Transcription profiling of EDS1- and SA-dependent genes in nudt7-1

Project description:nudt7-1 mutant plant exhibit stunted growth, enhanced basal resistance, elevated SA levels and spontaneous leaf cell death that is restricted to single cells but does not spread. All of these phenotypes depend on EDS1, a key regulator of plant innate immunity. Moreover, SA depletion in nudt7-1 results in EDS1-dependent cell death exacerbation and strong growth retardation. In this this experiment, we used 4-week-old soil-grown plants to seek for EDS1- and SA-regulated genes in nudt7-1 to identify novel components of the EDS1- and SA-signalling pathways.

1-deoxydihydroceramide causes anoxic death by impairing chaperonin-mediated protein folding

Project description:Ischemic heart disease and stroke are the most common causes of death world-wide. In both pathologies anoxia, the lack of oxygen, triggers profound metabolic and cellular changes. Sphingolipids have been implicated in anoxia injury, but the pathomechanism is unknown. Here we show that injury is caused by the accumulation of the non-canonical sphingolipid, 1-deoxydihydroceramide (DoxDHCer). We found that anoxia causes an imbalance between serine and alanine resulting in a switch from normal serine-derived sphinganine biosynthesis to non-canonical alanine-derived 1-deoxysphinganine. 1-deoxysphinganine is incorporated into DoxDHCer which impairs actin folding via the cytosolic chaperonin TRiC/CCT, leading to growth arrest in yeast and increased cell death upon ischemia-reperfusion injury in worms and mouse hearts. Prevention of DoxDHCer accumulation in worms and in mouse hearts resulted in decreased anoxia-induced injury. These findings unravel key metabolic changes during oxygen deprivation and point to novel strategies to avoid tissue damage and death.

Project description:Heart failure is a leading cause of death worldwide, and failing heart muscle is marked by increased O-GlcNAcylation (OGN). It is unknown if excessive OGN contributes to cardiomyopathy and heart failure. OGN modifies serines and threonines, total OGN levels follow cellular nutrient and metabolic flux in addition to net activity of O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA). We developed new transgenic mouse models with myocardial delimited over-expression of OGA and OGT, and found that OGT transgenic mice developed severe cardiomyopathy and premature death. In contrast, OGA transgenic hearts had normal function, but were resistant to pathological stress. Interbreeding OGA transgenic mice rescued cardiomyopathy and premature death in OGT transgenic mice. RNA Seq and functional studies highlighted disrupted metabolism in hearts from OGT transgenic mice that was rescued by OGA transgenic interbreeding. Here we show excessive OGN causes cardiomyopathy, identify gene programs responsive to pathological OGN, and suggest attenuation of OGN may be an effective therapy for heart failure.
