIS) in harsh environmental conditions, including low temperatures, high light, and low nutrient availability. Chlorophyte algae bloom on snow, while streptophyte algae dominate bare ice surfaces. Empirical data about the cellular mechanisms responsible for their survival in these extreme conditions are scarce. This knowledge gap was addressed by quantifying proteins for both algal taxa from samples on the southern margin of the GrIS. We show that the streptophyte glacier ice algae have a relative enrichment in proteins involved in environmental signaling and nutrient transport, indicative of cellular readiness to dynamically respond to extreme GriS environmental cues, linked, for example, to photoprotection and the rapid update of scarce nutrients. In contrast, the chlorophyte snow algae have a high abundance of proteins linked to lipid and nitrogen metabolisms, providing evidence for the biological processes sustaining the cellular carbon and nitrogen stores necessary for survival in an oligotrophic environment. We also identify proteins in both taxa linked to the synthesis and breakdown of key cellular pigments. Our study gives novel insights into the cellular biology of these algae and their adaptation to extreme environments.

Project description:Current research into bacterial dynamics on the Greenland Ice Sheet (GrIS) is biased toward cryoconite holes, despite this habitat covering less than 8% of the ablation (melt) zone surface. In contrast, the expansive surface ice, which supports wide-spread Streptophyte micro-algal blooms thought to enhance surface melt, has been relatively neglected. This study aims to understand variability in bacterial abundance and production across an ablation season on the GrIS, in relation to micro-algal bloom dynamics. Bacterial abundance reached 3.3 ± 0.3 × 105 cells ml-1 in surface ice and was significantly linearly related to algal abundances during the middle and late ablation periods (R 2 = 0.62, p < 0.05; R 2 = 0.78, p < 0.001). Bacterial production (BP) of 0.03-0.6 ?g C L-1 h-1 was observed in surface ice and increased in concert with glacier algal abundances, indicating that heterotrophic bacteria consume algal-derived dissolved organic carbon. However, BP remained at least 28 times lower than net primary production, indicating inefficient carbon cycling by heterotrophic bacteria and net accumulation of carbon in surface ice throughout the ablation season. Across the supraglacial environment, cryoconite sediment BP was at least four times greater than surface ice, confirming that cryoconite holes are the true "hot spots" of heterotrophic bacterial activity.
