Project description:Type 2 diabetes is recognized as one of the primary contributors to cardiovascular diseases, with vascular endothelial dysfunction (VED) being a crucial mechanism underlying its development. In this study, we present the aberrant expressions of CircHMGCS1 and miR-4521 in diabetes-induced VED. Overexpression of CircHMGCS1 or silencing of miR-4521 expedited the onset of diabetes and aggravated VED. Mechanistically, CircHMGCS1 upregulated ARG1 by sequestering miR-4521, resulting in the inhibition of vascular NO secretion, enhanced adhesion molecules expression of VCAM1, ICAM1, ET-1, and increased ROS generation. Consequently, these events accelerated the impairment of vascular endothelial function. These findings underscore the physiological roles of CircRNA and miRNA in cardiovascular diseases triggered by diabetes and suggest that modulating the expression of CircHMGCS1 and miR-4521 could serve as a potential strategy for preventing endothelial cell dysfunction and the development of diabetes-associated cardiovascular diseases.

Unconventional role of HMGCS1 in driving pathogenic-Th17 cell immunity and autoimmune diseases

Project description:Identifying factors controlling pathogenic-Th17 cells is of importance, for their vital role in inflammation and immune-pathology. While intricate ER stress signals have been implicated in T cell biology, its role in Th17 cells remain poorly understood. Here, we demonstrated that HMGCS1, a cholesterol biosynthesis precursor enzyme, was highly induced by inflammatory cytokines and preferentially expressed by pathogenic-Th17 cells in vitro and in vivo. HMGCS1 specifically dictated pathogenic-Th17 cell differentiation and augmented autoimmune diseases. Nonetheless, HMGCS1 role in pathogenic-Th17 cells was independently of cholesterol metabolism but necessitated its catalytic Cys129 residue. Notably, HMGCS1 leveraged IRE1a-XBP1s-dependent ER stress response which was essential for pTh17 cell generation and pathogenicity by transcriptionally activating Rorc loci. Mechanistically, HMGCS1 located to the ER membrane, where it bound and stabilized IRE1a protein via restraining IRE1a interaction with K48-linked ubiquitination-dependent E3 ligase MARCH5. Of note, interfering HMGCS1 and ER stress impeded pathogenic-Th17 immunity and mitigated autoimmune diseases. Therefore, our work reveals a non-canonical axis wherein HMGCS1 sustains ER stress to license pTh17 cell differentiation during autoimmune responses.

Our study investigates the pivotal role of circular RNAs (circRNAs) and MicroRNAs (miRNAs) in diabetes-induced endothelial dysfunction, a critical aspect of cardiovascular diseases. CircHMGCS1 (hsa_circ_0008621) emerges as a specific circRNA that effectively targets miR-4521. The upregulation of CircHMGCS1 acts as a miR-4521 sponge, facilitating the expression of arginase 1 and accelerating the progression of diabetes-induced endothelial dysfunction. Furthermore, intricate associations among circRNA, miRNA, and mRNA are revealed, illuminating the underlying molecular mechanisms involved in diabetes-induced vascular endothelial dysfunction. These findings significantly advance our understanding of cardiovascular diseases, providing potential therapeutic targets for intervention.

Arg1 expression defines immunosuppressive subsets of immune infiltrating cells in cancer

Project description:Tumor-associated macrophages (TAM) have attracted attention as they can modulate key cancer-related activities, yet TAM represent a heterogenous group of cells that remain incompletely characterized. In growing tumors, TAM are often referred to as M2-like macrophages, which are cells that display immunosuppressive and tumorigenic functions and express the enzyme arginase 1 (Arg1). Here we combined single cell intravital imaging with scRNA seq to uncover the topography and molecular profiles of Arg1+ macrophages in mice. We further assessed how immunotherapeutic interventions impact these cells directly in vivo. We show that: i) Arg1+ macrophages are more abundant in tumors compared to other organs; ii) there exist two morphologically distinct subsets of Arg1 TAM defined by previously unknown markers (Gbp2b, Bst1, Sgk1, Pmepa1, Ms4a7); iii) anti-Programmed Cell Death-1 (aPD-1) therapy decreases the number of Arg1+ TAM while increasing Arg1– TAM; iv) accordingly, pharmacological inhibition of arginase 1 does not synergize with aPD-1 therapy. Overall, this research defines subsets of immunosuppressive myeloid cells with powerful complementary single cell analytical approaches that pave the way for a more intricate understanding of TAM behavior.
