We characterized protease activity in lymph nodes and found that antigens were rapidly degraded in the subcapsular sinus, paracortex, and interfollicular regions, whereas low protease activity and antigen degradation rates were detected in the vicinity of follicular dendritic cells (FDCs). Correlated with these findings, immunization regimens designed to target antigen to FDCs led to germinal centers dominantly targeting intact antigen, whereas traditional immunizations led to much weaker responses that equally targeted the intact immunogen and antigen breakdown products. Thus, spatially compartmentalized antigen proteolysis affects humoral immunity and can be exploited.

Project description:This data set reports RNA sequencing results obtained from whole cerebral cortices of NEFH-tTa/tetO-hTDP43∆NLS transgenic (“rNLS8”) mice that were actively immunized with C-terminal fragments of the human TDP-43 protein. The widely used rNLS8 mouse model overexpresses cytoplasmically mislocalized human TDP-43 in a doxycycline-inducible manner, mimicking ALS/FTD-like CNS pathology and motor dysfunction (Walker et al., 2015). The purpose of this study was to evaluate the safety profile and therapeutic potential of various human TDP-43 epitopes (up to approximately 40 amino acids in length) which were used as antigens for active immunization in a rapidly progressing mouse model of TDP-43 proteinopathy. To this end, we pre-screened 15 peptide antigens, collectively spanning the entire TDP-43 protein sequence, for immunogenicity and safety. Next, we repeatedly immunized “rNLS8” mice with the most promising antigens prior to transgene induction, and performed bulk RNA sequencing of the neocortex, since this region is considerably affected by TDP-43 pathology in both humans and the mouse model. We asked whether active immunization with two different peptide combinations leading to high antibody titers would affect – i.e. prevent – transcriptional alterations upon transgene induction. In summary, we provide a bulk neocortical gene expression profile of actively immunized (and mock-treated) ALS/FTD-resembling “rNLS8” mice.

Project description:Eliciting a robust immune response against tumors is often hampered by the inadequate presence of effective antigen presenting cells and their suboptimal ability to present antigens within the immunosuppressive tumor microenvironment. Here, we report a cascade antigen relay strategy integrating antigen capturing nanoparticles (AC-NPs) and migratory type 1 conventional dendritic cells (cDC1s), named Antigen Capturing nanoparticle Transformed Dendritic Cell therapy (ACT-DC), to facilitate in situ immunization. AC-NPs are engineered to capture antigens directly from the tumor and facilitate their delivery to adoptively transferred migratory cDC1s, enhancing antigen presentation to the lymph nodes and reshaping the tumor microenvironment. Our findings suggest that ACT-DC improves in situ antigen collection, triggers a robust systemic immune response without the need for exogenous antigens, and transforms the tumor environment into a more “immune-hot” state. In multiple tumor models including colon cancer, melanoma, and glioma, ACT-DC in combination with immune checkpoint inhibitors eliminates primary tumors in 50-100% of treated mice and effectively rejects two separate tumor rechallenges. Collectively, ACT-DC could provide a broadly effective approach for in situ cancer immunization and tumor microenvironment modulation.

Project description:Introduction: Natural killer T (NKT) cells and CD8+ T cells are key in the immune response against multiple myeloma (MM), a largely incurable blood cancer. Immunization is a promising strategy to activate these T cell populations. To our knowledge, immunization with mRNA and α-galactosyl ceramide (αGC) have not been studied in MM, as knowledge on clinically relevant antigens in preclinical MM models is lacking. Methods: Transcriptomics and immunopeptidomics were used to identify candidate antigens for immunization in 5TMM models. Galsomes, lipid nanoparticles containing antigen mRNA and αGC were used to immunize 5T33MM bearing mice. This treatment was combined with a CD40 agonist. Tumor burden and activation of NKT cells and CD8+ T cells were studied using M-protein electrophoresis, cytospin, flow cytometry and ELISA. Results: Transcriptomics revealed survivin as a candidate antigen. Prime-boost Galsomes therapy targeting survivin significantly reduced M-protein levels despite low survivin-specific T cell responses. Further analysis showed potential T cell fratricide. Immunopeptidomics revealed HSP60, Idiotype, PICALM and EF1A1 as candidate antigens. Prime-boost therapy with Galsomes targeting these antigens reduced MM growth significantly when combined with a CD40 agonist, coinciding with significantly improved antigen presentation, co-stimulation and cytotoxicity of NKT cells and CD8+ T cells. Conclusion: These findings highlight the potential of Galsomes, an mRNA vaccine designed to activate CD8+ T cells and NKT cells, for MM therapy, and emphasize the importance of combinatorial approaches, addressing immune anergy for effective MM immunotherapies.

Project description:Our understanding of T-cell-dependent humoral responses has been largely shaped by studies involving model antigens such as recombinant proteins and viruses. In these contexts, B cells internalize the entire antigen or pathogen, and present a range of antigens to helper CD4+ T cells to initiate the humoral response. However, this model does not account for large pathogens (such as parasites) that are too large to be taken up by individual B cells, and the mechanisms by which B cells acquire and present antigens from large complex pathogens to T cells remain poorly understood. Here we used Plasmodium, the causative parasite of malaria, as a model to investigate the requirements for T cell help for B cells targeting the Plasmodium surface circumsporozoite protein (CSP). Upon Plasmodium sporozoite (SPZ) immunization, CSP-specific B cells can form a synapse-like structure with SPZs and take up CSP and non-CSP surface antigens. As a result, CSP-specific B cells can receive help from CD4+ T cells specific to antigens that are located on the surface but not cytosol of the Plasmodium SPZ. Therefore, B cells can obtain help, not only from T cells with the same protein specificity, but also from T cells specific for spatially linked antigens. This flexibility in T cell help may enhance the initiation and maintenance of humoral immune responses to complex pathogens.
