Project description:Inflammatory bowel disease induced by treatment with DSS induce destruction of gut barrier inducing massive bacterial translocation to distal tissue. Our findings suggest that gut microbiota translocation induces trained immunity in Granulocyte-monocyte progenitors that can underlie pathologies related to low-grade systemic inflammation. The identification of E.faecalis translocation to the bone marrow as a main driver of this effect and its sensing by Mincle receptor in bone marrow progenitors offers potential targets for intervention in pathophysiological settings where gut permeability is altered. Moreover, intravenously and intranasal administration of HK-EF provided protection against systemic candidiasis in wild-type and Rag1-deficient mice lacking functional lymphocytes, indicative of innate immune-mediated protection.

Project description:MV130 is an inactivated polybacterial mucosal vaccine that confers protection to patients against recurrent respiratory infections, including those of viral etiology. We showed that MV130 induces long term heterologous protection against viral respiratory infections in mice. Moreover, intranasal administration of MV130 provided protection against systemic candidiasis in wild-type and Rag1-deficient mice lacking functional lymphocytes, indicative of innate immune-mediated protection. As trained immunity acts via modulation of hematopoietic stem and progenitor cells (Kaufmann et al., 2018; Mitroulis et al., 2018) we hypothesized that MV130 could confer systemic long-term protection through reprogramming of hematopoietic precursors. For that we measured the chromatin accessibility landscape in multipotent progenitors (MPPs) coming from mice treated with MV130 or its excipient using ATAC-seq.

EF-P dependent genes in Salmonella Typhimurium

Project description:Elongation factor P (EF-P) is required to enhance peptidyl transferase on pausing ribosomes, which can be induced by certain codons on mRNA. It regulates expression of genes in specific conditions. Therefore, to search EF-P mediated ribosome stalling motif, ribosome profiling has been used on organisms such as yeast and E. coli. However, it has not been addressed on the intracellular pathogen Salmonella Typhimurium. Through the ribosome profiling, 135 sites in 128 genes were identified as EF-P dependent sequences and many of these sites had consecutive proline codons specifically at P site of the ribosome. As a motif level, PPG was the most abundant amino acid sequence at EPA site of the ribosome and followed by APP and DPP sequence. Also, our findings revealed that salmonella-specific gene, ugtL, is regulated by frameshifting caused during translation as ribosome stalled in the DPP motif. In addition, we showed that both proline motif and EF-P are engaged in this regulation, and it affects the resistance to antimicrobial peptides. In conclusion, this study emphasized the importance of poly proline motif on EF-P and suggested a possible mechanism how EF-P affects the physiology of Salmonella.

Ef-cd locus shortens rice maturity duration without yield penalty

Project description:The contradiction between ‘high-yielding’ and ‘early-maturing’ hampers further improvement on annual rice yield. Here we reported the positional cloning of a major maturity duration-regulatory gene, Early flowering-completely dominant (Ef-cd), and demonstrated that natural variation in Ef-cd could be used to overcome the above contradictory. The Ef-cd locus gives rise to a long noncoding RNA (lncRNA) antisense transcript overlapping the OsSOC1 gene. Ef-cd lncRNA expression positively correlates with the expression of OsSOC1 and H3K36me3 deposition. Field test comparisons of early-maturing Ef-cd near-isogenic lines (NILs) with their wild types as well as the derivative early-maturing hybrids with their wild-type hybrids conducted under different latitudes determined that early-maturing Ef-cd allele shortens maturity duration (ranging from 7 to 20 days) without a concomitant yield penalty. Ef-cd facilitates nitrogen utilization and also improves the photosynthesis rate. Analysis of 1,439 elite hybrid rice varieties revealed that the 16 homozygotes and 299 heterozygotes possessing Ef-cd matured significantly earlier. Therefore, Ef-cd could be a vital contributor of elite early-maturing hybrid varieties in balancing grain yield with maturity duration.
