Project description:Antimicrobial proteins of the REG3 family represent, like a wide variety of C-type lectins to which they belong, the first line of protection against infections and transformed cells. Their expression is inducible, particularly in response to inflammation, making their role in cancer biology less clear and controversial, as an immune-inflammation background may pre-exist or co-exist. We show that REG3A presents a suppressive role of REG3A in hepatocellular carcinoma (HCC) in a mouse models of HCC driven by c-MYC. By comparing the transcriptional profiles of liver tumors from MYC-REG3A-TG mice with those from MYC-TG mice, we found that MYC-REG3A tumors belonged to the S3 and G5 molecular subclasses of the human HCC classification proposed by Hoshida et al. and Boyault et al., respectively, subclasses that concentrate well-differentiated and CTNNB1-mutated HCCs. Tumors from MYC-TG mice exhibited features of S1 and G2/G3 subclasses. Results of the GSEA analysis indicate marked differential gene expression profiles. MYC-REG3A tumors and adjacent liver are enriched in metabolic pathways including bile acid, xenobiotic, and fatty acid metabolism, oxidative phosphorylation, and activation of Wnt/β-catenin signaling, whereas transcriptional reprogramming of MYC-TG mouse livers resulted in activation of cell cycle-, inflammation-, and glycolysis-related pathways as expected.

Project description:Gene expression changes were examined in transgenic MYC-driven liver cancers at different time points as tumors formed and upon early regression. Time points evaluated include: Control (non-tumor bearing), Pre-tumor (mice were removed from doxyclycine in their diet to induce MYC oncogene expression for 4-5 weeks), Tumor (tumor nodules from mice that had been off of doxycycline for 8-9 weeks) and Early tumor regression (tumor-bearing mice were placed back on doxycycline for 72 hrs to inhibit MYC oncogene expresion). MYC-Driven Mouse Tumor Models are described in Schahaf, et al., Nature, 2004 and Goga, et al., Nature Medicine, 2007. 11 Total Samples: 3 Control, 4 Pre-Tumor, 4 Tumor, 4 Early Tumor Regression. Control = LAP-tTA transgenic mice. Others = TRE-MYC x LAP-tTA double transgenic mice (doxy off).

At 5 months, the livers of transgenic mice exhibited mild to moderate hepatocyte dysplasia, this time point represents the preneoplastic stage. At 11 months, the tumors were visualized in WHV/c-myc transgenic mice and consisted mainly of well-differentiated, trabecular-type HCCs, and this time point was considered as neoplastic stage.

Project description:MYC and RAS signaling are two oncogene pathways important in human liver cancer. We sought to model liver cancer driven by either MYC or RAS using conditional LAP-tTA crossed to either TRE-MYC or TRE-HRAS models in the FVB/N background, and to understand oncogenic signaling pathways that are distinct between each model. Non-tumor tissue, from LAP-tTTA (LT2) mice were used as controls. In this study, we generated tumors from either MYC or RAS driven tumors and compared global gene expression changes to each other and control samples.
